Hi RMM too me it’s a mis-leading tweet that has no relevance as it’s about targeting the viral mediators too stop the evasion of the immune system! It’s about stopping Covid19 inhibiting the JAK/STAT pathways, it’s basically jumping on too anything that say JAK in it!

So it looks as if you need a Tyk2 GOF (Gain of Function) rather than what the pharma’s have with Tyk2 LOF (Loss of Function)!

I don’t see this tweet in relation to inhibition of Tyk2 which is trying to be achieved in molecule design by companies, as this reads like COVID-19 evading the innate immunity, by COVID-19 inhibiting JAK-STAT pathways which is what the Tyk2 inhibiting molecules do, so common sense would lead too the creating a molecule to stop COVID-19 inhibiting theses pathways, the innate immune system is first line of defense and this is not the over reaction on the immune system and cytokines storm with the death of patients in the ICU, so in my mind the only relevant nature of the tweet is that it mentions JAK-STAT pathways!

I think this is a difference in how BMY and Nimbus are progressing their Tyk2 programs where their Tyk2 are classed as Allosteric inhibition, and are claiming this is the way forward, I’ve also put the link for the Catalytic site for other methods of inhibiting Tyk2, it would be good too look at this too see the difference in the development design between Tyk2,s other than selectivity profiles!

Damion is it because the research the company has done has implicated JAK1 inhibition as per articles but then then is also Tyk2 ones but have seen more evidence of JAK1 inhibition, as is thought to make more sense too dampen down the covid19 immune system overreaction than Tyk2, it could be that tofacitinib because it’s approved too market they could get through faster, but it is interesting that no other companies with Tyk2 assets have chosen this path, the Pfizer molecule which there is 2 the main one is more JAK1 than TYK2 slightly more you could can it a dual JAK1/TYK2 Inhibitor, but then it has the other one that has a similar profile to SAR’s with it being very selective for TYK2 but there is no evidence that they have them in mind too trial for Covid19!

The only way too see if Sareum is best in class is by results and gauging against the competition, Pfizer CEO has said that their Tyk2 is best in class, and also BMS is saying their pseudo-kinase Tyk2 is the way forward to inhibit Tyk2!

If the judgment is the selectivity the specificity of Tyk2 then Nimbus Tyk2 is more selective in their molecule, just because it is said too be best in class in doesn’t mean it is, it just means it might be best in class, but until it’s in the clinic and can compare results then that is the true representation of the status!

Hasn’t Novacyt got a test kit that’s in production that’s in high demand so there’s not the risk associated with trialing a new molecule too see if it works or not, just because a company is in the same sector that’s had some astounding gains, me myself don’t like comparing a company unless there are striking similarities involved so then you can compare the attributes of that company’s merits like for like, obviously I know this is a chat see and have seen over the years with members posting the companies with high valuations and referring too the company their investing in, could be a dangerous thing too do just because a company works for certain it might not for another, not saying Sareum is one it’s just not prudent to do so!
