I do find it very odd that those who claim.to have been here years choose to knock the company.

Some only started posting here very recently.

What was their objectives when they invested all that time ago?

Was it to make money?

If it was then why not sell when from the lows to the highs is around 5000% increase. Clearly they did not take advantage if this.

If it were to invest long term to the the point of an on licence or commercialisation or even takeover and take the corresponding financial rewards than this, then surely they would be looking at ways forward or constructive debate without spouting out constant negativity.

Frustrations l can understand from long term holders here and yes they are pretty much genuine looking on the brighter side of things. They know who they are.

The self confessed who claim to have been invested here years and only recently posting on LSE, l find in most cases very dubious. They have never contributed anything of any significant value, either company or industry related.

Those that claim to have been here years would have invested when the price was low. For what we have in the pipeline and our current value, the company value is grossly below where it should be. That my opinion.

Very little difference in potential gain here at these prices and the gains that have been made in the past. Future gains will come from satisfactory data results and that applies to all of our compounds.

Any discussions that may be taking place will certainly involve suitable compapatible in combo partners. To enable any agreements ie licence then trial progression will rely heavily on the intentions of the pharma.

As much research has been carried out into areas such as SCLC, HGSOC, TNBC, Anal, Head and neck cancers then this needs careful evaluation py a pharma.

Clearly a pharma will not wish to pay over the top whilst CPF will not want to on licence gor peanuts. Any negotiations will take months with a month or so at least to write up with the legal aspects a contract agreeable to both sides.

SDC1802 l see we sit in a very sweet spot Aith Sareum continuing with translational studies and at a time when the critical aspect of finalising the compound candidate and identify the particular cancers likely to respond best to 1802.

1801 should pose no threat with regards to efficacy due to attention to detail carried out in preclinical. In vitro and especially on in vivo.

Sareum BoD appear very confident in their SDC pipeline.

Yes the current market cap is a very poor reflection as to the worth of our compounds. News of any significant development should propel the SP considerably higher.

As you say there has been considerable progress made over the past 10 months. 1801 in clinical trial and chk1 available to on licence.

Who stated 737 is apparently being considered by pharma for in combo for trialling?

Whilst there is no direct statement that a pharma's is considering in combo therapy involving 737 there are very real events that a checkpoint 1 inhibitor in combo take place.

Acrivon with Prexasertib trials in combo with LDG and or proposed Parpi or PD,1 or even Wee1.

Bebet the Chinese pharma are using a CHK1 inhibitor with LDG and propose in combo developments of their CHK1 inhibitor to a nigh identical path to that of Sierra Oncology with 737.

Since 737 showed a superior safety profile in some aspects over prexasertib and showed good efficacy in mono. ( we need take into account here how the checkpoint kinase one works)

The added advantage is that 737 can be administered orally and showed no indication of any cardiac problems associated as was with prexasertib.

I really wonder on here how some people understand the pharmaceutical industry!

There is nothing wrong with an Aston Martin but it is not going to be bought by someone to use in heavy haulage.

Likewise a checkpoint1 kinase inhibitor is unlikely to be purchased by a company that does not have access to suitable compatible in combo therapies and the research abilities to carry this out. Also the finances now at a time when we are verging on recession.

We have had good preclinical results with PARPI in combo and impressive results when used with LDG. Niraparib was one.

We proved excellent results preclinical with Pd-1 and LDG. T Sen gave a brilliant talk at ASCO 2019.

Been posted here many times.

An area of slight concern which required further investigation was toxicity in combo not as a direct result of 737 but rather the PD-1 or Parp inhibitor and long term effects associated with.

Not all inhibitors are the same as there are as we know differences in efficacy and toxicity amongst similar similar treatments. Pd-1 and particularly Parpi are no exception.

A later generation Parpi in combo with a checkpoint kinase 1 inhibitor and LDG would in my opinion look to be a valuable asset in the treatment of difficult to treat cancers and especially areas of unmet need.

There is a later stage Parpi in development that has little to no off target effects that are associated with earlier Parp inhibitors.

