Covid19 is far from over. Whilst most new strains present mild symptoms and our bodies immune systems can cope there exists the possibility of a covid 19 strain evolving of deep concern.

There are well documented reports on Covid 19 and relate to feron cleavage ie the addition of added material. This does not happen in nature and is thus created by reverse engineering.

There was reports the corona virus had been spliced with HIV.

The covid 19 is but a mutation factory in people with HIV. There is a well documented report on an African woman with HIV that had had developed over 30 different mutations of Covid 19.

Ultimately new strains overcome our bodies immune defences but it is the cytokine storm that can develop in some patients that require medical attention at times on an urgent basis.

If we are not suitably prepared then there will be severe repercussions that can result in death or prolonged periods of incapacity.

Where we fit it l do not know. Dose escalation results which are good is definitely a step in the right direction as a possible form of clinical treatment.

I of the very strong opinion that our SDC1801 of which there have been ongoing discussions with interested parties which have been going on for what seems like an eternity, will reach deepening discussions with either a partnership, on licence and increasingly a probability maybe of takeover from any time onwards on positive safety data with regards to our dose escalation trials.

Interested parties require data ie something concrete as a basis to work from.

On good safety data and the understanding that Tyk2 Jak 1 is very highly likely to prove greater efficacy in Psoriazis as opposed to Deucravacitinib it will enable a pharma to evaluate the possible value of SDC 1801 taking into account just this indication.

Biomarkers will be used in later stages of the phase1b clinical trial and this will give strong Indications as to the efficacy in other auto immune diseases.

What we have here is a series of stages ie

safety profile,

efficacy in Psoriasis

establishing and identifying further Indications that the 1801 compound can probably achieve greater efficacy in (There are many) along with

an acceptable safety profile above all else which is currently used in the clinic.

With regards to 737 which is fundamentally in the hands of the CPF, my take on this is that there are interested parties.

The dilemma here is funding a partner where a checkpoint 1 inhibitor will fit into its pipeline.

You will be looking at a pharma that has Pd-1/PD-L1, possible PARPi or various other forms of treatment that can make use of what a DNA Damage Repair inhibitor will work with to give greater efficacy but limit associated side effects. In effect the upside in efficacy must not be destroyed by adverse side effects caused by a drug. The benefits must outweigh the down sides.

There are many possible in combo therapies that may prove highly beneficial here and along with preclinical and clinical work taken out by SO and a plethora of other investigational studies by other highly regarded establishments for example CRT to name but one there will exist a way forward for the possible commercialisation for 737 in como therapy. Highly likely more than one.

Opportunities arise for any serious negotiations once certain stages are reached particularly relevant to SDC compounds. That is the nature of the game.

Lots of ifs and when's but many milestones have been reached so far. People forget at times how far we have come.

I will also add that Keytruda is a PD-1 inhibitor and was one of the options labelled by SO in 737 combination therapy either on its own or in combo with LDG.

737 would certainly fill a very appropriate gap in a pharma pipeline.

From memory here but we do know that much research was carried out on the former candidate prior to 245737 prior to tweaking of the compound on head and neck cancers by M Garrett and Co for ICR. Results showed significant positive results.

As stated in the link you posted here, by definition there still exists a gaping whole in an area of unmet need with regards to head and neck cancers and as we all already aware (apart from a couple of recently joined posters) still areas of high unmet need in HGSOC anal and genital cancers as well as small cell lung cancer.

Very coincidental l will add that after the availability now of 737 as back in the hands of the CPF Z-along with all relevant data from SO and indeed other investigational bodies that Merc have now decided to discontinue development of keytruda plus Lenvima.

Taking into account recent events it would not be out of the question for Merk to have a keen interest in 737.

Interest here l may add may already have been shown giving Merc an option should the results prove disappointing results.

A big if certainly but certainly not out of the question.

The reason they are here is to spread doom and gloom and create negative sentiment.

They both tell lies. But of course they do have some success in sucking the weak and gullible into their mindset for their own gains.

They admit to shorting the stock at least there is a degree of honesty there.

They know know little to nothing of sareum or its compounds. The method of finance used to enable continued development of 1801 and 1802 they have portrayed as a death spiral.

I will rind you that Sareum has raised money for development and not to pay off existing debt unlike many companies.

As for rumour that Sareum will be selling for multiples of current share price, l myself have not heard of this rumour.

What l have heard is that some people have postulated that Sareum may be bought out.

Sareum stated in their RNS that this is what they considered their best option. They did not state it was their only option.

Good morning. I believe the 1801 debacle of performing better than Deucracitinib misinterpreted here. At the AGM stated may not prove better. That is a mere possibility and open to being misinterpreted.and can only be confirmed by clinical trial. The statement can be interpreted taken both ways.

Tim Mitchell stated that they believe 1801 to outperform other inhibitors and that definitely included Psoriasis. He stated it will prove better in some Indications in some areas and not in others.

TYK2 Jak1 has far more efficacy over Tyk2 in Psoriasis that is a fact now proven but dear old Beprocitinib dropped the proverbial boll..k with problems associated with JAK2.

Sareum has greater selectivity of Jak one over Jak2 than Bepro and that us stated in the AGM.

It is just no good to select a piece of information saying Beprocitinib has been discontinued on trials involving certain Indications. Yet put no reasoning as to why eg is Bepro still being developed in Psoriasis?

Limitations as to dosing related to adverse effects due to Jak2 put a little bit of Kybosh on Bepro. It in no way whatsover disproves the possibility that Tyk2 Jak1 has greater efficacy over allosteric Tyk2.

One thing Tim and Co cannot state is 1801 is superior over a certain competitors compound. It would effect market price and would be false leading to misconduct.

So to put it may not prove better than ...........

certainly does not mean that the BoD believe it will not be better.

I believe Beprocitinib suffered problems that resulted in a reduced treatment dose of around30 mg per day. The primary belief by Sareum was true to adverse effects associated to Jak2. We have greater Jak1 selectivity over Jak2 than Bepro. Pfizer passed Beprocitinib on to Priovant.

All about getting the required or to be precise optimum selectivity.

So yes C79 we can take Beprocitinib as the main challenger, however l am of the belief that the attention to detail that Tim and Co have taken over candidate selection we will definitely have the edge.

Thanks HbD for your contribution regarding our performance of 1801 over competitors. Tim did state in an interview from memory with Proactive investor? He believed we could out perform Deucravacitinib basically as you have correctly stated of dual inhibition.

So as it stands we ought to have better safety profile enabling higher doses over Beprocitinib without adverse side effects and perform with better efficacy over allosteric Tyk2 Deucravacitinib.

Take into account patent protection, capsule formulation and patented crystallisation structure of our Tyk 2 Jak1 compound which in effect protects the manufacture of our compounds then this should put us at not only best in class but also extremely attractive for a would be interested party.

The science comes into its own in the clinical trial. Tim and Co and those chosen in the preclinical development of our compounds have the benefit of learning from the failings ( what Tim and Co percieve the problems to be) of competitor pharmas.

Having been involved with the development of SRA737 and Aurora +FLT3 it will give them a degree of expertise in the translational studies on 1802.

Deucravacitinib is not the most effective treatment for Psoriasis. So there is definitely room for improvement
