Well I’ve lost the impetus somewhat now, as basically the MTD dose hasn’t been reached. Maybe it doesn’t have to be though, if the 8mg/kg performs well enough at the next stage. Will be interested to see how the liver copes with a higher dose, after borgie brought that up, but it doesn’t have to double again. Maybe 12 mg/kg? As the 8 mg/kg performed so well to stop that particularly aggressive cancer in its tracks. That was the really tough patient to get under control, so was the big test for me in the results. I was slightly baffled as to why there was only one patient at 8 mg/kg and further testing was done at 4 mg/kg, when the point was to try and increase the dosage. Another cohort could be added easily to test higher doses in the next phase, but as it was working at these doses already, then I can see why they possibly didn’t feel the need to push it at this stage. The pharmacokinetic profiles certainly show that 8mg/kg looks fine, with a high level of certainty and opens the door for that to be increased, if needed.
