Good post. I can think of no reason as to why 737 cannot progress to a phase 2 combo with LDG and maybe PD-1 inhibitor or similar. Excellent results l believe we're achieved albeit preclinical but l believe would have given GSK Tesaro bought pipeline a run for its money. GSK squeezed their pipeline into Europe as a treatment in an area of unmet need and from memory was not without its problems with regards to safety profile which was under the watch of the EMA..

We would not be the only chk1 inhibitor progressing through trial if and when progress starts again. Addressing the DDR pathway is an extremely important aspect of cancer therapy and l am of the strong impression that although there is competition to get later ideas into development and treatment we have not properly given good potential earlier compounds sufficient chance to eclipse maximum potential.

In combo is the way forward and has been suggested by research groups many times.

Our compound achieved as much as you can expect from the mechanisms of a checkpoint inhibitor working. The compound is fine itself but by the nature of the way CHK1 inhibitor works it needs in combo therapy.

Sareum now have designed and developed their own compound in the area of auto immune. They have decided on the pathways used and the sectivity of the compound. All the molecules in the compounds are patent protected as are the compounds themselves as well as the controls of manufacture and of course very nicely put in a capsular formation.

As an example here to show advantage of capsule formulation has anyone ever experienced the benefit of the Neurofen capsule over the tablet?

With regards to 737 it would not surprise me if an indication would be in SCLC of which very favourable results were achieved in triple combo 737 LDG and PD-1/Parp inhibitor albeit preclinical.

As we know the small pharmas have suffered significantly over past 18 months.

A spike with covid in the sector then a massive fall.

This small sector being the backbone of future improved innovative therapies and treatments.

It needs financial assistance in the current difficult financial world economis climate created by the war in Ukraine and to a lesser extent Israel.

Much investment been put into companies that are in anyway connected to military production whereby you are basically guaranteed a return on your investment. Hence the biopharma market with potential far higher financial gain but associated higher risk of failure has subsequently suffered.

The question now is has the military production reached or is reaching its peak and has the small phsrma sector bottomed out?

Unless significant finance via investors is made then small pharma will suffer which will have a knock on effect by creating a void of new innovative drugs to the big pharma that are suffering from patent expiry pipeline and the threat of losing out to generic produced compounds.

For investment to be made there must exist a certain degree of predictable success.

As for a benefit to Sareum notwithstanding the restarted development of 737 but the development of current trials with SDC1801 and with the likely success of a more than satisfactory safety profile then l am certain that we will attract significant investment on data indicating the aforesaid.

Biomarker data will also come into play here and when combined with a satisfactory safety profile then there really should be now problem attracting investment.

A very pertinent point you have made there rv84.

Biomarkers which have been used in dose escalation studies will indicate Indications that 1801 will best perform in.

I don't see any problem with 1801 doing well in psoriasis. A PASI score of 75 reached over a 16 week period with Deucravacitinib. I would hazard an educated guess we can reach that score in 12 with dual inhibition.

However, this is just one of many Indications that

1801 has the potential to do well in. We will not be best in every indication ut then again that is not the objective here.

At present we need good safety profile data from clinical trial. Once obtained then we ought to see a considerable up tick in SP.

With regards to RF the only headache l see at the moment is the original drawdown of 2 million which does not attract interest. Whilst additional shares will be required to cover the shortfall the later drawdowns attract interest and therefore I do not believe it is in RF interest to sell these shares.

A waiting game at the moment.

Good evening Luton l share the same opinion as you in that the company can now be evaluated with the news of developments of 737 and of course we need the data for 1801 which is crucial and expected in H1.

Once developments progress to phase1b then the company can expect offers for takeover.

Good news in that 737 is being progressed.

The importance here is that it is being progressed so do not place too much concern on the low upfront payment.

Although we know that there will be at least one indication we do not know what it is, what we do know is that very satisfactory preclinical data has been produced in SCLC. Ie PD-1, LDG and 737.

Good news was also released with regards to anogenital indication ( I don't believe that 737 was expected by Tim and co produce this type of success). We now have good data albeit preclinical for 737 and A Wee 1 inhibitor in CRPC.

A small company in a sense is far more adept in carrying out further research in attempt to find the best Indications and also particular attention for most effective in combo therapy and also dosing regime.

Once an indication or several Indications of satisfactory efficacy are found then it may well be that the small pharma will sell to large pharma for commercialisation.

