I haven't screamed about anything as you put it. I have just linked an important science paper and linked various commentry from very senior scientists who are very influential in decision making. I have just shared the research for people to make their own conclusion.

Thanks Scinv commonly the ****tail contains just 2 monoclonal antibodies. Idea being virus becomes resistant to one the treatment will still work. However what happens if the virus becomes resistant to both!

So re-read the twitter thread in context with the recent paper I shared. See that Eli Lilly mabs ****tail now has both its monoclonals made ineffective against the SA variant.

Now effectively calling Jeremy Farrar ignorant because he says we shouldnt be using single monoclonals is a bit silly :)

This new preprint by David Ho’s group shows that 501Y.V2 already "is resistant to a major group of potent mAbs ..., including two authorized for emergency use”.
As @JeremyFarrar
told me: “We shouldn't be using single monoclonals."

Thats going a stage further senior people saying we shouldnt be using them. Jeremy is the Director of the Wellcome Trust.

Yes its a major issue from the science paper you can see there is a change of thinking with mAbs. They are not a good solution for longer term use or stockpiling as they likely become redundant if there are mutations.

Fauci, Horby and other key influencers will be concluding the same.

Another important line here 'Several other mAbs in development are similarly impaired'

I need to research more but this could be bad news for AZN mAbs too. These E484K and other mutations could render them ineffective.

Important new data released I suggest everybody read and the implications for mAbs treatments if Brazil or South Africa (or any E484K) mutation becomes prevelent.

Our findings have relevance to the use of mAb to treat or prevent SARS-CoV-2. Both UK?8 and SA?9 are resistant to neutralization by mAbs directed to the NTD supersite (Figs. 2c, 2d, & Extended Data Fig. 3b). More importantly, SA?9 is resistant to a major group of potent mAbs that target the RBM, including two authorized for emergency use (Fig. 2c). LY-CoV555 is inactive against SA?9, and the activity of REGN10933 is impaired (Fig. 2b) while its combination with REGN10987 remains potent (Fig. 2e). Several other mAbs in development are similarly impaired (Figs. 2c, 2e, & Extended Data Fig. 4b) against this variant. Decisions on the use of these mAbs will depend heavily on the local prevalence of B.1.351 or B.1.1.28, thus highlighting the importance of viral genomic surveillance worldwide and proactive development of next-generation antibody therapeutics, including combinations that target antigenically distinct epitopes.

The 8% efficacy number comes from an unconfirmed german government source according to the reports. Could just be a mischief news story but with how bad the AZN trial was its not one we can dismiss out of hand.

Its what the EMA do on Friday thats important - if they dont approve it for over 65's then its a big blow for AZN. What sensible person in the UK will want an AZN vaccine? You will ask for a phizer vaccine.

Unsourced reports in German newspapers say the AZN vaccine in over 65's has just 8% efficacy.

On Friday EMA will make decision on approval. If they dont approve for over 65's that will be a major disaster for the UK government/MHRA.

Moderna works on the UK variant but has a 6 fold decrease in neutralising titers vs South Africa variant (and likely Brazil variant due to E484K mutation). Also bear in mind in the UK and most EU countries we do not use the Moderna vaccine (yet).

Looks like the E484K mutation found in South Africa and Brazil will have a serious impact on the vaccines effectiveness and companies will need to update/amend their vaccines.

Yes Bojo locked down too late to prevent spread from Brazil / South Africa. We know other countries like Argentina, Tanzania, Italy have seen the E484K mutations to on smaller scale and people have been flying in from these countries.

From UK we know these variants are here and we have detected them early. Its down to our government stopping the spread and pot luck with how the virus evolves.
