The main problem DNA vaccines have to overcome is that the vaccine needs to gain access to a cell nucleus

Modi1 is a peptide vaccine not a DNA vaccine. It is the Immunobody vaccines that are DNA.
However , I reckon Prof Ottensmeier is a fan of both Moditope and Immunobody vaccines.
He was, in fact, a clinician on the SCIB1 trial and his name also appears on some of the SCIB1 articles.

Type 4 is concerned with T Cells and is delayed and also particularly associated with CD4+ T Cells.

"Delayed hypersensitivity is maintained by T- lymphocytes. T-cells (lymphocytes) have two main types—the CD4+ cells and CD8+ cells. Type IV hypersensitivity requires CD4+ type. The spe­cial group of CD4+ cells take part in type IV hyper­sensitivity and are called T-D cells (delayed). Again T-helper cell (TH cell) includes T-D cells which constitutes the bulk of CD4+ T-cells. TH cells are again distinguished into TH-1 and TH-2 type, of which TH.2 cells are mainly responsible for acti­vation of B-cell to produce immunoglobulins and TH-1 cells are involved in causing the inflamma­tory responses including delayed hypersensitivity reactions (Fig. 11.6)."

Causing that inflammatory response is the first stage of Modi1's mode of action. The inflammation is essential for Modi1 to then progress to the killing stage. I suspect Lindy is such a fan of these CD4+ cytotoxic TCells since they do the whole job - inducing inflammation and then killing. CD8+ TCells are not required to do the killing.

Bojo, always worth re-listening to one of Lindy's talks when she tries to explain Scancell's science in a such a short space of time. It gets more compressed with each AGM as the science develops.

It's fascinating Wild, and I remember reading that the immune system limits clonal expansion once it thinks that it has enough T Cells of a particular TCR. So there is a natural breaking mechanism to having too many T Cells. I will try to re-find that particular article.
I was thinking more in terms of clonal expansion whilst the tumour is regressing.

Aren't the exhausted T Cells replenished by those resulting from clonal expansion?
But I agree that it is probably better to have steady rather than explosive regression of the tumour

SCIB1 - very successful phase 1/2 trial already carried out for stage 3/4 melanoma with outstanding results in patients

Covidity (Covid19) - Trial finished and results are being written up (uses Avidimab)
SCIB2 waiting in the wings for non small cell lung cancer

Moditope - used to develop vaccines for late stage cancer. The results in humanised mice were exceptional.
Modi1 - trial underway for 4 cancer indications show great initial promise
Modi2 - planned to go into the clinic in 2024

Glymabs - these attack cancer via malformed sugars. Five have been developed but Scancell are looking for more.
First pre-clinical deal done recently with Genmab for 1 Glymab only
3 other collaborators may also be seeking deals

Avidimab - a way of increasing affinity of any mAb
Scancell are currently talking to 2 parties
