To be precise I would say the capsule form of oral administration will be used in all aspects of immuno treatment and that will include treatment for Covid 19. It appears that we are the only company being able to treat the later serious stages of covid. The only other inhibitor that might have proved some degree of effectiveness in this is Baricitinib, which of course we all know cannot be used on its own and included safety profile concerns. The FDA withdrew its EUA many moons ago.

A mixed bag of news. Spot on with capsule manufacture. Much easier uptake by the digestive system as opposed to tablet. All made possible by crystallisation of a compound patented invention. I would hazard a guess that this work was commenced on receiving news of the granting of patent. Higher concentrations possible too.

A few months delay in the present climate is not surprising. finalisation of preclinical toxicity and safety profile also held up due to delay in manufacture of capsules by companies affected by covid 19. Overall looks extremely encouraging outside of the delays with regards to an effective treatment and also will attract interest from 3rd parties.

Another question I would ask, is are you in a position at this moment to produce sufficient manufactured compounds for phase 1 trials use?

Take Il-6 as an example. Tyk2 inhibits via one pathway and Jak1 another. Think of it as 2 roads. ! is blocked so we use the other. I have a feeling that pharmas have jumped aboard TYK2 and shut out Jak1 due to toxicity issues which are not big with Jak1 and way way below Jak3.

jaks work best in pairs and our preclinical data so far indicates we are pretty well spot on.

Indeed it will be phase one trials that will confirm our safety profile.

That proving satisfactory the world becomes our oyster.

Nothing out there as far as I can see near us. There are not many i know. We are not the first but odds on favourite to be 1st in class I feel.

All Tyk 2 inhibitors do not exhibit the range. Many do not inhibit IL-6. Interstingly I find that IL-6 is inhibited by Jak 1 and Tyk 2. However both signal through a different STAT pathway.

Jaks work best in pairs. So I am assuming that as Tyk 2 will block a pathway it is rendered useless on its own so in a sense this is not utilised by pure Tyk2 manufacturers. Jak1 is also required . Think of it as two different roads. One is blocked and the other remains open. In an effort to remove Jak1 which has side effects they have in effect severely limited full potential of TYK 2. Too late to change their molecule me thinks.

As stated by Tim an optimal amount of jak 1. Clearly they new or could see the importance and relevance thereof.

In a nutshell the formation of molecules in the make up of a compound are ensured by crystallisation. The invention covers such measures as the quality control and preciseness of such.
Each molecule has a target and the correct arrangement of these molecules are vital to its success.

An interesting point at one time earlier on it was stated that it would require 20k virions to create infection. That figure has now been significantly to 1000 virions or less. Whether this is due to the differing strains or poor initial research i cannot answer.

Typical size of Covid 19 virus is 50 nm x 100nm.
1 mm is 1000 microns
1000 nm is 1 micron.
the thicness of a hman hair on the head is around 2 one thousandths of an inch. 25 microns is one thousandth of an inch,
500 virions laid end to end is around the thickness of a human hair,
we can go 1 step further. area of a circle is Pi x radius squared.
therefore we can use 250 x 250 x3.14 = 62500 x call it 3 as am doing in my head. that is 187,500 .
finally volume of sphere = four thirds Pi times radius cubed 250 x 250 x250 x 3 x 1.33

= 62,343,750
In a sphere the diameter of the thickness of a human hair you could fit this amount.

Fortunately the virus will not be found in such concentrations.

Droplets similar to when we sneeze are around 100 to 200 microns in diameter. The smaller the droplet the longer it will become airborne and hence drift.

What does a vaccine consist of?

An inert or weakened virus is used for vaccinations. The body has mechanisms ( in this case macrophages and neutrophils) which are released by our immune system.
These little buggers recognise the protein and consumes it. It know nothing of its RNA.

The spike protein can change and does change. The bodies immune system does not at first recognise the change in spike protein. An RNA virus can only replicate once absorbed into a cell whereby the virion then uses the hosts DNA to replicate many thousands of copies. There will be mutations and not all these will be harmful.

One virion can produce over 1 billion copies in a 24hr period.

A change in the nucleic acid in the case of RNA single stranded can and does alter. It is this change to the RNA which has the ability to mutate to survive that causes the biggest problems. The RNA learns and when invades new cells, resistance is part of the inherent nature of a virus to survive, the RNA will modify itself to survive using the hosts own DNA. thousands of mutations will take place and nearly all will pose no problem at all. Occasionally there will be a mutation or mutations that will cause problems. In deed many of the mutations will die off.

