To further confirm that GPR56+ TEMRA cells were major components of IFN-?-producing cells in response to DENV, we sorted GPR56+ and GRP56- TEMRA cells from secondary DENV-infected donors (Supplementary Fig. 1c) and measured their production of IFN-? after stimulation with DENV CD4 T-cell megapool. We observed that a higher proportion of GRP56+ TERMA cells produced IFN-? compared with their GRP56- counterparts (Fig. 7d), further supporting the notion that DENV-specific TEMRA cells predominantly adopted a GPR56+ phenotype. Taken together, these data suggest that DENV-specific TEMRA cells are associated with a cytotoxic phenotype characterized by the expression of markers such as CD244, perforin, and especially GPR56. This is consistent with the observation that these cells were clonally expanded, which could result from multiple rounds of DENV infections.
