IMo mistakes have been made in the past.
But I see no point in dwelling on those mistakes. I believe the trial strategy is correct now. CH's timescales have been wrong, but personally I have no issue with that since I always thought they were over optimistic.

ImO The kudos of having a US trial run by KF took precedence over the urgency of production of trial data. I thought that at the time and have not changed my view.

No preclinical prep during the early stages of the scib1 trial. Too much confidence that a deal would be done on scib1. Again I thought that at the time.

I agree entirely about your remarks about obsolescence.

But I don't agree with your remarks about learning about the science. How can you invest in a scientific company without having some understanding of the science of that company and its competitors? As a BB surely we should learn together by asking questions and raising the general level of knowledge. We'll never become experts but at least we won't be making totally blind bets.

The ozzies have probably got a better post removal attack than us.

But seriously, what it shows is that the science is ever advancing on different fronts. Lindy seems to be adept at starting new projects using her extensive network.
But I agree very much with Bermuda. The latest collaboration is with a serious commercial partner and it is non exclusive.

The extra avidity that Avidimab confers intrigues me. We know that Lindy has added structures to the mab tail allowing the tail to bind to proteins on the tumour surface. This binding then augments the normal antigen binding that is comparatively weak in the case of glycans. But the information we have so far also talks about a 3rd type of binding namely the Mab tails binding to each other. It sounds like this also adds to the overall avidity.
It looks like pure genius to me.
In addition the tail can also be used to carry "warheads". I had to laugh at that when I saw it in the Trinity article.In all 4 different types of killing mechanism are allowed for.
So bring your tired Mabs to the Mabs service centre and we will turbo boost them.

Yes. I think the section on Avidimab in the website and the update from Trinity are both interesting in that they impart different aspects of Avidimab. Maybe written by different researchers within Scancell.
It is certainly much more of a platform than I expected. The improvement seems to have come about from the humanised Mab losing the killing action. So Lindy did modifications to the tail (fc region), this is the bit that I believe to be unique.

For those not familiar with the science, these are modi2 targets that inan is talking about. So it looks like another range of targets and another range of cancers.
Quite a lot for Lindy to talk about at the agm.

I notice in the pipeline page that Modi1 has now got renal cancer as a possible target. I wonder if this is the CAB at work.

As I understand it, the modifications to the Fc region (the tail) of the Mab allow either drugs to be delivered to the tumour or mediation with TCells close to the tumour to kill the tumour. As such, this is a breakthrough that is relevant to any Mab. It also boosts the avidity of the Mab.

So, for instance, take a CI Mab that blocks off a certain protein expressed on the surface of the tumour. This same Mab could also now be modified to provide a killing action. So it gives this Mab both blocking and killing potential.

I would have thought this will attract a great deal of interest. Is there anything like it already out there?

All the above is in addition to the 4 Glycans Mabs that Lindy's team have already developed.

The Fc region of the antibody is the tail. So Lindy has found a modification of this region of the antibody that enables the tail to bind more strongly to receptors on the tumour. The tail is NOT the part of the antibody that binds to the antigen. So this is the reason that this Fc modification can be applied to any Mab! So potentially increased avidity for any Mab with this modification. It also confers direct killing ability with certain targets!

So 4 methods of killing and can theoretically be applied to any antibody.
So, if an antibody disappoints in either its low level of avidity or its killing potential then AvidiMab could be the answer.
