Physiomics Demonstrates Hantavirus Translational Modelling Capability

Physiomics plc (AIM: PYC), a leading mathematical modelling, data science and biostatistics company supporting the development of new therapeutics and personalised medicine solutions, is pleased to highlight the development of a hantavirus translational modelling framework demonstrating the Company's quantitative pharmacology and mechanistic modelling capabilities within infectious disease applications.

The framework illustrates how Physiomics can integrate multiple evidence layers into a coherent translational workflow, linking:

· in vitro antiviral screening;

· concentration-response potency modelling;

· pharmacokinetic ("PK") exposure translation;

· mechanistic PK/PD viral dynamics; and

· virtual population simulations.

The modelling workflow has been designed to demonstrate how quantitative systems pharmacology approaches may be used to assess whether antiviral activity observed in laboratory systems could plausibly translate into meaningful viral suppression under simulated human exposure conditions.Illustrative workflow components include:

· simulated 384-well phenotypic antiviral screening;

· EC50 and Emax concentration-response modelling;

· human PK exposure simulations against potency-linked targets;

· mechanistic viral-load dynamics modelling across differing antiviral modalities; and

· virtual population and time-to-clearance simulations.

The Company believes the framework demonstrates the breadth and flexibility of Physiomics' modelling capabilities and highlights the potential role of mechanistic translational modelling in supporting infectious disease therapeutic development programmes.

The material has been prepared for scientific and illustrative purposes only and does not constitute evidence of clinical efficacy for any specific therapeutic compound, nor has there yet been any commercial application of the hantavirus modelling framework.

Investors can view the modelling workflow here on the Company's website at https://www.physiomics.co.uk/hanta/

Mike Whitlow, Executive Director of Physiomics plc, commented: "Recent global concern surrounding hantavirus outbreaks highlights the continuing need for more sophisticated translational modelling tools capable of supporting infectious disease therapeutic development.

"This framework demonstrates how Physiomics can integrate assay data, pharmacokinetics, viral dynamics and virtual population simulations into a coherent scientific workflow capable of supporting exposure-response assessment and programme prioritisation.

"Beyond hantavirus, we believe similar modelling approaches may have broader applicability across other infectious diseases, including Ebola and related viral threats. The Company is continuing to explore additional infectious disease modelling frameworks and will provide further updates as appropriate."Dr Ghaith Aljayyoussi, Senior Pharmacologist at Physiomics plc, commented: "During the COVID-19 pandemic, translational pharmacology and modelling approaches became increasingly important in helping researchers and developers understand antiviral exposure-response relationships and optimise therapeutic strategies under rapidly evolving conditions.

"It is exciting to see similar scientific frameworks now being applied to hantavirus and other emerging infectious diseases. By combining mechanistic PK/PD modelling, viral dynamics and virtual population simulations, we believe these approaches can provide valuable insights that support more informed and efficient therapeutic development decisions.

"Infectious disease modelling remains an area of significant scientific interest for Physiomics, and we look forward to expanding these capabilities further."

The Company notes that further experimental, preclinical and clinical data would be required before any conclusions regarding therapeutic efficacy could be drawn.

Dr Shaun Pennington, who helped lead SARS-CoV-2 therapeutic screening activities at the Liverpool School of Tropical Medicine during the COVID-19 pandemic, commented: "One of the key lessons from COVID-19 was that speed matters, but so does biological and pharmacological context. Screening compounds for antiviral activity is only part of the picture. The most useful approaches are those that combine preclinical efficacy data with what is already known about human pharmacokinetics, exposure, safety and dosing feasibility. For emerging threats such as hantaviruses, this type of integrated analysis could help identify repurposed candidates that are not only active in experimental systems, but also have a plausible route towards clinical impact."
