I think it is to do with the build up to the Modi1 trial starting.
It probably started with the Paris conference with no less than 6 posters.

I don't quite know why she is presenting at a neo-antigen conference.
Lindy has previously said that she is not that keen on neo-antigens.
Perhaps it is just a reminder to her audience that Modi1 targets very commonly expressed epitopes and the mode of attack (CD4+ Tcells) is novel.

So, the idea is to determine the neoantigens for an individual patient and then create a vaccine specifically for that patient.
Time consuming and costly with no real time to thoroughly test the vaccine.

I would love to hear Lindy's presentation at this conference to a group of fellow scientists.
Likewise the lecture she gave when presented with the Waldenstrom prize.

I guess that with the separate enolase patent for autoimmune disease, Scancell wanted to emphasise that they had the patent for cancer treatment.
I assume Karolinska are looking at ways of blocking the enolase epitopes.
Perhaps Moditope is giving them clues as to how to do this.
Who knows, Avidimab may also help in this area.
Mabs to block the enolase epitopes i.e not binding to APCs but binding to the areas affected by RA.

Do we know why Scancell considered that a patent specifically for enolase citrullinated peptides was required over and above the previous Moditope patent.

I too picked up that it is now a multi stage process.
The solubility has been one (maybe the main) problem. Let's hope that has now been solved.
The other problem of conjugating with Amplivant looks to be sorted now.

To summarize, it looks like the process is:

a) Polypeptide to conjugate each of the 3 peptides with Amplivant - complete
b) AMRI Combining the 3 target elements into an antibody structure - complete I think
c) AMRI Ensuring solubility - ongoing (at time of AGM, may have been completed now)

I, for one, have consistently said "expect delays".
When doing something new, there are always the unexpected problems to overcome.
With Modi1 is was manufacturing difficulties.
With the BionTech collaboration it was HSE issues around creating a viral vector.
With SCIB1, we have the FDA not approving Trigrid 2.
With SCIB2 we don't know what is causing the delays

Of these, the SCIB1 delay is the only one that annoys me. The lack of a fallback plan, when quite early Scancell knew there were problems getting Trigrid 2 approved is what bugs me. This is not a hindsight issue, It should have dawned upon Scancell that they needed a plan B once they knew Trigrid was not approved. Any fallback plan did not involve extra research or development so it would have had minimal effect on timescales of other projects.

With the SCIB1 combo trial currently stalled, one of the following 3 look more likely to be the breakthrough in SCLP's fortunes

a) Start of Modi1 trial - hopefully this will be multi-centre from day 1 and NOT delayed any further
b) The Biontech collaboration. Delayed by HSE issues but presumably back on track
c) The Avidimab collaboration

All of these are being worked on by Scancell.
I have always thought that the Modi1 trial is the most important of all of Scancell's activities; past, present or future.
It is wider in scope than the SCIB1 combo trial and it should be easier to recruit patients. There is no issue about paying for CIs and it has the backing of the CAB. Presumably the CAB will also advise on the best centres for the trial.

Here's a man who has a very extreme opinion of the FDA.

It could just be a case of Ichor having bigger fish to fry. But then again surely they want to get a Trigrid 2 trial underway in the US.
It does make you wonder about conspiracy theories.

I don't think they've missed the boat. I just think they are still waiting for it to leave, with the FDA inspectors still looking at the engine. What on earth can be their objection when the 2 trials in Europe have the go-ahead (1 is ours) and 1 in Korea.
