If AVA6000 crosses the placental barrier, I doubt treating pregnant women would work as the foetus will be growing, cells will be dividing and thus will be high in FAP and so would cleave AVA6000. Could have a role as an abortifacient in that case.

If AVA6000 doesn't cross the placental barrier, then no problem, but I think that would be unlikely.

If AVA028 was originally PD-L1 Affimer® / IL2 bispecific (no detailed description on the Pipeline page), the research code seems to have been reassigned to AVA028 – PD-L1 Affimer® / ImmunoCytokines (with detailed description on the Pipeline page that doesn't mention IL2, only a number of potential ImmunoCytokines). I don't know why they would do that.

It is still a safety and tolerability study. It is also a range-finding study to identify susceptible cancer types. The cohorts have consisted of end of life cancer patients, who have already revceived one or more lines of treatment. You don't know what benefits the patients on the trial have gained beyond the 65% tumour reduction because you are not invested and YOU HAVE NOT BOTHERED TO FOLLOW THE 12th December PRESENTATION, where some benefits to STS patients are described but not all benefits to all patients.

So AVA004 is the basic PD-L1 Checkpoint Inhibitor, the AVA02x compounds are bispecific compounds but AVA028 is the group name for various multispecifics of Affimer plus (>0) immunocytokine. AVA032 comes under this, or they just haven't unpdated the website from this:

AVA032 & AVA32

You have to understand that Avacta have chosen a really weird way to number their research compounds, AVA6000 & AVA3996 included. It seems that AVA028 has not been assigned to one compound but to a class of compounds (Affimer-cytokine), of which AV032 (sometimes written AV32) is just one. If I didn't know Avacta better I'd say this was designed to confuse.

Well it's been two years of abysmal performance for Avacta's SP but, like many things in life, investment goes in cycles and the best time to invest is at the bottom of a cycle, not at the top.

The US had it's customary Santa Claus rally (coinciding with the end of their tax year? - equivalent to our mad dash to get cash in by 5 April) and subsequent early January fall-back. So hopefully we all kick on from here.

I think the complete data for Arm 1 C1 to C7 will be presented at AACR in April. Just my opinion.

New intellectual could be revealed at the same time. I'm sure it will be protected by then (patent applied for). It's just a matter of whether they want to reveal it if it hasn't been published (put into the public domain) by then.

Q3 was one of mine (Q2-Q6. plus Q37 during the meeting). I don't think "protecting new intellectual property" could refer to a pharmacologically active compound in any way as that could not be introduced in a first-in-human Phase 1 trial of another potential drug without extensive IND enabling study and approval. The timescale is too short.. I suspect it will be a method or process patent and my money's on a diagnostic method to identify FAP levels. There are several clues as to why that might be. 2+2+2+2...

If not obvious from my 3rd paragraph, I think there could be multiple licence deals for the pancreatic, colorectal, and head & neck cancers to be included in Arm 2 (see the 13 December deck slide 11) with the licencees paying the costs for Phase 2 and, if necessary, Phase 3 studies.

I also have a sneaking suspicion that Avacta are able to identify high FAP cancers.😉 Or is just that these Arm 2 cancers are fast growing and hence fast cell dividing and hence susceptiple to doxorubicin?🤷🏻‍

Avacta are not presenting at the JP Morgan Healthcare conference because, I think, it is too early. I would expect Avacta to present Phase 1 data at AACR in April and to follow up with a peer-reviewed article. If they are not even going to the JPM event, then they must be very confident about whatever negotiations are currently taking place. However, I suspect that they will be attending, to network if nothing else.

The move to 2-weekly dosing is common sense in the quest to find the optimal dosing regimen. Pfizer have a one/third strength weekly dosing doxorubicin which is claimed to be more efficacious and have fewer side effects than the standard dosing. The idea to do this could have come from Avacta themselves (it was mentioned at Science Day and I was in contact with Andrew Saunders about it afterwards), a suggestion from the FDA or indeed may be a potential licensee's requirement.

A much more likely potential licensee's requirement would be the additional indications in Arm 2, although that could also have come from within Avacta or from the FDA as it makes sense to go after ALL high FAP cancers as low hanging fruit in this pre-registrational phase. This was one of the most satisfying pieces of information for me from the 13 December update as, no matter where the idea came from, it will make it much easier to take AVA6000 forward in indications other than the rather restrictive - in terms of both available patients and market size - STS subtypes they will be selecting.
