The way I see it is that 2 checkpoint inhibitors are likely to be better than 1, if they are blocking off 2 different checkpoints. Its like having a centre half and a goalie. But maybe the law of diminishing returns applies here. So perhaps adding a 3rd CI may may little additional affect.
So, attackers like SCIB1 are maybe what is needed to raise the bar even higher.
Remember that SCIB1 mounts its own powerful attack. In contrast CIs rely on the patient's own immune system to mount an attack and if that patient's immune system is weak then it is unlikely to halt or reverse the cancer.
So the question we should be asking is
"How much better is a SCIB1 attack compared to the immune system without stimulation?"
Well we have a good indication of the answer to that question already from the Scib1 mono trial.
But I agree we could do with more than 25 patients to statistically put the result beyond doubt.
