The study featured in the link has made the headlines because of the cutting edge genetic engineering technology used and quite rightly so. So the news is more about the way in which the therapy has been created rather than the results it has produced.

However, no matter how good the process, the success of any treatment still depends on selecting the right targets. Personalised therapies such as this which target neoantigens (new antigens) created as a result of genetic mutations in a patient's tumour use algos to select the best neoantigens. Only time and much larger studies will tell how good those algos are.

As you know, Moditope also targets neoantigens, but because they're created by a post translational modification rather than genetic mutation, they don't vary from patient to patient . We also know that Scancell are working on developing Moditope TCRs. Let's hope that both therapies are hugely successful.

SITC is a prestigious scientific conference and good to see Scancell presenting there. Worth noting that in order to be accepted for presentation, the poster must contain new, previously unpublished research.

I'm sure these conferences are great for networking, making contacts and highlighting R&D achievements, all of which may ultimately lead to a deal, but they're not a place where deals will be signed. It takes months and sometimes years to negotiate a deal, complete due diligence and all legal work etc. . So in answer to your question - no I don't think SITC22 itself will lead to an announcement of a deal, but who knows what seeds may have been sown at other conferences in previous years? We just don't know.

By the way, I'm not even sure LD is attending SITC 22 - I suspect she's not.

As posted previously Scancell are presenting a poster at the SITC annual meeting which starts today in Boston. The full abstracts have now been published and link to Scancell's is below. As we can see, Scancell have discovered another potential target for the Moditope platform, in this case the citrullinated GRP78 protein. A couple of quotes:-

'We have shown in animal models that the efficient stimulation of citrullinated epitope specific CD4 T cells results in dramatic elimination or regression of tumours'

'his data leads us to propose that citrullinated GRP78 is a candidate tumour antigen and that vaccination against citrullinated GRP78 may provide a promising approach for future tumour therapy'

Yes the trial has been designed to gather data from a wide variety of settings to inform the design of the next stage of development, whether that be a phase IIb or III trial.

Just a couple of points. As I understand it, ovarian and triple negative breast cancer arms will only be as a monotherapy and not in combination with a checkpoint inhibitor. This is because in ovarian cancer no CI's have been approved and in TNBC they've only been approved in combination with chemo but Modi1 can't be given with chemo.

Also would need to check again, but am pretty sure the neoadjuvant arms are only being run in head and neck cancer patients and there'll be a monotherapy and a checkpoint inhibitor combination arm. Perhaps someone can confirm this?

In other words, it's not all 4 cancer types in all 3 settings. Hope this makes sense.

hyms - they're for the current trial. At the moment, only 14 are listed on clinicaltrials.gov. I assume they will add further study centres if and when required.

They say a picture speaks a thousand words. This picture shows the beautiful Charlotte and was taken 3 years before she died at aged 19. She had a brain tumour on her brain stem which meant tissue biopsy wasn't possible. Liquid biopsy would be an option for all those patients who are unable to have a tissue biopsy because their tumours are inaccessible.

Thanks for the link, excellent news for TNBC sufferers..

Worth noting that both Keytruda and Roche's PD-L1 checkpoint inhibitor Tecentriq in combination with chemo have been available on the NHS for some time for patients with advanced inoperable TNBC (same setting as Modi1 trial). This new approval means that patients can now receive Keytruda at a much earlier stage. It will be available in combo with chemo in the neoadjuvant setting (before surgery after initial diagnosis) and as a monotherapy in the adjuvant setting (after surgery).

As I say, great news for patients but no impact on the Modi1 trial.

Do whatever you need to help you understand.

Worth having another read of miavoc's 30th Oct post:-

Datar’s approach is service based and they do not have a product that could be offered to hospitals. Datar is a customer rather than a competitor.”

If you have concerns/questions regarding Datar and see them as a competitor to Angle, I'd really recommend that you email Andrew Holder at Angle - in the past I have found him to be very responsive and able to simplify the complexities of different liquid biopsies and how they sit in the marketplace.

I believe that all of his share options have vested and therefore assume/hope that he is allowed to keep them. The bulk of them expire next year, but the largest tranche (3.5m) has an exercise price of 33.2p. Worth noting that Richard Goodfellow also holds just over 6.3m joint ownership shares.

Thanks for your posts today. I assume that Scancell will have carried out some research to test SNAPvax before signing on the dotted line and hopefully that will be published at some stage. It will be interesting to hear LD's comments on whether or not SNAPvax is also a potential option for Modi1 in the future or whether each Modi is likely to require a different adjuvant.
