I pointed out a number of concerning facts in my first post which didn’t include kidney damage. I added that when some idiot said half life wasn’t an issue because we could just give patients loads of AVA6000 and make more money

The Pom poms picked on kidney damage as an argument they could win and conveniently forgot about AVA6000 half life versus rate of dox cleavage.

Hopefully arm 2 results when published will prove that AVA6000 will be viable and save a lot of lives but I really don’t like the way that the rhetoric is shifting to how great the follow on pipeline is going to be. By my reckoning that’s 3 years behind and as you all know me so well you know that I might not live long enough to enjoy my Avacta investment. Though I could possibly benefit from Avacta products

40 ish patients

15 results shown

10 showing any shrinkage

5 significant shrinkage

So about 5 out of 40 doesn’t seem that great (unless you are one of those)

So possibly 30 patients where tumour carried on growing and you wonder why BP aren’t falling over eachother to buy us

I can’t say that. But equally you can’t say the opposite.

It’s all about trying to analyse why we are where we are.

And also counter the unending wishful fantastical predictions by the Pom Pom girls. This isn’t a slam dunk. The market and BP are not as thick as mince and they probably have better access to the data than we have. How many niaive investors have been drawn in by 3 years of chanting things like it’s all about the science etc?

How many times have we been told buy now go all in at 180, 160, 150, 120, 100 and now 50, 40. You’ll never get the shares this cheap. And yet every time you have been wrong and yet you don’t question it? It’s always a conspiracy. It can’t just be that there is a long way to go and some scientific hurdles to jump.

I believe we will make our investment back but not for a while yet and we are not going to be millionaires next year Rodney.

And in 36 out of 40 patients the doc cleaved had no significant impact on the tumour.

Next you’ll say it wasn’t an efficacy trial which just the point I’m making. We are 3 years in an haven’t yet started P2

So come on. Who out there can tell me dox cleavage rate versus AVA6000 concentration in the bloodstream?

i am not talking about kidney damage due to dox i’m talking about potential for kidney damage due to trying to urinate massive amounts of uncleaved ava6000. amounts that might result from very frequent high dosage regime. that is why ava6000 needs to be efficacious in ‘normal’ volumes.

but my main concern is rate of av60000 urination versus rate of cleavage. you cannot say this doesn’t matter because obviously the amount of dox being cleaved in the tme of any particular patient is proportional to strength of ava6000 in the bloodstream at any one time. and if it’s being ****ed away too quickly then that is a big problem. i’d be delighted if you can show me where the data is that shows that. otherwise this is my conclusion from all that’s happened over the last 3 years

I don’t see where it says that kidneys won’t be damaged by having to clear a lot of AVA6000 in a short time.

But we digress. My main thrust is rate of AVA6000 dilution versus rate of cleaving. But hey time will tell when the data is revealed

2. P1 trial extended and the arm2 with more frequent dosing - facts

3. New drug IP registered to double half life - fact

4. Kidneys can only handle cleaning so much crap out of the blood - biological fact

Oooh and if you really want a scare - will the modified AVA6000 with double the half life in the blood cleave at a slower rate in the TME - concern

And you say I am negative for asking so many questions but that’s because there is always so much going unsaid. So much to question.

If AVA6000 was so close to success why did Al and the others leave. I don’t believe it was just the crap financial deal because in the long term if Precision is the platform for a range of super efficient warhead drugs the that deal was neither here or there

Pom Pom girls FFS look at what has happened in the trials over the last three years. Look at the SP. Where are BP?

P1a was going to be 4 cohorts. Why has it stretched to seven. Why is CC being coy about arm2 results.? We’ll be going for daily dose rate soon. Although even that might be acceptable in a lot of patients.

Al had to go because all he could see was his baby being successful but it has a big flaw it’s not in the body long enough to do its work. Al said all along that the full effort was on AVA6000 and nothing would change that. And now he’s out the door, well AVA600 was just a minor step to the pipeline to be revealed in 2H24 that’s gonna knock our socks off.

You are right and I did point this out after the December presentation and was shouted down by the Pom Pom girls as usual.

The fact is AVA6000 is being metabolised more quickly than it can be cleaved in a lot of cases. I’ve already said this.

It is the LFT all over again. Lauded to the skies until the fundamental flaw is revealed. But just as AVA6000 came to the rescue of the LFT disaster, so the fabulous new pipeline comes to to rescue the AVA6000 disaster. AVA6000 will be paused to be tweaked just like the LFT, never to be seen again. Anyone got that sinking feeling?

Anyhoo who cares. We will make our investment back with one of the targeted drugs coming through in the end.

My understanding but not tax advice:

When you die any US shares you hold will be subject to US Estate tax regardless of where they are held. So if they are worth more than $60,000 currently, you will be taxed on the excess. You can set that off in the UK against your overall IHT bill to avoid double taxation.

What seems silly is that if you sell the shares the second before you die you’d be ok🤣🤣🤣

I was purely pointing out that chemotherapy has a nasty ring to it. Vaccines not ADCs are the long term answer. Hence BP reluctance to commit money to yet another chemotherapy treatment at the moment.

But as Bella says, Novartis will commit as soon as arm 2 completes successfully but we won’t see a massive leap in SP.
