A new paper from LD and her team was published a few days ago in the peer-reviewed journal Pathobiology.

Unfortunately the full paper is behind a paywall so you can only access the abstract but essentially they're looking at the role of PADI2 and PADI4 in colorectal cancer. Protein arginine deiminases (or PADIs) are the enzymes responsible for triggering citrullination and other post-translational modifications and the research seems to be looking at PADI2 and PADI4 expression in both the cytoplasm and nucleus of colorectal cancer cells as a prognostic factor.

I'm not sure whether the research is funded by Scancell or UoN but it's clearly linked to Moditope. Colorectal cancer isn't a target for Modi1, perhaps they're looking at it for Modi2?

There is a fundamental flaw in EB's post which suggests a lack of understanding of the setting for Parsortix. It doesn't have the potential to reduce medicine use at all, that's no what it's about and if anything it increases it. Just a couple of examples:-

1) Ensuring that patients are on the correct therapy for the genetic profile of their tumour at any given point in time and that changes can be spotted quickly and treatments adjusted accordingly. Currently, when patients are on a targeted therapy, the first indication that their tumour has stopped expressing that particular target will be when their cancer has spread and that's too late.

2) Monitoring of patients who have been successfully treated and are currently clear of cancer by spotting any relapse early and also potentially identifying dormant cancer cells lying in wait in the bone marrow. Rather than living in dread of their cancer returning, patients would have the reassurance of regular monitoring via a simple blood test and if the worst happens, treatment can be started at a much earlier stage.

So Parsortix isn't about reducing drug use, it's making sure the right drugs are used on the right patients at the right time.

Sorry I meant to add that although the primary completion date is April, it's an open label trial and it's likely we'll get updates along the way and that the SCOV1 results will be released when available.

I know it's frustrating but ethics committee approval has only recently been received and if you think about the logisitcs of physically starting the trial, ie. shipping over the vax, the PharmaJet devices, training staff etc. then it really isn't long at all.

Yep it's not the cash, it's confirmation that JZP815 continues to be developed. In terms of value changing news, perhaps a more appropriate term is value confirming news. Sanofi prepared to pay $1.9 billion to get their hands on Kadmon's ROCK2 inhibitor showing firstly it's a product big pharma want and secondly they're prepared to pay top dollar to get it. The only other ROCK2 inhibitor in the world in clinical development is REDX's.

Completely agree about your point on liquidity though.

I'm not sure this rise is driven purely by the ESMO presentation - take a look at the charts.

It started with the milestone payment from Jazz on 2nd September and then gained momentum with the article and buy tip from The Sunday Times on 5th September. Then we had the announcement of Sanofi's acquisition of Kadmon and their ROCK2 inhibitor on the 8th September. So a quck succession of events all highlighting the potential of Redx and the Sanofi acquisition in particular will not have gone unnoticed by bio investors.

Well they have that option of course and only AMYT can decide which is worth more - the sale value of the voucher now or saving it and using it to shorten the review period on the next product submitted for FDA approval. However, look at the pipeline, it's going to be a long while before they're sumbitting another NDA to the FDA on any of the other products and I suspect $100m now to fund the clinical development of that pipeline might be more useful.

Also, don't forget, Filsuvez was granted priority review by the FDA in its own right, no need to use a voucher even if they had one and given the field they're operating in, the same may well apply to future applications.

We were told at the end of last year that they were working on manufacture scale-up and preparing the supply chain in anticipation of approval. Apparently the product is now sitting at a speciality pharmacy, in pre-filled tubes ready for packaging and distribution. I'm not sure they've announced their plans in relation to global distribution yet.

As for AP103, it's still in preclinical development and has shown some very encouraging results producing similar collagen levels to viral gene therapies. The phase I/IIa trial is due to start in H2 next year.

It's only a few short weeks until the FDA decision on Filsuvez and the EMA's CHMP decision follows shortly after that.

There is currently no FDA approved treatment specifically for EB, if approved Filsuvez would be the first and will have orphan drug exclusivity in Europe, the USA and Japan. Worth also remembering that FDA approval would also bring a priority review voucher which Amryt can sell on to the highest bidder. The going rate for PVs seems to be on the up again, perhaps a knock-on effect of Covid as companies look to overcome potential delays at the FDA. Last month Prometic Biotherapuetics sold their voucher for $105m.

Not only would FDA approval of Filzuvez be amazing news for EB sufferers, it would also be a major inflection point here with the added bonus of an instant windfall of potentially around $100m. So it wouldn't be at all surprising to see investors starting to take positions now.

Jelenko, Parsortix can't capture ctDNA but it can use the same tube of blood to isolate CTCs, perhaps that's what you were thinking of. This from Angle's website:-

'If you also have an interest in ctDNA – that’s great. We can obtain ctDNA and CTCs from the same blood sample. If you are already doing ctDNA analysis – contact ANGLE and don’t waste the circulating tumor cells!'
