Not 350, C7 will be at 385mg/m²

Doses have been:

C1: 80 (90% of the standard 60 doxorubicin dose)

C2: 120 (increase of 40)

C3: 160 (40)

C4: 200 (40)

C5: 250 (50 after MHRA approval to increase)

C6: 310 (60)

C7: 385 (75)

That's a very big jump to C7. Probably their estimate of how much they can increase whilst being confident of still being under MTD. I'll bet they're kicking themselves though that they didn't start off at 100 (equivalent to 67.5mg/m² bound doxorubicin, so 90% of the standard 75 doxorubicin dose rather than the 90% of the 60 dose that they actually started with) and go up in steps of 50 all the way to 350 in C6 and 400 in C7 but hindsight is a wonderful thing and they could only do what MHRA would allow them to do.

The change does seem rather hurried and unplanned. He hasn't posted on LinkedIn for 3 months nor commented much in the last month, both of which would indicate personal reasons rather than a professional 'falling out'.

Governments don't "oil the wheels" of regulatory approval of drugs. Imagine a world where that happened! 'Take Clorox for your covid! It's government approved you know y'all!' goes the TV ad in the US where such direct advertising is allowed. No, it's 'evidence based' and the reputations of scientists, companies and regulatory authorities are at stake. If the MHRA started approving ineffective or downright dangerous drugs for sale in the UK what do you think that would do to confidence in the UK health system? And what would it do to the companies that had those ineffective or dangerous drugs approved? The whole system is based on faith in everything being done correctly and above board.

Do you have source for an alleged interest in a £10m fund raise for Affimers? And why Avacta would need to do an urgent fund raise for such a small amount last month given that they knew the AVA6000 data they were sat on?

Hi JT, I take your point, but you would agree that 209mg/m² is the MAXIMUM amount of doxorubicin that could be cleaved by FAP from 310mg/m² of AVA6000? The PK data will tell us everything that needs to be known about what the equivalence actually is - agreeing that the equivalent amount of straight doxorubicin, were it to be administered, would be fatal!

A nice summary by RAH of key points from the announcement but I would take issue with one thing that was written, namely:

"At 310mg of Dox (sic [AVA6000]), 209mg of AVA6000 (sic [doxorubicin]) equivalent (but targeted and therefore super charged) AVA6000 continues to be received safely."

We cannot assume that 310mg/m² of AVA6000 is equivalent to 209mg/m² of straight doxorubicin, only that 209mg/m² is the MAXIMUM amount of doxorubicin that could be cleaved by FAP since some may be excreted uncleaved just as some straight doxorubicin may be excreted without acting on cells. The PK data from the trials, Phase 1a and now also Phase 2, will provide that information for both AVA6000 and straight doxorubicin. It is certainly absolutely true that AVA6000 targets and concentrates doxorubicin in the TME and will be delivering massive doses there.

How do you know that AVA6000 can be administered via a simple back of the hand cannula? As far as I'm aware, Avacta has never mentioned how AVA6000's intravenous infusion is administered. When/where/who did you ask?

I think we'd all agree that AIM investors are mostly just interested in chasing the next (or last) big thing and that UK institutional investment managers are pants at buying into new tech at an early stage.

The US however is different. Short of listing on NASDAQ, an option is to get tech-, biotech- and pharma-savvy investment managers interested in Avacta. Not sure whether US investors were also targetted for Science Day but Science Day seems to have had little to no effect on institutional investment in Avacta, let alone US institutional investment.
