Proposal process for COVID-19 treatments
An open nominations portal will be managed by UKRI, who will collate information on compounds based on the evidence and data provided by academics, clinicians and industry experts as well as independently sourced information. A core team of due-diligence managers will be responsible for validating the information and will create briefings on potential compounds. This information will feed into the UK CTAP’s specialist sub panels who will triage and prioritise the compounds for consideration by UK-CTAP.
If you wish to submit a proposal for a therapeutic to enter RECOVERY+, REMAP-CAP, PRINCIPLE or AGILE , you should complete a proposal form. Following the submission of your proposal you will receive an acknowledgement email with details of the onward process.

Sareum continues to believe that the licensing agreement amendment signed in November 2020 between CRT Pioneer Fund and Sierra will help expedite the SRA737 development programme.

Patient benefit is a paramount goal so we need to ensure that innovations arising from CRUK-funded research are continually developed and introduced to market in a timely and efficient manner. To this end, we need our partners to commit to:
• Diligence provisions which address product development in oncology applications and pursuit of marketing approval of such products in major markets, specifically the UK.
• Generation of a development plan (and provision of annual updates) covering significant activities, associated timelines and key decision points.
• A diligence monitoring mechanism focused on constructive two-way dialogue and a commitment to regular face-to-face interactions to review development.
• A right for us to regain rights to a project (including partner’s IP to the extent required for onward commercialisation) should our partner no longer be developing it (in exchange for a revenue share reflecting the contributions of the parties).
• Provision for biomarker IP of generic value to patients/ projects to be used freely (including by 3rd parties).

Hi Celtic - given that it was less than a month ago (and less than 6 weeks from the scheduled end of the UKRI funded preliminary studies) that the bod commented as below, it would surely be very surprising if 1801 is progressed further via Agile or similar.
“We were also pleased to receive UK grant funding to investigate the therapeutic potential of SDC-1801 in severe phase Covid-19. Early indications are encouraging, and we expect to see the final results from this initial six-month study mid-year 2021. If this study continues to be successful, it may allow us to apply for further non-dilutive government funding from the recently announced AGILE platform to advance this programme into clinical trials. We are further encouraged to note the UK Government commitment to accelerate the development of Covid-19 therapeutics through initiatives such as the DHSC-led Therapeutics Taskforce and believe that SDC-1801 could benefit from this.

Specific inhibition
Janus kinase (JAK) inhibitors are powerful therapeutics used to treat inflammatory diseases but are also linked to immune suppression and complications associated with viral infections. Schnepf et al. show that selective inhibition of tyrosine kinase 2 (TYK2) can block potentially noxious type I IFN signaling but does not alter IFN-? signaling, whereas the JAK1/2 inhibitor baricitinib blocks both types of IFN responses. Epithelial cells did not require TYK2 for IFN-?–driven gene expression or antiviral responses, and TYK2-deficient mice have intact IFN-?–mediated responses and are protected from influenza A virus infection. These findings indicate that TYK2 inhibition may be a better treatment option for type I interferonopathies.
Abstract
Inflammatory diseases are frequently treated with Janus kinase (JAK) inhibitors to diminish cytokine signaling. These treatments can lead to inadvertent immune suppression and may increase the risk of viral infection. Tyrosine kinase 2 (TYK2) is a JAK family member required for efficient type I interferon (IFN-a/ß) signaling. We report here that selective TYK2 inhibition preferentially blocked potentially detrimental type I IFN signaling, whereas IFN-?–mediated responses were largely preserved. In contrast, the clinically used JAK1/2 inhibitor baricitinib was equally potent in blocking IFN-a/ß– or IFN-?–driven responses. Mechanistically, we showed that epithelial cells did not require TYK2 for IFN-?–mediated signaling or antiviral protection. TYK2 deficiency diminished IFN-a–induced protection against lethal influenza virus infection in mice but did not impair IFN-?–mediated antiviral protection. Our findings suggest that selective TYK2 inhibitors used in place of broadly acting JAK1/2 inhibitors may represent a superior treatment option for type I interferonopathies to counteract inflammatory responses while preserving antiviral protection mediated by IFN-?.
