Can I ask if you are talking to potential partners and sign NDA can you discuss preclinical data not in the public domain?

If discussions go nowhere you have created a third party who had info others do not. By publishing the data, even in poster form you make it public in a way that also ticks some promotional boxes. We know Avacta have two other animal models on addition to mouse but these have not been published before.

The other thought is you don't publish data that gives competitors a hand or before you have patents positions in place.

Hence the excitment about the poster?

Choosing US to do it maybe hits timeline, target audience and the ability to give a doctor a pen / Tshirt (not in UK tsk)

We are looking at trial for its dose escalation. What's the highest tolerated dose.

AS has previously stated the likely treatment strategy will be built around higher doses at tumour site for any given dose and lower cardiac exposure enabling more frequent dosing. I'm guessing you want a dose that fully saturates the FAP enzyme taking into account serum half life of the drug but no higher. Im getting a sense the next dose escalation will be that balance point

If these were slides used in roadshow then key info such as financials and the lfd progress could have / should have RNS.

Interesting. The highest dose for standard dox is cumm max of 550mg. I'd go off his bnf tweet rather than Canadian.

The DE is much higher than this. The clinical experts have ok'd this so presumably systemic dox in cohort 1 was low otherwise you would be exposing patients to same risk as normal dox.

One question is what % of prodox gets converted at tumour site. We know in mice it is very high 18x normal can be reached for same cardiac exposure.

Mice do not have same pK or Fap activity as man.

I'm guessing 18x the concentration is way above the dose required for max therapeutic response. Therefore can lower dose with no loss in efficacy. Might find cohort 2 dose hits max clinical response - local tumour concentrations being high. Balance between cardiac risk and tumour killing.

For at risk patients the cumm max dose is 400. If ava6k does achieve max tumour effect for very low systemic exposure then no dose adjustment required.

Trial RNS was August. Second cohort Feb. AS stated Covid had slowed stuff down. Standard cycles are every three weeks but I don't know how many cycles. Guess at 4 and all patients start same day gives 3 months minimum.

I don't think they escalate dose until cohort complete (don't know as a fact)

If doing third cohort then that makes it six months. So September. If only two (going for more cycles not highest dose - see presentation) then maybe July. Therapeutic max at tumour site may be achievable on DE dose. Can see local conc if do a biopsy.

Good point. What outcome were they expecting. The only info not in public domain was the struggle they are having with LFD and they are trying to do biopsies in the trial. Tone was a bit of someone who had been put in detention - I know regulatory is a nightmare but has the feel of shrugging shummper shoulders. SP doesn't affect balance sheet but come on. Watching it was like eating sand paper and if going to talk about FAP activity, epitopes etc in an investor meeting then take off the mortar board and tell us why IRS important to the value of the company. Otherwise it smells of smoke.
