IFN-I signaling, determined by the physical-chemical properties of interactions between IFNAR and various IFN-I species, serves as a gateway to activation of selected genes generating different biologic phenotypes. The dichotomy uncovered between a and ß IFN responses during various pathogenic infections suggest there are unique molecular and signaling pathways controlled by IFN-I that await discovery. Clearly, further careful mapping of a and ß activity and receptor binding coupled with development of decoys and therapeutic molecules is likely to open new avenues for successfully understanding pathway usage and mechanism by uncovering specific activation signals, roles and activities for various IFN-I subspecies. Correlation of screening studies that illuminate the activities of interferon stimulated genes on pathogens (Liu et al., 2012; Schoggins et al., 2014; Schoggins et al., 2011) with study of the types and timing of the IFN-I species and the genes they activate during infection will lead to a better understanding of the IFN-I mediated chess game between the pathogen and host and generate strategies to manipulate the IFN-I pathway for biomedical benefit.
From my non-scientific but well read pov:
Interferons, naturally occurring human innate antiviral proteins, are powerful and complex and we are only just beginning to understand them some 50 years after they were discovered. If we can harness their power safely, we can really smash the s**t out of viruses.
This is why SNG is so bad*ss. For almost 20 years, they have been painstakingly researching interferons and their application, learning that the top trump was INFb and by bypassing the bodies full system via direct inhalation to the lungs, they can apply just enough INFb to blow any virus out of the water. This overcomes the negative side effects of injected interferon, which takes its toll on the body (this is why you'll see papers arguing against it as an injection)
It's a genuine breakthrough discovery tbh. So exciting to be a part of :)
