Physiomics Demonstrates Hantavirus Translational Modelling Capability

Physiomics plc (AIM: PYC), a leading mathematical modelling, data science and biostatistics company supporting the development of new therapeutics and personalised medicine solutions, is pleased to highlight the development of a hantavirus translational modelling framework demonstrating the Company's quantitative pharmacology and mechanistic modelling capabilities within infectious disease applications.

The framework illustrates how Physiomics can integrate multiple evidence layers into a coherent translational workflow, linking:

Β· in vitro antiviral screening;

Β· concentration-response potency modelling;

Β· pharmacokinetic ("PK") exposure translation;

Β· mechanistic PK/PD viral dynamics; and

Β· virtual population simulations.

The modelling workflow has been designed to demonstrate how quantitative systems pharmacology approaches may be used to assess whether antiviral activity observed in laboratory systems could plausibly translate into meaningful viral suppression under simulated human exposure conditions.Illustrative workflow components include:

Β· simulated 384-well phenotypic antiviral screening;

Β· EC50 and Emax concentration-response modelling;

Β· human PK exposure simulations against potency-linked targets;

Β· mechanistic viral-load dynamics modelling across differing antiviral modalities; and

Β· virtual population and time-to-clearance simulations.

The Company believes the framework demonstrates the breadth and flexibility of Physiomics' modelling capabilities and highlights the potential role of mechanistic translational modelling in supporting infectious disease therapeutic development programmes.

Dr Shaun Pennington, who helped lead SARS-CoV-2 therapeutic screening activities at the Liverpool School of Tropical Medicine during the COVID-19 pandemic, commented: "One of the key lessons from COVID-19 was that speed matters, but so does biological and pharmacological context. Screening compounds for antiviral activity is only part of the picture. The most useful approaches are those that combine preclinical efficacy data with what is already known about human pharmacokinetics, exposure, safety and dosing feasibility. For emerging threats such as hantaviruses, this type of integrated analysis could help identify repurposed candidates that are not only active in experimental systems, but also have a plausible route towards clinical impact."