There are many Indications shown by similar preclinical xnd investigational studies with 737 that have indicated further development.

Head and neck cancers was one example and of course this includes the brain ( early work by M Garrett) an area with an unmet need. Another was SCLC with Triparna Sen talking about the micro environment and singing the praises of 737 LDG and another inhibitor (not sure if Parpi or PD-1 here)

Also note that SO had undertaken considerable development with 737. 737 with LDG. 737 with LDG and Parpi. Indeed they were at a stage of finalising trial design in combo with either PD-1 Parpi or a Wee1 inhibitor.

The potential here is not to be underestimated.

Clearly it would be of benefit as not only for AZ but also Sareum and more importantly a treatment in later stage prostrate cancer patients where inevitably leads to metastases.

Not all cancers are the same as you know. But someone somewhere has instigated this investigation in the first place and if you say that data had been derived by May 23 then intention to carry out this investigation would have months before hand possibly around the time that SO announced return of 737 to the CPF.

Very little point in starting before this as the decision to return had not been made and licence still held by SO.

More to the point here is had this interested party attempted to make any contact t with SO when they held the onlicence?

With regards to treatment l am of the firm opinion that it may well be lower dosing required to treat CRPC than HGSOC.

HGSOC becomes very aggressive and can develop and normally does acquire resistance to treatment very quickly. Prostrate cancer by comparison is somewhat slower to develop and may well not be as prone to develop treatment resistance over such a short time as HGSOC.

Clearly there is interest and l dare say that should this be the case then we will be updated in the not too distant future.

It is evident here that there is interest in 737.

My view of this indicates to the leaning of further development of 737 not by a large pharma but to a company similar to the lines of Acrivon that took an onlicence for the discontinued Prexasertib.

AZ discontinued development of Advorsertib in 2022. Basically adverse effects.

However whilst this can appear a tad gloomy it is worthwhile noting that in combo therapy can allow reduced dosaging that will reduce toxicity issues whilst at the same time maintaining or improving efficacy.

Already proven with CHK1 inhibitors and Gemcitabine in early and mid stage clinical trials.

Reports by Sierra Oncology in combo with LDG and other reports by the Chinese company Bibbert indicate greater efficacy with satisfactory safety profile. Indeed SO we're looking at further in combo therapy of Wee1 Parpi and PD-1 inhibitor in there final design stage prior to return of licence to CPF. Bare in mind here that there was no development only intended development indicated by proposed final design stage that stood stagnant for nigh 18 months.

From memory SO would of have to hand back 737 as this was part of the original licence agreement whereby no updates on any developments in a 12 month period. The CPF granted a change in the milestone payment due to the financial situation of SO with its change in commitments due to development of Momoletonib. Yes they robbed allocated funds to 737 too. Exploring methods of raising funds which never materialised. A long delay then the long awaited final trial design.

Back to the incombo therapy. Whether or not the addition of Gemcitabine has the potential to further increase efficacy without compromising safety in this particular indication l have no idea, but may well be a possibility.

I suggest the most likely outcome here is an agreement of some sort with a company that specialises in further investigational work that can develop inhibitors whether currently in use but more particulary discontinued that can be by utilising combination therapy to maximise the potential suitable to be approved for clinical use and importantly for us commercialisation.

Some way away? Yes.

An agreement here of course would not only involve the development company and the CPF but also the owners of AZD1775. A 3 way negotiation along with all the other aspects including all clinical data along with its Indications.

This scenario alone would explain the lengthy time span with no news on 737 development.

I don't think RF will be off loading shares as l have said before, the initial track of 2 million does not earn them interest so understandable that they sell to recoup funds ASAP. The following tranches attract interest so therefore in RF interest to to make as much money from this as they can.

My own view here is the SP had bottomed out at around 60p a share. I did mention this a couple of weeks back.

On no selling by RF SP ought to climb a tad.

Not the best form of finance l will agree but at the same time should the SP climb then this will be of further benefit as RF can see a lower risk and at the same time will want to loan money.

Of course 737 could get a licence but not think this side of Christmas.

Just an addition here with regards to clinical trials in OZ.

Sareum has not yet carried out any trials with regards to 1801 in Psoriasis patients. Clinical trial information and data results would be kept closely under scrutiny.

Whilst l have the utmost confidence in satisfactory DE results and indeed the treatment in Psoriasis, we are still at DE stage.

I am of the firm belief that in clinical trial we will have favourable results with reaching a PASI score of 75 over a 12 week period.
