Based on the extensive recent data on Avacta’s AVA6000 and its pre|CISION® platform, the likelihood of clinical proof of concept (POC) success appears very high, supported by several key points:
• AVA6000, a FAP-activated doxorubicin conjugate, has demonstrated a favorable safety profile with no maximum tolerated dose reached up to 385 mg/m², indicating excellent tolerability compared to conventional doxorubicin.
• In patients with FAP-positive salivary gland cancers (SGC), AVA6000 showed a 91% disease control rate, with multiple confirmed tumor responses (partial and minor), and median progression-free survival (PFS) not yet reached after more than 25 weeks follow-up. This PFS nearly doubles the benchmark PFS of 3.5 months for similar patients on conventional therapy.
• Tumor biopsy analyses confirm that active chemotherapy is released specifically in the tumor microenvironment, validating the targeted delivery mechanism of the pre|CISION® platform.

Given these strong clinical signals and platform validation, a reasonable estimate for the likelihood of clinical proof of concept (POC) success for AVA6000 in FAP-positive tumors, particularly salivary gland cancers, would be in the range of 70-80%. This reflects the robust early-phase efficacy, excellent safety profile, and confirmed tumor-specific drug activation demonstrated so far.