Interestingly here, I will add, a woman who were HIV positive having produced over 30 different variants of covid over a 200 day period. It is not normal to have more than one virus in our bodies as a virus in their effort to survive will become dominant over the other.

Rather odd then that a virus can share and live with ( take it as recognising allies) . Covid will live and mutate as it wishes in an HIV infected person. The immune system in an HIV infected person is weakened. Too such an extent the viruses will be exposed to our antibodies that are insufficient to eradicate completely. In this environment the virus will mutate at will. It will also live with many variants of its own making.

To kill the virus our immune system must first recognise it. Once it recognises the outer protein as an invader our innate and adaptive immune system will seek to destroy. Macrophages and neutrophils being highly effective killers which emit powerful enzymes which kill and consume the cells ( phagocytosis) . We then attract lymphocytes which learn to recognise the invader and add this to our adaptive immune system memory.

Such is the replication factor caused by the Corona virus into our bodies that by the time it has learned of the invader an overeaction or to be more precise a cytokine storm is produced which not only seeks to destroy the invader but will also destroy our healthy cells.

The above is a fairly simple idea how viruses work.

Complacency, complete disregard for others and a blatant refusal to take necessary precautions by anti government urchins. There still exists those that believe this does not exist and is all made up to impart control over ones life. A bad time of year for it to happen. You can now expect significant rises in people testing positive followed a short time later by a rise in fatalities.

Of course the virus will be exposed to people who are already vaccinated. Being subject to antibodies it will mutate in an effort to survive. The virus and strains it produces will thus become more resistant.

Viral loading may very well be reduced. However, mutation of virus will evolve to combat this.

i would expect news due very soon on how our 1801 immunotherapy is progressing in this area. It was not originally intended as a target. Cytokines are caused by our immune systems and it does not matter how much a virus mutates, a fine balancing of our immune systems, to prevent over response, will offer far more in the longer term as an effective treatment to seriously affected individuals. Whether used in conjunction with an anti viral or as a stand alone treatment.

I can come across as being very much a ramper. It is my beliefs and understandings I post.

Small companies are down trodden by the large. I remember dys of Sareum with 15 million market cap. Many hurdles had to be overcome. These hurdles have been overcome. There exists still an element of doubt. Nothing is 100% guaranteed.
We have two potential wholly Sareum created kinase inhibitors. Although there exists much competition in this area we are at the forefront. A small company that has struggled and overcome problems beyond its control to acheive where we are today.

We are the only TYK2 Jak 1 inhibitor available to licence without complications.

We have demonstrated excellent safety profile in in-vitro and in vivo preclinical trials. Safety has always been a matter of concern with Sareum.
Our compounds have been very well researched. Our efficacy has been top notch. Problems with formulation of compounds with regards to solubility yes. These have been overcome.

We must be the only company that has 2 compounds with over 9 years of development and have, starting next week a 17 year period of patent protection. Any big pharma that had achieved results similar that we have achieved would be regarded as a blockbuster.

Biggest is not always best. It is dedication that counts. We are on the radar big time. Deals are not done overnight. Big pharmas patents are running out and they have failures galore in their up and coming pipelines.

Auto immune and cancer treatments are multibillion dollar revenues per year.

Sareum shortly will play a part in this. Our compounds are unique and now very well protected. All that remains is they prove a safety profile in human trials then the world shall become our oyster.

If and when news is released and it is just a matter of time then it will blow.
Any indication of an on license even with lowish upfront payment will see todays prices pale into insignificance. It will remove the shackles of a financially restrained company.
Market cap minimum of 500 million I would suggest on news for 1801 alone. Irrelevant of Covid 19.
I think bigger in store with Covid 19 and also the benefits should Covid 19 take a turn for the worse,notwithstanding the risk of another outbreak of a virus that creates similar respiratory problems via cytokine storm.

Sareum never had an income and very little raised by up front payments. Once a significant milestone is made then the doubts of surviving financially will be removed.

HNWI have invested. Deal of some sorts is approaching fast. We are the only TYk2 Jak1 inhibitor at late clinical stage without any complications. Excellent LADMET properties too. Stars are pretty well lined up I feel.

I am not ramping but this has been many years in the making.
