As a dual inhibitor of Tyk2 and Jak 1 it is predicted to have greater efficacy over Tyk2 alone.

737 was trialled in combo with ldg ie low dose gemcitabine.

although mentioned sra737 with ldg and cisplatin a platinum based therapy you will find nothing mentioned of this.

gemcitabine and cisplatin would be referred to as gemcis.

whether any trial took place with gemcis l do not know but what l do know there was a degree of toxicity associated with 737 gemcitabine and cisplatin.

i have the pdf on my laptop my no access to at the moment.

however in light of the above it was recommended that a combination therapy be trialled of ldg 737 and an ici ( immuno checkpoint inhibitor referring to a pd-1 inhibitor which significantly increased the effectiveness of 737 ldg combo.

with respect to a cessation or han**** of a compound eli lilly halted development of pexasertib from memory due to a toxicity issue associated with heart problems.

eli lilley ceased development of prexasertib april 2019 and acrivon gained an on licence june 2021 a period exceeding 2 years.

in addition the chinese company bibert have developed their own chk1 inhibitor to be used on combo with ldg. the wording is nigh as exact of the so description of 737 and ldg.

bibert also noted the cessation of development of 737 in 2020. bibert also intend to develop combo therapy with wee1 and or a pd-1 inhibitor.

i am not 100% sure here but l believe the bibert chk1i is available for use in clinic. claims of very successful tumour reductions have been made by bibert.

737 is proven to be too a toxic combination with platinum based therapies ie cisplatin.

Cisplatin is a relatively cheap yet effective form of chemo.

737 can still.be obtained for preclinical investigational work.

Of course since the return of 737 by Sierra Oncology for whatever reason, GSK has suffered setbacks with the 5.1 billion Tesaro pipeline.

Nirapirib PD-1 inhibitor already evaluated in preclinical with 737 and LDG by SO.

Preclinical results with LDG, SRA and a Pd-1 inhibitor were very impressive.

Bearing in mind that SO had undertaken this work and was indicated in their final design plan where SO would have needed funding to continue.

Indeed on SCLC preclinical in mice all mice had tumour reduction and 8 out of the 10 had no detectable tumour up to 60 days post cessation of treatment.

A very promising route to pursue.

Should GSK have any interest in this then my opinion for what it is worth will be a 3rd party to continue development in this area.

Absurd as it may sound Sierra Oncology would be most qualified to continue this, as not only familiar with compound but have carried out excellent work previously as indicated by approval of Momo and the clinical studies carried out on 737 in mono as well as 737 plus LDG.

The RNS informed us that full trial data results.

Full trial data results would not be required for a deal of some sort with a pharma.

A deal could happen at anytime. However let me explain to you as you clearly have difficulty grasping the modus operandi of Sareum.

Their intention is to licence at end of preclinical to early clinical stage. As milestones are reached so the value of SDC1801 increases as does the value og Sareum.

At end of phase1 dose escalation l would put a value of minimum 250 million on 1801.

I will remind you that albeit at a later stage of development A similar albeit not exact pipeline of inhibitors reached a deal size of 4 billion dollars.

Whilst l do not see a value of that amount being reflected in the Sareum market cap at the moment it will on successful dose escalation results considerably raise the value of Sareum.

Perhaps you would be so good as to give any reasonings as to why this should not be the case.

I suggest you take the death spiral approach out of your argument, but then again l am of the opinion you have nothing else to go on.
