NEWS: In a new study, scientists have found that small molecules that stop cancer cells from copying their DNA can boost the effectiveness of another type of cancer drug called CHK1 inhibitors.

This could offer a new, effective way of treating cancer by combining two drugs.

I do find it very odd that on this type of news that CHK1 ceased development at the same time as Nick Glover resigned.

I have tried very hard to find but to no avail. Closest l can get is to MEI as connected to Nick Glover an ardent fan of 737 and the CHK1 inhibitor. Plenty of research work collaboration with M Garrett too!

Of course there are those that shout that MEI are a public company so could not possibly be them.

The on licensee details ae clearly protected.

Nick Glover resigned 2020 and much here depends on the terms of resignation.

What l find most intriguing was the removal of SRA737 from the shop window early 2020. Cessation of development.

With our SDC 1801 it needs to be established thst there is such a degree of safety that it is not affected by black box warnings.

This would then place it head and shoulders above Deucravacitinib on Psoriasis alone.

Ropsacitinib is a TYK2 Jak2 inhibitor and the owners believe they can out perform Deucravacitinib and personally l see them as the most competitive competitor in this field.

As it stands you are looking at evaluating the worth of a compound derived from.its potential commercialisation value.

Psoroasis is nowhere near as big as SLE forecast of world market size of over 300 billion in a few years doubling the current 180 billion per annum.

Ideally looking at initial commercialisation into Psoriasis then other Indications. There are some big markets and at the same time strong competition.

Myself would be looking at modest upfront payment with high end milestone and Royalties.

This is all down to negotiation and the cards that Sareum hold in their hand.

50 plus million up front with milestone payments 1 billion plus l see as ball park.

It still leaves SDC1802 of which the resuls of 1801 will have great knock on effect.

It could all go tits up but l very much doubt that.

We have 2 wholly owned compounds in development ie SDC1801 and 1802.

Both are Tyk2 Jak1 inhibitors.

1801 is for immuno therapy and is believed to better Deucravacitinib.

This has been stated by Tim on more than one occasion. Theoretically is certainly should give better and more rapid efficacy over Deucravacitinib ie PAS175 score over 12 weeks and not 16 weeks.

Effectively looking at 75% or greater reduction in initial skin affected surface area.

This is merely one indication. There are a myriad of Indications that 1801 may prove to be best in class in.

Triple negative breast cancer and ovarian particularly high grade serious ovarian cancer there exists a need for improved treatments.

My perception is that selective chk1 forms a necessary component in the treatment of cancer.

How else do you remove DDR checkpoint without 100% removal of complete cancer and that includes individual cells. Surgery is in many cases an option but it remains that any cancerous cells not removed will multiply which is then likely to move to metastases.

Looking at it logically this is an ideal opportunity for a DNA damaging agent and CHK1 inhibitor and dare I say PD-1 inhibitor too.

Effects of 737, LDG and PD-1 in combo, over any single application being remarkable.

It has been posted before but good that you have found and posted again. There exists additional reporting.

737 was good at what it is designed for.

Half on here don't know what it was designed for and that was to prevent cancer growth.

Very good results given in most Indications that chart showed stabilised disease.

Cancer cells are not apoptic which means like healthy cells they have no programmed cell death.

In combo therapy with the addition of DNA cell damaging agents is thus the way forward.

As is also PD-1 which is a process of recognising a cancerous cell and its destruction thereof. Pd meaning programmed death.

PD-1 inhibitors as far as l know are monoclonal antibodies. Clue here is that compounds ending in mab are monoclonal antibodies.

As we begin to destroy cancer cells so does the cancer rapidly want to replicate but also will develop resistance to a treatment.

Mutation at P53 checkpoint in over 50% of cancers.

HGSOC this figure reaches 98% in its advanced stages.

Indeed cancers can overcome the P53 DDR checkpoint and develop an alternative pathway, but by stating this it also gives value to the importance of the CHK1 pathway in the first place.

On the subject of mutations l trust the two damaged cells continue to marvel what is posted here.

I formerly to BoD will know of trial results specifically no adverse effects observed.

The full data analysis of patients that would have had follow up medical observation and tests post cessation of dosing will need to be recorded and analysed.

We have no adverse events observed and Sareum will not know a great deal more, with the exception being biomarker data.

A very pessimistic 10% risk factor on this on a one year turnover in SLE alone is 1.83 billion.

We have as we know as contained in previous RNS that we have a similar profile to Baricitinib.

Below is a link and the 2nd paragraph in particular is good reading as gives an indication as to what immuno disorders 1801 could initially prove to be successful in.

Not mentioned in the link is Baricutinib being trialled in Alzheimers. Started from memory around end of 2023.
