They can't. 4 June '20 RNS said the number of shares issued could be increased out decreased. Last year's RNSes, etc didn't include that.

Yesterday Avacta wasn't listed so I filled in their simple form and today the company is there.

Whilst such patients may well benefit from AVA6000 as a marketed drug, they are excluded from this first-in-human trial on the precautionary principle, the same as restricts the total amount of AVA6000 that can be given and the dosing interval.

There will be no quantitative, statistical assessment of efficacy, only qualitative observations as the number of patients is too small for meaningful quantitative conclusions.

Patients may well have been on other, non-anthracycline types of chemo, no chemo (radiotherapy or nothing at all because +too late or refused therapy) or low amount of anthracyclines. One just can't make generalisations.

Doxorubicin resistance is due to prior doxorubicin exposure not to how poorly they are. Patients are recruited onto the trial on the basis that they have not previously received much doxorubicin (or other anthracyclines).

Exclusion: "Received a prior total cumulative anthracycline dose of >350mg/m² doxorubicin hydrochloride (or equivalent anthracycline dose)"

There doesn't seem to be much rationale for recruiting doxorubicin-resistant patients onto a doxorubicin trial, even a Phase 1a where the availability of volunteers is not a limitation (as stated by the CEO).

Looking at Avacta purely in financial terms from RNSs, the company website and regurgitating what other have written, it's very easy to see Avacta's 'massive' recent SP rise as just froth. Little wonder then that we've not seen the SP reflect all the good things we see. Shows how it pays to be well researched.

I'll have one last attempt. Let me put it this way.

If cell death > cell growth, then tumour regression
If cell death < cell growth, then tumour growth (e.g. an aggressive cancer)

A biospy will not tell you this. Measuring the tumour(s) will.

For this trial, neither dose nor frequency of dosing have been optimised. The tumours may be regressing. Ones that aren't may be caused to regress by upping the dose and/or the dosing frequency. Monitoring of size will be by scanning (X-ray, CT or whatever), not by biopsy.

I'd be very surprised if tumour sizes are not being monitored by scans in this trial.

Biopsies may show that the mode of action works (cells dying as expected). They do not show regression (decrease in tumour size). For that, the tumour needs to be measured.

Proof that it works by:

1) Side-effects profile.
2) PK following AVA6000, doxorubicin, doxorubicinol and other metabolites around the body and out, quantitatively.
3) Any imaging of the tumour.
4) Examination of cells from biopsy.

Of these, only 3) will show regression is taking place, that is killing of cancer cells > growth in number of cancer cells.

UK govt backing this horse is like their preference for nuclear over renewables, it's out there on the future certainly possibly maybe definitely perhaps. The questions are Does it work? How many patients can be treated per year? What's the cost of treatment? How much can the Tories and their family and friends screw out of it? and Are there alternatives just as effective, but cheaper?

A combination of pre|CISION to shrink or eradicate FAP-rich cancers and mRNA vaccine to finish them off if pre|CISION doesn't do it would be good for everyone: patients, hospitals, NHS budget and Tories if Alan can get in there quick with a brown envelope.

Agreed. pre|CISION with FAPI PET will be a game changer in cancer treatment (for those cancers with upregulation of FAP).
