Watch this video AHS and if you understand the mechanism of pre|CISION along with the statement it works you will understand the potential of the platform. Interesting article from Leeds University ; Enzyme Switch Sensors, can detect the amount of therapeutic monoclonal antibodies (TmAb) that are present in a patients system. The sensor is composed of three different proteins - detector, switch and reporter. As can be expected the detector proteins are Affimers which bind to the therapeutic antibodies. Result a Diagnostic test to measure the amount of antibody in a patients system thus informing clinicians how a patient is responding to treatment in 30 mins. Procedure takes place at patients bedside.

One more example imo why Affimers will challenge mAbs in the Diagnostic market dominated by a single reagent.

JT, " Affimers are unrelated to pre|CISION ,two separate platforms". Unrelated in AVA 6000 however TMAC combines the companies two platforms, Affimer immunotherapies and pre|CISION. TMAC to my knowledge is covered by the patent. The plan Avct has employed is to validate the two platforms separately before entering clinical trials with TMAC. Not having this plan covered by the patent makes no sense. That is my interpretation of the facts presented.

The pre|CISION platform patent covers Affimers and a wide range of other binders against oncology plus viral / Inflammatory targets. It also covers a wide range of drugs to which binders can be conjugated. The potential is priceless in the oncology market alone. The patent description turns pre|CISION into three separate potential therapy outlets, put a price on that.

Doxorubicin was the first choice cytotoxin because it was the standard first-line therapy for STS and the fastest route to the market. In no way would it have been the first choice if pre|CISION had been a clinically validated platform.

Listen to AS ( 46 min in ) when asked about the way to get maximum value for shareholders using the pre|CISION platform. It is quite clear the use of the most potent & effective cytotoxins is the way ahead. Avct is in no position to take the platform forward at pace, that is why BP imo will license out the platform. This is the next step forward imo too maximise the potential of the platform. Dox may be most cytotoxic drug in chemotherapy however there are far more potent cytotoxins out there. Somebody has to be interested in finding out which is the most potent & effective. Dox is getting pre|CISION out of the blocks, the sprint to the line is underway and there are at least eight lanes available in the race.

Coris / Avct has this covered with a Diagnostic test available to the market place however as with all approved tests Antibody reagents dominate the market, only one other company can provide a proven alternative reagent. Focus on AMR a really good move imo.

Trendz, up until a several months ago the ADC technology ruled the roost in targeting a cytotoxin. This bombshell statement "pre|CISION works" has turned a multi-billion $ market into a second rate delivery technology. The platform will deliver licensing revenue and imo negotiations with BP are already underway. The alternative is BP stick their heads in the sand and ignore a platform that works.

The pre|CISION platform is where the revenue will be generated, way beyond any individual drug like Doxorubicin. In short take the ADC cytotoxin MMAE, so toxic it cannot be used in chemotherapy, it can now if the BP get involved.

Doxorubicin is not the choice cytotoxin of the ADC market, ADC toxins target crucial intercellular pathways to induce cell death and can be over 1000 fold more potent than chemo therapeutic agents. Statements anybody can find as a fact on Google.

It would therefore be safe to say if pre|CISION works Dox will not be the choice cytotoxin for any of the ADC producers. What pre|CISION does for Dox can be achieved with much more toxic drugs. This imo is why BP will be interested in the platform and waiting to find out if AVA 6000 is clinically proven makes no sense, its the proven platform not the drug BP will be acting on. ADC's we are informed in some cases demonstrate sufficient efficacy and safety to warrant FDA approval. The clinical use of ALL ADC's leads to substantial toxicity in treated patents and many ADC's have failed during clinical development.

J&J $100 million licensing deal is based on yet another mAb doing what no other mAb has achieved in the last 20 + years of ADC development. Toxicity will be major problem.

Faced with the information available regarding the inefficiency of the ADC targeting technology and AS stating "We can use the pre|CISION platform in the linker in Drug Conjugates" why would BP not act on this information. AVA 6000 will carry on through the various clinical trials and discussion will have been underway for many months to secure a licensing deal imo.

J&J dip their toe into the ADC market with a upfront payment of $100 licensing deal. Further payment of a $ 200 million option exercise milestone -based payment and tiered royalties.

Would it be safe to assume J&J is aware pre|CISION works and a mere $100 million + punt on a licensing deal with Avct is not a bad investment. Again seems a no brainer.

Do not list in the top 10 ADC producers and without doubt this is where the all the BP are pumping their cash / hopes. Unfortunately all the approved ADC's searching for the magic bullet drug delivery technology are using the wrong ammunition. Firing shot gun pellets has proved a bit of a hit and miss strategy. The target does not lie.

The bullet is now delivered by pre|CISION and on inspection of the target it hits the bullseye. J&J like all the other ADC producers have a simple decision to make imo, face reality and pick their own choice of cytotoxin. License out Pre|CISION start again with a proven drug and combine it with a proven delivery technology. It's a no brainer, AVA 6000 proving pre|CISION works opens the door to much more toxic drugs than Doxorubicin. Who knows what these can deliver with a better quality of life for the patients.

This class of drug conjugate delivers the potent drug in the TME unlike ADC'S, they require cellular internalisation of the conjugate.

As said pre|CISION works, it does what it is designed to do, deliver drugs efficiently. The ADC market is massive and the only way forward for the BP is to license out pre|CISION and find out which toxin works best for them.

Although ADC's have demonstrated sufficient efficacy to warrant FDA approval the clinical use of all ADC's leads to substantial toxicity in treated patients.

So far research efforts have not come up with a solution to off-site toxicity, pre|CISION works and therefore my understanding of the situation is, companies can license out the platform using their own choice of cytotoxin. Potentially pre|CISION renders the Antibody in ADC'S redundant and therefore a direct replacement therapy imo.

One for the Affimer platform, Avct has already produced Affimers which inhibit the virus from entering human cells. A potential therapy treatment once the Affimer is proven in humans.

A click- to - release technology, unfortunately it's an ADC with a big question mark over targeting efficiency. Interestingly the cytotoxin used is MMAE which would probably be the choice of Novartis if they wanted to take out a license for the pre|CISION platform. With the majority of approved ADC's using this type of toxin it would be an ideal opportunity for Novartis to be a player, much sooner than the Tagworks platform would deliver.

Conclusion : ADC drugs have undergone 3 generations of changes so far and there have been breakthroughs in Antibodies, drug carriers, connectors etc, especially in the use of payloads. Although the Specificity and cytotoxicity of the new generation of payloads have shown significantly improved characteristics than those in the early stages, the current payloads still have limitations.

The pre|CISION door is open, who will step through and seize the opportunity, Pfizer, Roche, GSK, AbbVie, Sanofi, Mersana, Gilead, maybe Daiichi.

Pre|CISION works, Avct has a platform that works and therefore open to many many more applications. This is a point AS keeps ramming home, unfortunately it falls yet again on fallow ground. Happy days ahead if you are not fitted with blinkers.
