"AMG319 is a PI3Kd inhibitor. Preclinically, target inhibition abrogates Treg mediated immunosuppression, augmenting CD8+ T-cell anti-tumour activity. "
The aim was to produce an inhibitor that would give the immune system a better chance of attacking the tumour. So the same basic aim as CIs like Keytruda but with a different target to inhibit.
As you say, crumbs, the trial does not appear to be going well.
Perhaps Scancell knew about this trial and decided to compete but I think inan's comment about the switch was probably more relevant. It is fine tuning as Scancell test Modi1 against more and more cancers in mice. Also perhaps, the work in the BionTech collaboration has some relevance to this.
I always get the impression that Lindy strives to produce vaccines that go after the most difficult cancers to treat. So there also might be an element of this too.
But it would be nice if CRUK wanted to do deals on Moditope trials with a similar framework to the SCIB2 trial.
You do see Nigel Blackburn's quote in you first link:
Dr Nigel Blackburn, CRUK’s director of drug development, said: “We’re delighted that the collaboration between Amgen and our Centre for Drug Development is moving into Phase II trials. It means we’re getting closer to providing a new treatment for cancer patients.
I wonder if he is also thinking:
"It would be better to be involved in Moditope trials"
