I’m curious if anyone knows the logic behind choosing the split of 1-1 Drug vs Placebo

I’ve seen other trials choose 2-1

I wonder what effect if any this has on the way data is reviewed and weighted.

We wouldn’t want to be on ACCORD currently anyway if we had been put on ACCORD we would have lost 8 weeks of time. Much better to finish our trial get the result we are hoping for.

Then you could envisage starting a much larger phase 3 study under an ACCORD umbrella or similar.

I’m wondering if they have managed to fill the final 11 patients on the COPD trial.

They didn’t explicitly rule out recruiting to the trial if one of the patient pool contracted Covid.

It would be a nice surprise to get two lots of data in June.

Maybe if the Covid trial is promising they’d unblind the COPD trial anyway.

All speculation.

Something I’ve never seen posted here is the acknowledgment that for the company to log an RNS costs money.

I think it’s in the hundreds of pounds region.

So you don’t need many RNS’s and you could have paid for a dose of treatment, I’m sure that weighs heavy on a scientists mind.

Nolupus - Just trying to broaden my understanding.

Perhaps I should wait until the Roche and Sonofi results until I try and draw too many conclusions.

I’m conscious that after C19 my portfolio of shares should probably have a larger bias towards areas that I would usually not invest in.

As you say TILS has more to it than C19 so whilst I might currently think there are better ops for C19, TILS is definitely on my radar beyond this.

Thanks to those who did form a response to my question there was no motive other than to try and better educate myself by seeking out opinions that don’t necessarily agree with my own. You can get blinkered if you only communicate in tight circles with the same agendas.

Woodaldo - Does the Novacyt portable PCR have an expected release date on it? Sounds very much like the GDR system which the US military use.

Seems like NCYT are drifting in the direction of GDR tech.

Anyway you guys had a rotten end to the week this week I’m sure this will spring back in due course probably oversold currently as traders jump out.

TamerInvestor - I agree with that sentiment.

I guess my question was getting at the fact that the half life of anti-il-6 in vitro is sufficient that it gets to the lungs with a reasonable dosage. So I was wondering what benefit of direct to lung would be in this case.

If you had a sufficiently short half life of the molecule then direct to lung would significantly reduce the required dosage which is an obvious benefit, but I don’t think that’s true here.

So I’m trying to work out the other benefits of the direct to lung technology.

I can see nebulisers are friendlier to the patient than IV treatments and can be used as an outpatient.

Would welcome any points I’ve missed.

You’d have to track 100 patients outside of the trial that had the required entry requirements to get on the SNG trial.

Then you could probably make the links you are suggesting it would get more accurate the larger the study size.

Hi all I am not a holder here (yet) I was wondering if some of the more researched holders here could advise why they think nebulising anti-IL-6 drugs is a better approach than the conventional anti-IL-6 drugs which are given via IV or SC?

Nothing sinister in the question just trying to understand the view here.

SNG001 could prove to be a first line of defence against a multitude of viruses that effect the lungs.

It’s use could be very broad and common place which would be fantastic.

Primary objective here is to get the required data, phase 2 phase 3, and where possible emergency use authority.

The above would normally take years, we could see real value here quite quickly as the story unfolds.

There will be no one drug wonder for Covid.

However there has been a lot of good data coming out surrounding interferons so I am confident the trial will give us the result we need.

Scoutt - I agree with you.

ShareStings link proves that they are likely to have access to blinded data.

The purpose of this is believe is to be sure that patients are actually developing adverse symptoms from infection, pointless for keep running the trial if no one is getting ill.

What the blinded data does not tell you by its very nature is who the data belongs too.

So to draw conclusions is not possible. You could do analysis on the data and lump it into two distinct groups.

But once unblinded it’s likely both groups you have formed will contain both medicated and placebo patients. There will always be outliers and with only 50 patients on the drug it won’t take many to effect %’s.

We just need the last patient to receive their dose I bet the company are itching to unblind their data!

PeterJW - It hasn’t been discussed in great detail and is obviously a sensitive subject.

Pros for Synairgen
- We have self funded operations to date no government money or grants.
- We are a small organisation so the fact we are contributing a potential therapy is significant in itself.
- COPD was our main objective so we cannot be accused of taking advantage.

To distribute globally Synairgen will need partners, those partners might have the balance sheet to be able make a generous gesture which IMO would benefit the future sales of the drug.

The Gilead gesture was a drop in the ocean for them but Remdesivir could be set to become a standard of care drug in the US which would be a huge result for them off the back of some lack lustre trials.

The phrase "preclinical experiments" implies they have not yet begun clinical trials of any form so they have a huge body of work ahead to prove safety before beginning to think about efficacy. Probably talking in years not months.

There is no coincidence that the Synairgen trial is being used as a model for future trials.

Whilst others may be publishing results ahead of us, take note that since the trials have not been conducted as double blind placebo trials the data whilst promising is only indicative and requires further trials to be conclusive.

We are likely to be the first to deliver data that will stand up to scrutiny globally.

Synairgen are certainly making all the right calls currently.

I'm not experienced enough with Level 2 to really use it to any great advantage plus on AIM you get so many late reported trades etc that you are still half blind.

It's main uses for me are:
- Live price and spread so I can try and place clever limit buys/sells that might get filled during auctions
- Live trades so that you can pick out who is telling porkies on the chat boards and you can gauge a little bit the momentum of a share.

Unfortunately there is no substitute for Research, Patience and a Bit of Luck.
