I have recently spent several hours looking into computational chemistry which we know TM has a PhD in.

Such aspects as quantum chemistry are used in a prediction how electrons will end up creating molecules which produce or inhibit enzymes.

Take an enzyme as a precursor to an event happening.

Wave particle duality and Shcrodinger equation involvement in prediction but hence only accurate in single electron atoms ie any Hydrogen isotope.

Multi electron atoms are in a sense to gain anything of reliability is basically impossible.

Now this is modelling of a molecule. Molecules are used in our compounds. Our molecules are heavily patent protected so anyone else has to develop their own.

Our molecules are not new but unique snd owned by Sareum. The molecules can be proven in certain parts of early clinical trial to give a resultant efficacy.

Ultimately it is the phase 2 trial that really indicates the potential of a compound.

But importantly initial safety must be achieved first.

Interesting links and goes to show the ever increasing amount of competition.

In preclinical trials there is and l will use the word tendency to administer higher doses with regards to efficacy.

You then know the ability of a drug in a potential range of Indications

However, the down side is that the amounts administered to humans as in therapy are way below this.

Now if we take it that the compound works in this preclinical activities it will work in humans.

However, the dilemma here is off target effects and adverse events.

As the dose increases so does the risk of adverse events.

We can be fairly sure of success in phase1 as merely looks at the safety.

Phase 2 in a trial is looking at and testing in a given indication and administering optimum dosages.

Now a large percentage of drugs by the time they have achieved satisfactory efficacy have also achieved unsatisfactory off target and or adverse events resulting in failing to meet primary endpoints and hence failure.

1801 results with regards to the safety profile and importantly refer to the initial pharmokinetic and even more so to the pharmacodynamic will enable a starting point for dosing a particular indication in humans.

Of most importance here is the full range of data from the SAD & MAD studies of the 1801 in Phase 1 clinical trial and the interpretation thereof.

You are nothing more than a doom and gloom merchant when it suits you. Then also predicting 1 to 2 billion valuations also when it suits you.

How much do you value the company at this moment in time?

The results of the multiple ascending dose should give a result on the pharmacodynamic which is the effect the drug has on the body and its various organs.

Pharmakinetics is the effect of the organism being treated on the drug as we know.

Pretty certain that Sareum have good idea as to the areas of indication for 1801 treatment as basically that is at the molecule design stage.

However they are being very quiet on this at the moment.

Computational chemistry and modelling at early design stage, Followed by in vivo in vitro testing and at this stage retweaking of the molecule if chosen.

Csn you explain exactly as to what you refer to by dosing levels not as predicted?

Dosing levels were carried out in single ascending dose and multiple ascending dose up to a predetermined maximum of 300mg per day ie 30% of the maximum therapeutic dose indicated by Sareum of 1000mg

Certain aspects here as therapeutic range by Sareum was given as mg per kilo of body weight

Now if you have a person who takes part and weighs 50 kg and takes 100 mg per day is it equivalent to a person who weighs 100 kg and takes 200mg per day?

Maximum therapeutic dose was 12.5 mg per kilo of body weight up to a maximum dose of 1000mg per day.

It is important to recognise that this is at the top end of therapeutic range and bugger all to do with MTD.

Side effects to kick in early would have been realised in the single ascending dose from the 5mg per day up to the 300mg per day.

There were no serious adverse events reported in this part of the trial.

This allowed progression to multiple ascending dose. This carrief out over a period of time.

This is no immediate concern of any serious adverse events.

This does not of course rule out non serious adverse events or reactions that have occurred in other kinase inhibitors that have been approved for clinical use.

The importance here is to establish that there is not sufficient adverse reactions leading to the restrictive black box warnings.

We are mainly Tyk2 which has selectivity over Jak1, 2 & 3

This selectivity should ensure very little off target effects to maximise safety profile.

There are other areas such as risk of zoster to myocarditis,

As for the market is expecting is 'obviously' expecting news in H1, personally l do not see it as obvious and further more no mention of this in the RNS

The RNS was released by the Nomad. An expectation is all it is nothing else. Has no legal binding or Jack 5hit.

As for the last part of why companies release an RNS on a Friday to protect from sharp falls, are you implying that companies withhold market sensitive information to protect the SP?

As with regards to bad news first of all we have to interpret what bad news is.

Yours is a country mile away from mine.

Bad news I interpret would be results that would prevent continued development ie failure.

I certainly do not see that happening. Any slight thought of that l would be out like a shot.

No need to inform of delay as not a deadline.

As for the wording of the RNS that as such, it does not matter. I say yes it does.

Potters just exactly what would you expect to be contained within a bad RNS reference to 1801.

The Indications given by the content contained within the RNS suggest to me we are on target for very favourable results.

You very likely see it differently.

One moment you are projecting a 1 to 2 billion company, the next thing protecting yourself by either not buying but selling in case of a bad RNS on a Friday

What different does an RNS release make on a Friday as is just another trading day.

They used the word expectation, some here trying to interpret as some sort of deadline and we are doomed if we are not informed of whether it is good or not.

On decent RNS l would hazard a guess in the 60 to 80p region, maybe more.

Then the downside of course is non dilutive funding which unless resolved in the very near future will likely cause a continually downward drift.

I believe the problem was not establishing a MTD.

It is has and as far as l know an industry practice whereby the Jaki inhibitors are permitted to be administered up to a maximum of 30% maximum therapeutic dose per day.

Since the maximum therapeutic dose of our SDC compounds was given as 1000mg per day

The maximum dosing allowed by the trial was thus 300mg a day.

I remember Thoth2 during the period of establishing an MTD of 30 times maximum therapeutic dose being sufficient in the absence of a non achieved MTD being reached.

I remember Tim in an interview with the smooth talking Aussie Andrew Scott, that 30 x had been reached and where do we go from here 100 times,?

MTD is basically a dose that can or may lead to death. Long term safety here is not the main concern.

This is of particular importance however to oncology.

Heavy dosing is not the main issue when treating a patient with an attempt to increase a life expectancy from a few months to a few years if there is a risk of an associated adverse side effect 5 or 10 years further down the line.

We await news and Tim and Co will release an RNS when they deem ot fit to ie following validation and data interpretation.

Until validated data confirms the correctness and suitability of 1801 and the clinical trial , Sareum are not going to release an RNS.

They got their fingers burnt expecting 1801 to go straight to phase1 via MHRA

Their exists international standards that must be complied with.

Validated data enables as in the case of Sareums 1801 to continue to the next clinical stage of development, without it, it cannot.

'Worthwhile continuing ' is very vague.

It involves many aspects, such as evaluating other parameters, such as finance, competition from competitors and being commercially viable.

Validation is a formal process which has achieved legal conformity.

Why would Sareum issue an RNS indicating a delay?

They await trial data results.

This data needs to be interpreted and released in the RNS

There will be pages of it to scroll through.

They may already have it.

Being pedantic here but the expectation is when Sareum expect the results. Results are not going to be sent to shareholders so stop seeing it such.

As l say there will be pages upon pages of data.

It gets closer by the day, be patient.
