Yes, Scancell's strategy has always been to develop LD's innovation from concept through to a product with clinical data and then to either license or sell the asset. There has never been any suggestion that Scancell would or could take a product all the way through to approval and marketing. Indeed the whole investment case for longs is the potential for a licensing deal/sale, whatever the product/platform.

Yes of course that's an entirely reasonable suggestion but the amount is a tiny fraction of the value of the SCLP shares some believe he has sold so I'm not sure it's too relevant. The cash in his ISA could have come from anywhere.

If you don't feel the tone and sarcasm in your response to my perfectly polite post merits an apology then fair enough. I have no idea what relevance you attach to David Evans closing down a private consultancy company that was likely set up to channel personal fees - what is your point?

As for whether DE is behind the sells, You could be right and he may well be selling his holdings. On the other hand he may be reassured by the change in leadership and may have decided to continue to support Lindy Durrant - we just don't know. The sells could be coming from anywhere. There are several funds sitting on very healthy profits, some who have been invested for many years. As Scancell's share price rises and hopefully continues to rise then we have to expect some fund managers to take profit and that may be what we're seeing. I'm not sure it matters that much either way - it's no big deal.

LL - there was no 1.5m buy for Intuitive Investments, it was a transfer of 758k shares between David Evan's nominee and ISA accounts.

Phase I trial results for RXC004 will be delivered by Principal Investigator, Natalie Cook via an oral presentation at ESMO on 20th September. Will be interesting to see the difference in responses between Wnt-ligand driven cancers and non Wnt-ligand driven cancers.

Yes, was presented at SITC20 but I believe at the time they hadn't carried out analysis of genomic alterations in all patients. Hopefully further analysis will have been carried out and the results will be presented at ESMO.

The neoadjuvant ESMO presentation is the one that interests me the most. The first line treatment wherever possible for virtually every solid cancer is surgical excision and the potential market is therefore vast - much larger than any other setting. It's also a market that is being targeted by the checkpoint inhibitor developers. It will very exciting if the neoadjuvant study can produce hard evidence that MRx0518 directly results in changes to patient's tumours prior to surgery which in turn results in clinical benefit.

Agree, SCIB1 recruitment was always going to be slow but Modi1 has none of the restrictions of SCIB1 and sadly there will probably be no shortage of patients who meet the entry criteria.

By the way, great to have Christian Ottensmeier as Principal Investigator - he is of course a member of Scancell's Clinical Advisory Board

The UK collaborator mentioned by PhamaJet is DIOSynVax. They're a spin out from University of Cambridge and signed a collaboration with PharmaJet last year. Interesting to have a quick look at DIOSynVax - several similarities to Scancell. Both collaborating with the universities they spun out from to develop a covid vax, both received very similar amounts of funding from Innovate UK, both using PharmaJet, both developing 2nd gen DNA vaccines.

Well spotted last night and good to see all study centres showing as open for recruitment.

It seems that the SCIB1 trial has had a major protocol overhaul. It has a new title, the number of patients has been increased to 44 and the treatment period has been extended to 85 weeks with follow-up for up to 2 years. Makes complete sense and gives SCIB1 a much better chance of proving its worth by increasing the power of the study. As it's an open label trial, there will no doubt be regular clinical updates along the way.

Interesting study published this week - 'Combining spike- and nucleocapsid-based vaccines improves distal control of SARS-CoV-2'.

The authors were looking at whether existing vaccines offered protection against involvement of distal sites such as the central nervous system in the event of breakthrough infections, their findings very much add to the evidence supporting Scancell's approach of targeting the Nucleocapsid as well as the Spike protein:-

'We show that after 72 hr of challenge, a spike-based vaccine confers acute protection in lung, but not in brain. However, combining a spike-based vaccine with a nucleocapsid-based vaccine confers acute protection in both lung and brain. These findings suggest that nucleocapsid-specific immunity can improve the distal control of SARS-CoV-2, suggesting the inclusion of nucleocapsid in next-generation COVID-19 vaccines.'

Moditope is a completely different kettle of fish to Covidity when it comes to delivery method. It's a peptide vaccine, delivered with an adjuvant (Isa's Amplivant) to target APCs and boost the immune response. The plan is to administer it with a simple injection - it won't need electroporation or PharmaJet to boost the response.

For the forthcoming trials, SCOV1/2 won't be using the peptide delivery method which I understand is still in development by the UoN. So they'll be using the PharmaJet device only. I believe in studies PharmaJet has produced as good, if not better cellular uptake of DNA vaccines than electroporation although I don't think it's ever directly been compared to Trigrid.

Crumbs the only reason SCIB1 patients needed two shots per visit was that the Trigrid device couldn't deliver 8mg in one go. With doses of 0.2mg or 1mg for Covidity , that shouldn't be an issue.

Finally, worth having a look at these children being given their measles vax with the PharmaJet device - not a single tear, not even a flinch. Looks pretty impressive.

WTP - not sure we can assume PharmaJet will be used for SCIB1. The requirements are different and it may be the solutions are too.

It's absolute nonsense to describe Redmile's dealings in Redx as predatory. For the sake of clarity:-

1) Redmile saved Redx from being forced to sell their IP at a vastly underpriced valuation and provided the finance to allow them to continue to develop their pipeline. Since their intervention, Redx have signed licensing deals with AstraZeneca and Jazz Pharmaceuticals
2) The ONLY reason Redmile made an offer for Redx was because a major shareholder who owned nearly 40% of the company wanted an exit. Redmile stepped in and acquired his stock which triggered rule 9 of the Takeover Code
3) At one point they owned 92% of the company - they now own 79%
4) Redx remains listed on AIM with no sign of any intention to delist

Far from being predatory, Redmile have been supportive and provided the finance and environment to allow Redx to flourish and they now sit with deals under their belt worth $1billion in future milestones and are pressing on with both preclinical and clinical development. I hope they will bring about the same transformation for Scancell.

Welcome to the SCLP bb and thanks for your observations.

It will be interesting to see if we ever discover more about Zakari. It may be that our new CEO decides against using the subsidiary at all, or she may decide to go ahead with the original strategy, whatever that may be. Scancell is first and foremost a vaccine company and you can see the logic of moving the IP for the glycan mAbs to a subsidiary. Avidimab isn't quite so straightforward because of course, it's now also an integral part of the ImmunoBody platform.

Will probably be teaching most here to suck eggs but the link below is to a really good infographic which explains the immune response to Covid vaccines.

Worth a quick read and while you're doing it, remember that's how it's meant to work in a perfect world. However, we know that your immune system doesn't recognise new variants quite so well so the response may not be quite so good. It's easy then to visualise another line of defence, a vaccine that gives all the protection in the infographic but also targets the nucleocapsid protein which is highly conserved between variants. So even if the spike response isn't quite as strong, you have those T cells recognising the N protein that will sweep in and destroy any infected cells and they'll do it for the current variant and the next and the next.
