Meanwhile the point that delivering any dose to the TME (eg 16x, 18x,20x) whilst exposing the heart to a "standard" dose of dox is what we are trying to avoid. Easy to miss the simple stuff though. Only Avacta know the human numbers but building fantasy data based on the mouse model needs to take this fact into the conclusions. Added to that the current c4 dose is nowhere near the mtd given in mice and if it were achieving even 10x the conc in tumours I suspect we would move on to next stage. Lack of side effects coupled with therapeutic concentration at tumour site would seem pretty good at this stage. Responses are even funnier than original given OP knows it is "a what if.

Hi. Can I just check where the 20x dox concentration in tumour has come from. RNS says therapeutic concentrations (ie 1x) at double dose and the mouse model is 18 x at 6x normal dose. Might have missed something.

I thought the point of MMs were you didn't need two sides of a trade. By holding inventory the MM can buy or sell without having to match. They have to eventually balance the inventory and will adjust prices and spreads to meet this need. I'm fairly simplistic but believe demand and supply effect the SP greater than anything else. Not surprised if we have days of large sells and days of large buys - and the SP goes up and down a bit. What individual holders be they large or small aggragates - happy with returns this month - confess it hurts to make paper loss on the day but maybe set the graph to 5 days or 1 month if its too much. Even better come back in June when a licensing deal has been signed

Whilst a more tolerable dox will result in larger doses and more frequent dosing I think it's more likely that financial returns will rely on securing a bigger part of the market rather than expansion. Health budgets are not unlimited. For Avacta that just increases their value to big pharma - fir avacta to gain market share someone else has to lose it

Add in penetration of a tumour and you could see a how more frequent dosing delivers therapeutic levels of dox for longer whilst also sparing cardiac tissue. Ava6000 changes the approach cause low AE opens up so much in way of dosing. Add this to cancer therapies are nearly always used in combination then the BD guys in pharma oncology will already have this on their sights. I know people disagree but that's why I think ava6000 will be licensed out - working out best approach rather than better than normal dox could be the next question and that will take a company with money, infrastructure and experience. Puts avacta on solid foundation and enables next drugs to piggy back.

The chosen posology will be interesting. What they want is a flat a pK as possible so that the Fap enzyme is working flat out all the time rather than becoming saturates and unconverted ava6000 excreted. More frequent dosing is a move towards that. Suspect the trial extension is intended to shed light on it

How many Pharma execs do you think have looked at Avacta and have sight of info you don't have. Would be daft to buy these huge chunks individually.

Call for abstracts is due soon with closing date of 20th Feb. Science day very shortly after.

1. Human data is more impactful than animal. Expect oral presentation
2. AS has said study is data rich
3. AS seems very excited about the biopsy data. Study on-going but given news to date expect interim analysis data

If submit abstract that data may be released earlier than science day. Doesn't always follow but given sensitivity not to release opens massive door for new insiders. Probably want to avoid that

Intellectual property concerns / control resource prioritisation

Just read a post about dox being used to manage rather than cure.

Dox is unlikely to be used on its own. One problem for its combination partner drugs is penetrative the tumour. If avacta can increase the "permeability" of the tumour (thinking those photos from mice" then perhaps it is a double whammy.

Not putting too much store on biopsy data since its a bit opaque. AS waving a hope rather than fact. However, the lack of side effects is meaningful. If they didn't think there was efficacy to be had why waste money continuing- added to that the data committee would stop them.
