Oh yes *smiley emoji with sunglasses lol*
Lools like a new assay. Bit of background on NSP16;

A pint of whisky that is Larry
SARS-CoV-2 NSP16 (Nonstructural Protein 16), also called as ribose 2'-O-methyltransferase, is one of the nonstructural proteins encoded by SARS-CoV-2 orf1ab. The polyproteins of CoVs are cleaved by virus-encoded cysteine proteinases comprise papain- and chymotrypsin-like proteases into 16 nonstructural proteins including the expression of NSP1 to NSP11 by orf1a and encoding NSP12 to NSP16 by orf1b. According to BLAST analysis, the sequence identity of ORF1ab protein between SARS-CoV-2 and SARS-CoV is more than 90% with the query cover of about 100%, while the sequence identity of NSP16 between these two viruses is about 93.24%
Looks like an assay thatll detection all coronaviruses?
CT Tks I've been trying to work it out but watching football at the same time doesn't help if you've nailed that it's extremely exciting.
Source for the below quote is here. https://www.activemotif.com/catalog/details/81318/recombinant-sars-cov-2-nsp16-protein
Would love to hear from some of our more learned posters on this.
Don't want to be a party pooper but nsp16 is detected by PROmate 2G
Just google 'PROmate nsp16" and it'll be the first link
We know there is tons on the way though, Ajan Reginald said recently they're planning 26 new products by end of 2022
From the first link i shared.
Coronaviruses employ between 20 and 30 proteins to carry out their viral replication cycle, including infection, immune evasion, and replication. Among these, nonstructural protein 16 (Nsp16), a 2'-O-methyltransferase, plays an essential role in immune evasion. Nsp16 achieves this by mimicking its human homolog, CMTr1, which methylates mRNA to enhance translation efficiency and distinguish self from other
Thanks Steve, you are correct.
It looks like the addition of NSP16 could well be from PROmate 2G.
I did wonder why it showed 3.2m sequences analysed if it was indeed new. Would have been more curious if it was say 100k. If they have been analysing it from the beginning, why add it in there now I wonder?
Fair point Steve page 20 of this doc (and I check manually)

So why only just appeared on weekly report?
Wilson -
'NSP16' is discussed first I believe in the Half Year Update 18.08.21:
"A key product expected to support second half growth in private testing will be the Company's PROmate® COVID-19 2G (2 gene) test. As announced on 23 April 2021, the Company's CE-IVD PROmate® COVID-19 (1 gene) test identifies the SARS-CoV-2 ORF1ab gene. Due to an emerging need in some countries, the Company launched a second CE-IVD PROmate® COVID-19 test to identify both the SARS-CoV-2 ORF1ab gene and nsp16 gene."
I checked the IFU for the non promate 2g test:
genesig® COVID-19 2G Real-Time PCR Assay
In vitro Real-Time PCR diagnostic test for COVID-19
does not reference the nsp16 gene.
Thanks B2, this was something I missed originally. I had always assumed it was either the M or S genes along with ORF1ab.
Reading the articles below gives me great confidence in the use of NSB16
CT it's nsP don't you know:))
B2 I'm on the floor with all the different brackets:)) if you could help though
I checked the IFU for the non promate 2g test:
genesig® COVID-19 2G Real-Time PCR Assay
In vitro Real-Time PCR diagnostic test for COVID-19
does not reference the nsp16 gene.???
I only send morse code
Oh no! - Morse code not in anyone's training syllabus now Wilson?
Oh ****..
https://www.genesig.com/assets/files/Path_COVID_19_2G_IFU_Issue_101.pdf?timestamp=1606160727
A wordsearch does not reveal 'nsp16'.
B2 so nsp not in genesig Covid 19 2g
but is in promate covid19 2g
In other news once we have all been blown to smithereens by nuclear warheads I will be able to communicate using morse..every cloud.
You'll be deed man, - we'll all be deed, - there'll be nobody to signal to.
We're all dooomed, just as we were about to hit the big time..
Glad to see you got involved with other debates tonight byp

Goodnight all.
Oh look another fabulous contributor, must dash.
Wilson….what am I contributing in….I see the original tweet has been deleted to this thread…..why is that? Lol
JUST IN : africa cdc maps regional spread new covid .
Morning Larry I was just about to say similar
perhaps we are the same person or I am porky who knows.
On the positive side it started an interesting debate where personally I read some stuff I'd wouldn't normally read, your research is invaluable please continue to share.
Back to listening to the rain.
Hi Wilson, idd, it was interesting Reading about nsp16.
Bad part is i created myself Hope, and maby to others too.
But well, thats how it is. You try looking At stuff, think its new or have some ideas about it , and in the end its all wrong.
But also, who knows, as we are able to target this, maybe it could be of interest for other tests still !
