Cambridge, UK, 30 July 2021 - Sareum Holdings plc (AIM: SAR), the specialist drug development company delivering targeted small molecule therapeutics to improve the treatment of cancer and autoimmune diseases, announces that the United States Patent and Trademark Office has issued a Notice of Allowance for a patent in respect of an invention associated with Sareum's proprietary SDC-1802 TYK2/JAK1 Kinase Inhibitor Programme (the "SDC-1802 Programme").

The patent (US Patent Application no. 16/343,639) will protect the SDC-1802 molecule and pharmaceutical preparations thereof as a therapeutic to treat cancer selected from pancreatic, colorectal and kidney cancers, melanoma, and B-cell lymphoma by inhibiting TYK2 kinase. This programme is in preclinical development.

The Company expects that the patent will be granted within three months, subject to certain formalities being completed.

This was ongoing as far as I know of the 'crystallisation of a compound ' that was originally applied for in April 2020.

I will look through old history and dig out for you.

Crogy34. You are welcome. my feelings of the secret patent is that the patent is for the 'crystallisation of a compound' using this method it enables the molecules to be arranged precisely.
if there were another compound invented we would have heard many moons ago. New or modified compound with intended target and hence received RNS notification.

i will post more on this later for you. But I will stress here the importance of molecular structure and formation into compounds to control cell signalling cannot be over emphasised.

The patent/ patents are big news.
Effectively by using the crystallisation of a compound to ensure maximum purity they now cover compounds ie 1801 and 1802 for a period of 17 years from the 26th of October 2021.
it may well prove extremely likely that should sareum use this method of manufacture for FLT+3 then this will also benefit from 17 years of patent protection. Although original patents worldwide for FLT+3 were granted around 2013 ( from memory) it effectively increase patent protection on FLT+3 alone from 2033 to 2038.

Patent protection on all of wholly owned Sareum compounds increased by the patented protection of its manufacturing process. Ie no other bugger can formulate our compounds to the exact standards required.

So yes very much underestimated.
Longer term protection of our compounds
100% potential purity obtainable by using this manufacturing process.

Mafuta, I have seen many things in the past that have failed to attention to detail especially in large companies. You can point out perceived problems but they will ignore just to get a product in to market.
Many years ago with FLT+3 Tim an co spent many months selecting suitable candidates for preclinical trials, they narrowed down to two compounds before carrying out extensive preclinical work to establish which one was best.

Effectively they have done the same here and we have 1801 and 1802. Many a company would have stuck to the one compound to treat all ailments.

The only problems they have experienced was not, does the compound work, but the difficulty in manufacturing the compound in sufficient quantities to enable MTD toxicity and safety profile.

My opinion FWIIW is the compounds are formulated from a selection of molecules pertaining to the later patent protection of Tyk 2 compounds. There are all sorts of molecules in here.

A separate patent protection for FLT+3 will longer be required as will be protected by the crystallisation of a compound. Clearly these formulation issues were know by Sareum. The Chinese could not formulate the oral route. Low and behold around the same time ( give or take a month) and we have a crystallisation of a compound patent applied for. As Tim state stated these formulation issues were outside their scope. Clearly they have resolved these issues by specialist in this field. Attention to detail again. It is all about getting it right. They have effectively not just looked at what is right with a compound but also identified, and now resolved these areas that posed problems.

Myself am more confident than ever. I wonder how much the HNWI's new when they came aboard.

AFAIK 1802 was used in investigations with regards to Lupus. Lupus is autoimmune and therefore should prove more favourable with 1801.
However, I see nothing to suggest why 1802 could not be used for Lupus.
Tim and co hypothesised that 1802 could be used for Covid 19 but were of the opinion that 1801 was better suited. Results for 1801 were pretty top notch.
They have access to SKIL ( sareum kinase inhibitor library for those that do not know) and in this instance would be regarded as experts in this field. As Tim said we are not experts in biology, but what we are very good at is chemistry.

A delay of a few months with regard to manufacture of capsules for SDC1801 which taken into context does not significantly affect progress of SDC1802 I believe.
It would be interesting to know whether or when likely hood of manufacture of SDC1802 capsule.

There will be interested parties which now can only gather in number.

If we are not already in discussions with GSK then informal discussions will take a while. Once an acceptable toxicity and safety profile has been established, then the blue touch paper will be have been well and truly lit.

However, I would not rule out an on license in the very near future given the importance of our crystallisation of a compound enabling the manufacture of an oral route via capsule.

Good afternoon Thoth. A lot goes on behind closed doors that we are not privy to both negative and positive.
We have been involved in discussions with interested parties for ages. Tim and co know who they are and most likely the HNWI's will know a tad more than we do.

