What value upfront payments and royalties paid by Medimmune if nuvec proves worthy of further joint development? The timescale on announcement is from now to end feb 19.

The reason for mentioning the below is that Nature is a very high impact journal

The PEI coating on the nanoparticle gives highly branching spokes which trap DNA on its outside (cf nuvec)

Does anyone have more info on how the 'spokes' on nuvec are produced? Is it patented?- Thanks

These are worth looking up on google/pubmed.
Silica nanoparticles bioengineered with PEI achieved x38 gene expression in murine neuroblastoma cells (Nature Jan 17). Good viability, non toxic. These particles are similar to Nuvec, and suggest this approach to a non viral vector is promising.

Also the Cell and Gene Therapy Catapult Centre in Stevenage got the green light to start making new therapies in collaboration with industry/academia- there is a good video on their website

What makes N4p compelling is that have chosen drugs which address very large potential markets. Also it’s cutting edge technology which may or may not work. They are early stage and so the risk is high. However the involvement or AZ and external validation by innovate uk etc were a game changer for me

Btw I hope Hugh Wilson is not David farrier. On the positive sid Hugh comes across as knowledgeable and I find his posts interesting but he is also an N4p fundamentalist. I am hoping that David is a ruthless savvy Pharma ex venture capitalist investor who spots winners. It is noticeable that none of the big investors have sold out here- is that correct?

On a serious note I agree with others that it is unlikely that they will shelve the high level of technology and patent protection they have built up. An issue is whether sildenafil is worth pursuing and whether there is a hint of efficacy in the data to make it worth persisiing with. Unfortunately the value of all of their drugs have been thrown out with this first failure and I am unsure this warranted. We are well below our rival cocrystal cooker Nuformix- they have not yet started trials. Also we are questioning Nigel and his teams competency and honesty. He comes across well on the videos and they have a lot of experience

So there is a licensed (mhra) version of sublingual Viagra- not fast acting

This gives me some hope for n4P! (my opinion)

Below is a link to an independent review on which PDE5 inhibitor patients preferred- there is evidence within it that some patients would prefer a drug which has BOTH short acting and long acting properties (good for n4p)

Also a trial from last year by an Italian pharma co (IBSA) comparing sublingually absorbed version of sildenafil (orodispersible film- now licensed) vs oral sildenafil
The good news for IBSA was that it was absorbed as well as the oral version- almost the same P/Ks- however it was not faster acting

The 3 month delay in the sidenafil was due to a manufacturing issue, and was red flag for some. For all the benefits of biphasic drugs a highlighted issue is cost and manufacturing-is this easy to tweak/ correct? Of note I am not aware of a drug on the market (happy to be corrected) that is both sublinguAl immediate release and oral extended release- this is quite an innovation as long as it can be made to work

I agree that we just need to bide our time for the next rns. The last one came across as overly negative. They only had the top line result without the underlying detail necessary to make a proper evaluation of the product

N4p is a small outfit. Is it wise to be ‘more than a one trick pony?’ Is it good to have multiple multi million opportunities? There have been a few patents listed online to reformulate sidenafil online inc one from Pfizer since 1998 though nil recent. I invested a significant amount following the MedImmune announcement. I am a doctor involved in clinical trials. I do not understand pharmaceuticals well and so not sure of what to make of the sidenafil reformulation and it’s chances of success. It’s biphasic action means either one or both aspects went wrong. Not sure what implications this has for the other drugs for which patents have been gained. Gene therapies are certainly cutting edge at the moment and N4p strategy of making the delivery vehicle seems a less risky route with the possibility of multiple collaborations- more likely if the MedImmune one is successful. Sorry have not got the ic article on me at the moment/ maybe someone else can help?