There is no getting away from the contribution that a CHK1 inhibitor in combination with regards to certain Indications in oncology.

For CHK1 to benefit it needs to be partnered with a good performing and highly compatible form of in combo treatment.

Regarding 737 TM stated that development would be in combination therapy and not mono at 2022 AGM.

They will know of certain developments as they have a financial Interest. Until any agreement is made with a 3rd party we will hear nothing.

Funnyguy, why did Sierra not progress?

It is very much akin to saying SDC 1801 did not progress as MHRA were unable to approve CTA application.

It takes months to overcome issues.

It was well over a year (closer to 18 months before Acrivon obtained Prexasertib from BMS.

I think some people here look it as if it is something for sale on Ebay.

The issues here are complex to say the least. It would take at least 3 months nowadays just for a contract to be written up. This would follow long periods of negotiation which take around 6 months to a year.

Apart from the complexities of patent protection including extension of patent protection by applying for patents in combo therapy there are a myriad of other details.

First of all you need a pharma that has either comparable in combo compounds or gain access to such.

Research purposes are one thing, the use of a competitors compound in clinical and human trial is something else.

We are compatible with Parpi and PD-1 inhibitors.

Parpi are not squeaky clean and have their own inherent non target side effects. Later generation Parpi much improved.

So there is plenty if scope for involvement of CHK1 in combo therapy.

737 is not very compatible with platinum based therapies ie Cisplatin which is a relatively cheap but very effective form of chemo.

Yes it will and does take time due to the intricacies involved.

Since Covid and people working from home things now take considerably longer to resolve.

I suggest some have a look at a typical contract.

Oh yes some will say copy and paste is you need to do.

Oh no sweeties, there will be major and minor details that need do be resolved by communications endorsed by both sides legal teams before the contracts are suitable to be signed by both parties.

Now this may very well be going on at the moment or maybe not.

I see very little if any benefit by being brokered by PH.

Sareum will come good Gunner, it has had its ups and downs in the past. 2.1 p down to 0.18p years ago. That is over 90% down.

SDC gives a satisfactory safety profile and performs as well as expected in Psoriasis alone then the market Cap will be heading way North despite the modus operandi of RF.

Bare in mind we have 27.5% financial interest with 737 and 1802 for oncology.

It really depends on what you chose to focus on Gunner.

There are those that are in it for a fast buck and in this case has been taken on by our newly acquired deramper. Some invested and do have concerns and will see both sides of the coin.

Whilst the raising here of finance may not be seen to be the best option, this option is by way far better than the collapse of a company due to lack of finance.

For Sareum to continue in clinical development they must be able to prove they can raise the funds before that part of a trial is approved. No different than over here where the MHRA were unable to approve application. We still never found out why not but highly likely to be concerning the pre clinical toxicity data with no MTD established.

However, we can now continue with dose escalation trials which have gone very well as expected by the BoD.

If like me you are here for the long time ie for on licence or maybe a buyout ( that cannot be ruled out even by that Mr Magic wand) then you will have no significant fears.

You quote a statistical figure of 7.9% as a success factor. This implies 92.1% failure.

Take for example BMS with deucravacitinib. Now correct me if l am wrong,

It had great results in Psoriasis

It failed in UC. It has had several trials around 4 which it has failed 1. Now we have a valuable commodity that has subsequently failed to meet its end points.

Allosteric Tyk 2 does have its limitations

A drug maybe successful in one indication and fail at 12 others. It still therefore has a value in the indication it has been successful in.

SDC1801 has been granted dose escalation trials.

A year ago the MHRA were unable to grant approval.

The SP of Sar was hammered at the time an although trials have been approved there has been very little recovery.

As you state it certainly is not a full gone conclusion but at the same time 7.9% success factor to me seems extremely pessimistic to say the least. That is how l see it. Based on what l know as opposed to based on what l don't know.

As milestones are reached then then the risk lowers considerably. The most important as we all know is the safety profile. I am 100% confident of a satisfactory safety profile.