Dexamethasone is a 60 year old corticoid treatment. As you know our profile is better in Covid 19 alone.
Dexamethasone is used in treatment of Lupus, ulcerative colitis and psoriasis. I would also suggest that our profile will be substantially greater in these areas. Steroids are not without their side affects and has been suggested that the use of Dexamethasone can increase the incidence of long term affects of Covid 19. A double edged sword so as to speak. Reports give the most effective positive data as 35% reduction in death as opposed to a placebo.
Plenty of room here for improvement via an alternative treatment.
Interesting to note also, is the US DoD funded money into SRI research into 1802 for treatment of Lupus bearing in mind that Dexamethasone has been round for 60 years and approved as treatment for.
Il-6 is not new as we know. Il-6 also plays a role in muscle regeneration.
A IL-6 can be signalled via Jak1 and Tyk 2.
Tyk 2 is just a pathway that is used to signal our innate and adaptive immune cells.
However, just because an inhibitor is Tyk2 does not automatically make it an inhibitor of Il-6. Fortunately ours does inhibit effectively IL-6 in conjunction with Jak 1 which signals via a different STAT pathway.

A few months delay given to us by the RNS and the BB here gets inundated with disruptive drivel.
Biggest plus is the capsule form of oral administration. I am not concerned with the negative posts of certain individuals that sing the praises of what other companies are intending to do to treat Covid 19, or the phases they are aiming to reach in the near future.

As the time delay inevitably reduces, so will the development of both our SDC compounds have time to reach their maximum potential. Years have been spent in development of our compounds by dedicated world class persons. Attention to detail has been second to none. This level of attention to detail is the difference between success and failure. Attention to detail confirmed via the oral method of administration via a capsule. ( would hazard an educated guess at liquid suspension here)

Summarise. Yes I get your drift. As you well know too, we are in discussions with more than one party. These companies will not be minnows and will have to have access to very deep pockets I believe.. Deals take many months to conclude. We will be updated of developments on the way with regards to our compounds.

Interesting post Jamesp21. Worthy of thought. My opinion for what it is worth that on a big buy price would have to rise to release limits triggering sells. Clearly not many shares available in free float.

Good post BD, an interesting question to put to the board would be do they have any intent to use, or do they consider the application of the 'crystallisation of a compound' to resolve formulation issues experienced with Aurora + FLT3?
Almost identical compound of molecules I believe. It is the crystallisation of these molecular compounds that would eradicate formulation issues.

Strean not been around around long. There are and will be others like him. Ignorance and lack of understanding may prove very costly to these people who will jump from share to share in an effort to make a fast buck. The in and out brigade. Then we have the HNWI's , With more money to invest than they will ever have in lives. They are not in and out brigade but looking for a decent return on their investment of 3, 4 , 5 or ten times and potentially more on their initial outlay. If it takes 6 months longer than anticipated do they really care. They are of a different mindset. It all comes to he who waits.
The likes of Strean neither understand Sareum, its compounds or the science behind it. We are where we are in our lives by the product of our own thinking. We invest, or should invest on the understanding of what we are invested in. If we do not understand then leave well alone.

I believe in its compounds as do the HNWI's with the added benefit of my buy in price is very much less than theirs,

Capsule as an oral route is outstanding and vastly underestimated in its ability to maximise its ADME. If you look at the original patent is included in the oral form as a method . That of course is paying attention to detail.

It is very good to keep your options open. I have kept my options open here for nigh 10 years. I am in no hurry to sell. Likewise I would hazard an educated guess that the HNWI's in no hurry to sell either, albeit significantly wealthier than me I would guess.

Nothing more than delays ultimately caused by

a. Delay in manufacturing process.
b. Lengthy time to prepare compounds to the requisite standard.

It is of paramount importance to produce our compounds to the highest possible standards available. Attention to detail pays dividends. Any lack or shortfall here and we compromise the effectiveness and possible safety profile. These concerns have both been realised and now resolved by Sareum. April 2020 Tim stated concerns of producing compound in high enough concentrations and looking at different means of manufacturing to overcome this. I cannot stress the importance enough.

The downside is inevitable delay in waiting for a patent approval and time consuming method in production thereof, not just restricted to the aforementioned but also delays effected by Covid 19 restrictions.

Nothing else has changed. Hurdles still need to be overcome ie completion of toxicology tests and then presentation of clinical trial details prior to CTA. I envisage no problems in this aspect with the exception of delays in manufacturing. Consultants have been appointed with regards to clinical trials. I am certain they are more than capable of satisfying the regulator with their detailed outlines and proposals.

In the great scheme of things a very positive note on the development of our compounds.

The delay unfortunately whilst not bad is not as good.
