Nothing Tim is aware of to mar the promising toxicology results. Until a final report is made all we can do is wait. There is very little an interested partner can do in the meantime as they will need to know in depth, the assessment of toxicology data prior to negotiating any form of a deal. Likewise it will be the same for any investors putting up money. Toxicology is the most important hurdle to cross. Looks like we have it in the bag, but of course not yet official.

I do not see any share consolidation arising prior to receiving outcome of toxicity data. There simply is no need. It is there as an option later down the line. PH can play as much as they want with the SP at the current time. It does not change one Iota the true value of 1801 and 1802 post official positive toxicity results.

Intellectual property
The Company had two new US patents granted during 2021 (in
January and September) that reinforce the patent protection
for SDC-1802 and its use in treating certain cancers (including
pancreatic, colorectal and kidney cancers, melanoma, and
B-cell lymphoma) by inhibiting TYK2 kinase.
Additionally, a patent application describing a number of
crystalline forms of a TYK2 inhibitor for medicinal applications,
filed by Sareum in April 2020, was published on 14 October
2021. This type of patent is standard pharmaceutical industry
practice and an important step to fully protect the intellectual
property surrounding the Company’s research programmes
and to extend the life span of its patent portfolio.

Encouragingly, in August 2021, Sierra announced the inlicensing of the BET inhibitor AZD5153 (now known as
SRA515) from AstraZeneca and noted potential combinations
with SRA737 as a possible pipeline expansion opportunity. In
September 2021, Sierra provided an update that referred to
the initiation of additional clinical studies with pipeline agents
including SRA737 in other haematologic and solid tumour
indications in the first half of 2022. Specifically, reference was
made to a potential role for SRA737 in combination studies
in solid tumours, including pancreatic cancer, where patients
have become resistant to PARP inhibitors.

I did read somewhere and cannot find again that the preclinical studies of SRA737, LDG and ICI (immuno checkpoint inhibitors) were superior to just SRA737 and LDG.

cancers. There is notable interest in the pharmaceutical

and JAK1, and particularly for those that can avoid side effects
from inadvertent activity via JAK2 or JAK3.
These differentiated programmes therefore represent unique
licensing opportunities for pharmaceutical partners as there
are currently no marketed products with the selectivity profile
of SDC-1801 and SDC-1802.

At the moment Blastoid Sierra do not have the funds to pursue SRA737. I have posted before and my belief they can do very little until shot of Momelotinib.
At the moment Sareum have taken them out of the equation. Raising funds via HNWI continues our progress unimpeded.
We can support early stages of 1801.
We have options of raising considerable funds and a very much likely hood of raising funds from a license. One we know about the other we do not.
Either way whether through investment or licensing both we will want in at cheapest price they can obtain. In this case we do not have one card in our hand but two.

Toxicology results to an extent will play a part here. Neither Sareum or a potential licensee will want to give too much information away to competitors.

Its all down to the individual molecules and the ratios thereof. With the capsule formulation alongside Crystallisation of a formula it will make us second to none.

PH are one of lives parasites I feel.

Look at the origin of Covid 19. it is found neither to exist in bats or pangolins. Yet we find it in mink and deer.

Covid 19 is just a small percentage of what an effective low toxicity Jak 1 Tyk2 inhibitor can treat. An effective treatment has yet to be found.

There are some big pharmas too which will lose patent protection from 2025 and big financial losses due to generic manufacture of drugs.
Good thing with our Tyk 2, is the crystalisation of a compound is patent protected for 17 years. This method of manufacture should ensure up to 100% purity of a compound. If this method is not used in manufacture then quality cannot be guaranteed. Not good for the generic manufacturers unless they want to pay us royalties.

Good to see SRA 737 get a mention. CHK 1 that prevents effects of DDR, the rapid cell mitosis and replication, the blocking of the P53 cell cycle checkpoint thus inhibiting cell replication.
As we know cancer cells do not suffer from apoptosis ( ie programmed cell death) like the rest of the cells in our body)
From memory phase 2 trials gave good results for our CHK 1. it is licensed to Sierra but sareum designed the compound in conjunction with ICR.

In addition taken from RNS 2019 prior to re negotiated Sierra SRA737 initial milestone fiasco.

Bare in mind in 2012 Tim also mentioned a transformational deal, which never materialised with a consequential drop in the SP, for which he were seriously lambasted for. He has not and will make the same mistake again.

We will get to know of just how good our results are when an RNS is released. Attention to detail and getting it 'spot on' will reap its rewards.

Pfizers 11a trials conclusion below.

Results: In total, 212 patients in 35 sites were treated; mean (SD) baseline PASI score was 20.8 (7.68). Decreases in PASI at week 12 were statistically significant compared with placebo in five treatment groups. The greatest change from baseline (least squares mean change -17.3 [95% confidence interval, -20.0 to -14.6]) was observed in the 30-mg QD continuous treatment group. Overall, 136 patients experienced treatment-emergent adverse events, including six serious adverse events in five patients and 13 discontinuations in treatment groups because of adverse events. No herpes zoster cases or major adverse cardiac events including thromboembolic events occurred.

Conclusions: PF-06700841 was generally effective and well tolerated in patients with moderate-to-severe plaque psoriasis.

QD for those that do not know, defines one a day ( from Latin quaque die)

Our proposed range for dosage was 12.5 mg total body, up to 10mg per kilo of body, weight maximum 1000mg per individual. No adverse effects were found in preclinical studies, and to establish an MTD at 30 times treatment levels did not produce an MTD, TM stated that we may need to go as high as 100 times treatment these levels. Extremely complex. It is the molecules that make up control of each target and their respective proportion in my view are of extreme importance. If I were pessimistic I would say at least as good as.

Covid is not going away soon. Eventually it will become endemic. An effective treatment will be required not just for Covid as it is at present, but also as a treatment in contingency plans should we experience any forthcoming serious SARS outbreaks.
