Normally companies announce they are presenting abstracts about a month or at least a few weeks ahead even if its a LBA.
This one is on the day of the presentation .

I'm no scientist so just speculating really .
Its good however that Faron have this nailed down on a patent . Imagine somebody else showing up with a drug that reduces Clever 1 effectively enough to challenge Bex .
And I see your point about how Bex may already suppress the Clever1 that the TLR9 was going to have a go at - so overall 2+2 would only =3 .
With Soluble Clever 1 and the fact that levels seem to rise again with long dosing intervals , there seems to be enough of this stuff knocking around to present opportunities to reduce levels both in the tumours and around the body , and if they are done by different mechanisms , then 2+2 could well =4 .

Its certainly an interesting development .

Just going back to the patent again - I found another version of it which included two charts .

So it looks like they have lab based findings that have proved their theory in mice ( as they did originally in Bex )

One shows the results of work in the lab on mice - showing rapid reduction in Clever 1 , and not seen using other agonists .

As seen in the FIG. 1, stimulation of alternatively activated macrophages with unmethylated CpG oligodeoxynucleotides (TLR9 agonist) induced rapid downregulation of Clever-1 expression by macrophages. A similar response was not seen with TLR3 (Poly(I:C)), TLR4 (LPS) or STING (2',3'-cGAMP) agonists.

The second chart showed the amount of Clever 1 found in various types of cancer tumours .
Not every patient is the same however , or cancers , so screening for clever 1 would make sense .

The most abundant expression of Clever-1 was observed in samples from colorectal cancer, vulvar cancer, pancreatic cancer, gastric cancer, breast cancer and glioma, but not all patients in even these cancer types are Clever-1 positive. Prostate cancer on the other hand is extremely low in Clever-1 positive tumour associated macrophages, but individual variation may occur. Therefore, the screening of the patients for Clever-1 positivity prior to treatment provides possibility to select patients suitable for the treatment method targeting Clever-1 positive macrophages according to the present invention and may improve the treatment response.

Expert opinion seemed to be here that as a monotherapy it wasn't enough to reverse the immunosupressive status of patients with a high tumour burden .
Importantly, the synergistic and complementary effect of TLR9 agonists and tumor immune checkpoint inhibitor therapy is expected to exert greater potential.

They do warn of possible side effects with combis such as chemo etc which require further study , but again , they seem to be in agreement that in principle these TLR9's look to be best placed as a complimentary treatment .

We are already running a trial ourselves for Bex as a neoadjuvant ( given before renal surgery )

And you can bet your life they are looking at how TLR9 affects 'soluble Clever1' too .

This discovery looks to be pretty significant I'd say , after a full read of the patent .
The patent was submitted 2 years ago - and now just published .

The patent covers the use of TLR9 agonists pretty comprehensively in the treatment of cancer patients experiencing high clever 1 levels , be that in tumours or circulating .

It covers treatment of ALL cancers

It covers all delivery methods - be that I/V , Intra muscular , subcutaneous and intra tumoural .

It covers use of TLR9s as part of a treatment alongside with other antibody treatment ( eg Bexmarilimab)

It covers screening for Clever1 positive macrophages prior to treatment in order to maximise success

As a follower of most Farons presentations , I can honestly say I have never heard of these TLR9s ever mentioned .
Yet here we have some pretty bold statements laid out in this patent .

Yet the patent appears to lay out conclusions that they have made discoveries regarding its efficacy .

Surely this should be an RNS'd piece of information - since we know how important Clever1 suppression is , through the Matins trial and much hyped ESMO late breaking abstracts , and here we have an indication that they have also found a whole new tool work along side Bex .

Side effects of TLR9's seem very minor also - fever etc , so again we have a very safe treatment .

This publication explains the basics . Their conclusion based here , is that TLR9 is a safe montherapy , and get this bit ...

"TLR9 agonist monotherapy is safe, although efficacy is minimal in advanced cancer patients; conversely, combinations appear to be more promising."

I know I'm starting to speculate now , but imagine if TLR9 combined with Bex - a drug that doesn't need to be given in high doses and that is very safe , combined with another similar treatment - that both reduce Clever 1 expression .

And we are already seeing 35-40% success rate on 4-5 different cancers with Bex alone .

If this turns out to be significant , you'd start to understand the reasons for further placings in order to pursue these findings before any deals start happening .

The invention relates to use of TLR9 antagonist in a method for treating cancer and/or chronic infection by downregulating Clever-1 expression on alternatively activated immunosuppressive macrophages.

This is from the patent text .....

"Now, the inventors have surprisingly discovered that a Toll-like receptor 9 (TLR9) agonists can be used to downregulate the expression of Clever-1 on alternatively activated macrophages. TLR9 agonist was found to reduce rapidly Clever-1 expression."

I am no scientist - so welcome any comments on this - but does this now mean that Faron have now two tools for reducing Clever 1 expression ??

I'm going to try and re-read the patent blurb - but on first scan this seems verrrry interesting .

