So consider how you could calculate how much ava6000 is converted to dox by the FAP enzyme over time....

That gives an indication of how much dox is hitting the tumour. That is the number that will affect the SP so grabbing pom poms and singing songs backwards just mis represents the good news when it comes.

So you believe ava6000 is a like for like dose with standard dox and 100% converted. Hence your repeating the same question and failing to understand the answer

The AUC may help you realise your misteak and give us an idea of how much dox is hitting the tumour. Then shout the proper facts and stuff from the rooftops rather than undermine what is some fantastic science.

If you understood what an AUC is then you wouldn't have to ask but feel free to make it up. From your response you are sticking with your original idea but maybe change the thick as mince target.

Only problem with your calculations is they assume 100% of ava6000 is converted to dox which is probably not true.

Given unconverted ava6000 is inert then it cannot have either a efficacy or a tolerability effect and is eliminated before either of those things. So comparing ava6000 dose to dox as if they are like for like is not valid. Sorry.

It's great news that we have heard about an increase in dose - this will almost certainly increase the concentration at the tumour which is the data we want especially area under the curve. This is not available yet.

Excited yes. MHRA have assessed - that is what I took from RNS. Not approval but a degree of endorsement that the trial adaption is valid and in the interest of patients.

With ava6000 the hypothesis is that very high doses of dox will be generated at the tumour but very low levels elsewhere. Initial data looking at tumour biopsy and systemic levels look promising.

MHRA agree that trial is worth continuing to higher doses. Suggest they believe further benefit possible. They will have seen the pK of the leaving group.

The trial design will be to demonstrate the next stage. The number of patients will also reflect this. Hoping regulators will somehow bend the rules for avacta is a pretty long straw and one I hope avacta is not relying on. Lots of promising drugs out there some with absolutely no alternative and therefore with a potentially better claim.

That said the generic nature of dox should shorten the process somewhat but this will be agreed with fda before trial initiation not after.

Proven means something different to regulators than some of the commentators. Frustrating to meet these requirements but probably cause they have patient interests first rather than impatient share holders.

Could build a model though based on animal data which they do have.

In comms they suggest that normal dox eventually has equal concentrations in all compartments - although this will not be precisely true the animal models will indicate a level of tolerance around this.

We also know the effect if dox in vitro cell models at different threshold concentrations. Could work it backwards. If you see an effect then that concentration has been achieved

Tumours may be difficult for dox to penetrate so maybe dox isn't ****genous through tumour. Works in ava6000 favour if multiple dosing to achieve flatter pk at TME is an option.

All a bit mathematical and built on assumptions but trial committee will be aware.

BITL. Truth is without RNS and therefore developments there isn't a lot new to be said here. Thinking if you need the break, and most here have taken them, then perhaps just set RNS message up.

Then if something does change we can all come back and join in the bun fight. Our tolerance of SP in both directions is in the range of anything that happens in mean time. Strange when we have to be rotated for some R&R from a board :-D

On slightly different note is the cash position which is straight line to 2024. That calc ignore potential milestone payments from other programs of which we know very little. Issue will have to be addressed at next update so perhaps some good news due. Perhaps not :-)

Volatility is the standard deviation of a stock’s annualised returns over a given period and shows the range in which its price may increase or decrease.

Personaly I am happier when the SP is higher but I have held from below 20 to the ATH and almost back again. Yes I want the SP to be higher but the news that we all have is out there irrespective of whether it was rns or not - unless you believe its the channel not the info that makes a difference.
