Hi Blastoid - investors using chat forums have to learn very quickly that for every dozen sincere posters there will be at least one contrarian claiming the end is nigh no matter what the situation on the ground is. Yes, our compounds could still fail but at the moment 737 has completed two Phase 2 trials and 1801 is progressing in Phase 1, so it's hardly time to turn off the lights in Pampisford just yet.

As we progress further the naysayers will need to be more creative. CoL tried to suggest we were stuffed due to the potential for nitrosamine contamination during the manufacturing process. I read up on it and ascorbic acid (vitamin C) is commonly used during manufacture of drugs to prevent the issues raised. Just to confirm things, I asked a related question at the InvestorMeets event and John said production met all GMP guidelines. Scaremongering nil, DYOR 1.

So if MAJ (or anyone else) thinks the BoD are determined to crash Sareum onto the rocks then, yeah, sure, whatever. Simply ask for evidence to back up their claim and await a response. Or just reply "up your butt with a coconut."

There's an issue with the pipeline page - the progress bars are way off to the right and the text formatting looks a bit shonky overall (cue some wag saying it's just a reflection of an incompetent BoD (yawn)).

I've contacted the site developers so we'll see if they correct it. I contacted them before when the new-look site went up and they corrected errors quickly (though they never thanked me for pointing out the issues).

Evening MAJ - so we can expect a flood of shares to enter the market next Tuesday? Thanks for the heads-up. I wonder what will happen to these cheap shares if (big if) we get an RNS in a week or two to say the safety review committee has looked at the initial Pt1 results and given the go-ahead for Pt2 to commence.

That would be a major derisk and 'excess' shares on the market would be hoovered up pretty quickly. But you probably know that.

Amor - they have committed to £2.6m funding which can be used to get us through Q1 2024. If certain conditions are met, we could then access more funding to take us to the end of next year (plus we'll have about £1.6m tax credit due to us).

Exactly, RMM. Even if a deal was in the offing for 737/1801, you still need to carry out business as if the potential deal didn't exist. This is the BoD ensuring they have a financial safety net ahead of when they really need it.

Hi Blue - I think they may have taken this route rather than an open placing to us punters as this way they have the option to draw down money only when needed. If a 737 deal comes along then they won't need the loan hence minimising dilution.

Curious how some posters see this as doom and gloom yet it isn't too dissimilar to HNWs buying in. They don't do it to spaff their money away, instead they see great potential.

Hi Laz - I'm guessing the third participant should have their follow-up visit at the end of this week. If so, we might hear something towards the end of next week. From the trial RNS - "Part 2 (multiple ascending dose) is planned to commence after the Safety Review Committee (SRC) has evaluated the available data from at least Cohorts A, B, and C in Part 1."

My timings are based on 6 nights in hospital then a follow-up two weeks later and if all good, the next cohort starts. So roughly 9wks from 5th June until there's the minimum amount of data required by the SRC to green-light Part 2.

Depending on how it's all going, the SRC may require more data before giving Part 2 the OK, therefore effectively pushing any news back by about 3wks per cohort. So perhaps next week is the absolute earliest for an update.

At least the various Sierra combo patents should play into our hands thanks to all IP being handed over to CPF.

Evening Basser - I don't wish to be mean but you can be silent all you want. I thought my speculation was reasonable based on a number of known factors. And for the record, I am patient.

Time for the F1 qualifying highlights......

1. First patients dosed on 5th June (per the trial doc) and will be in hospital for a week.

2. Follow-up visit and end of involvement about 2wks later.

3. Next cohort could be screened during this time so that they can be dosed straight after previous cohort ends (not sure if the next cohort could be dosed after the previous one leaves hospital or if they have to wait until after previous cohort have had their follow-up visit).

The RNS notes that Part 2 (the multiple ascending dose cohorts) could start after the first 3 single dose cohorts have been reviewed.

So if a new cohort has to wait until the previous one has had its follow-up visit, it's about 9wks from first cohort dose (5th June) until third cohort are finished (roughly 7th August). This is possibly the earliest that the Safety Review Committee would have enough data to look at and give the green light for the multiple ascending dose element to start and run parallel with the remainder of the Part 1 cohorts.

If I'm in the ballpark then the sp could remain bumpy until mid-August (unless there's some 737 announcement). However, if the SRC give the green light for the multiple ascending dose cohorts to begin then surely the sp will finally respond to the excellent progress.

Hi Desamax - tricky to do much of a comparison as Deucravacitinib (BMS-986165) is TYK2 whereas 1801 is TYK2/JAK1 therefore a bit apples and oranges.

Key inclusion criteria

4. Has moderate to severe plaque psoriasis at baseline as defined by a PASI score greater than or equal to 10 and a PGA score greater than or equal to 3 at Screening and Day -1.

So whilst the PASI score doesn't appear to be part of the measured outcomes (as we're looking for tox issues), I'd be surprised if they weren't measure as part of the assessment. A bit like taking your car in for new tyres but the garage doesn't bother checking the pressures once they're on - you only wanted the tyres fitted!

They are shown a two sells because both the sale and buyback are transacted below the mid-price in order to minimise any loss. You'd be well miffed if the transaction sold at lowest price then bought back at the highest!

Hi Potnak - whilst the aim of any P1 trial is soley to establish safety parameters, the fact that volunteers with psoriasis will be included helps us immensely as blood & urine results can only inform part of the story - if a patient says their itchiness has reduced etc. then we know we're onto a winner.