Yes seems to be more of a study into looking at Clever1 levels as a tool for predicting prognosis , and it looks to be a tricky problem pinning down any real correlation as the levels change during the stages of cancer , and even the sample collection may not be perfect .
So much involved at these levels that's for sure - you can understand why they can't just bang out a quick report explaining the data on these trials .

Yep , even I'm at a loss to chip in much these days .
We have literally nothing to go on .
We know they have gone back to work on the data , and we know that they are working on the final dosing and biomarkers etc , to define the next trials .
We know hibiscus is ongoing , starting late Aug , so at least 3 months into that trial with interim data supposedly expected in Q4 . But the Trial site has had no additional study locations added , so we are in the dark as to how many patients have been dosed .
We know they advertised for a Clinical Director - based on Haematology and Immunology studies .
We know they are now looking to recruit a new CMO .

We know they are fully funded at least going into 2022 , so at least that issue is sorted , plus the bonus of a few extra million from that arbitration .

Interferon is getting nothing but positive press in medical journals and studies . Just needs trials to prove up the most effective delivery route , and at which stage to be best deployed . With the data they have on Traumakine , this really has to be the best shot Faron will ever get . Despite Covid being the cause , you'd hope that they could replicate the ARDS data to some degree , with the steroids removed .

And the Bex story is really compelling . We are just left to ponder and speculate at the moment as to whats going to happen .
Surely they will have some good news on the biomarkers and dosing at least ( this is my worst case scenario ) , so they can move onto P3 trials .
But what about soluble Clever1 - this was the big ace up Farons sleeve that we are still waiting to hear about .
That would unlock so much more value , as well as the neoadjuvant combis that are planned , along with other Bex possibilities .

But its all just speculation . We've had a few months now of these repetitive presentations , the last few of which have been pretty tedious . I know it doesn't have to be exciting to be profitable and relevant , but even the ESMO abstract was nothing new to us lot .
It was like reading us a great book , but before they'd actually written the last chapter , that wrapped it all up to a conclusion .
I've given up making predictions now , so will just hold on tight now unless I see reason not to .
I take the lack of visibility at the moment as a sign that they are busy working on stuff - be that some kind of deal , or some very detailed analysis to present to us .
Something will come , and as normal , it will probably surprise us.

Update yesterday to the Activ2 site - they just added another hospital in the Philippines .

Had a small top up this morning ( last one I told myself ) . Unusually it took a fair while to get a quote ( when it was at £2.00 )

Explains why he came off the Trial sites last month .
People move on for many reasons .
Perhaps the job would now entail much more work in Boston , who knows .
Family sometimes can make those changes hard etc .

If you've ever watched a few hours of pharmaceutical commercials on US TV , you'd wonder why people ever took half the drugs they are advertising , given the list of side effects they come with , but people obviously do .
Unfortunately I think the majority of people still have faith that someone in a white coat is always on their side , and if they are offering you something , it must be a benefit .
It's only a minority that used to question that , but that minority has been getting larger every year .

I think acts like approving this pill will only deepen the mistrust many have .

Well the UK has ordered 480,000 courses , maybe they can put them in one of the containers full of unusable PPE .

SNG is still completely under the radar to so many people outside of clinical studies and us investors .
When i talk to people outside of the investing world , they generally have no clue . Most couldn't name one single therapy , apart from steroids maybe , or Trumps bleach , and when I mention Interferon they don't have a clue what I'm talking about .
When the results come out and we get the front page headlines , given what state our NHS will be like in mid January , never mind the rest of the world , I just can't even hazard a guess as to what demand there will be for this therapy .

Sold all my ODX shares on the bell this morning ( only a few grand , but just threw in the towel ) . After tonights meeting , I think I might be counting myself one of the lucky ones walking away with 28p .
Normally I would hold for the initial purge and wait for a bounce to sell , but today I just wanted them gone.

On the bright side , I put that dosh straight in here , so at least I clawed 5% odd back .

News is certainly due . Wouldn't say overdue yet , as we know what they are working on , but we just have to wait for it to arrive , be it Hibiscus interim data , Bex biomarker /dosing data , soluble Clever 1 data , or some other update .

Picked up on a tweet regarding Synairgen being on a German share index today , so had a hunt down the list and there was Faron Pharmaceuticals on the index too . Not that exciting , but at least it shows a few other people know we exist and have placed some value on us by including us in their index .

Just got used to the 1 sells , now in the midst of all the phoney sells we've just got one single buy today , of guess what , 101 shares( and at over the odds ).

If this does go to £20 , it'll go down as the clumsiest signalling effort in AIM history by a country mile lol

Definitely a buy . The round numbers may indicate its Mr 5000 loading up in order to satisfy demand on the Nas should some news arrive . Mind you , the nailing of the bid to 330-333 for so many weeks is just plain odd .
It did rise a tadge on Friday to 335 like Santhi said.
We are just in one of those periods where nobody is buying or selling apart from the MM's .
