I have just come across an article with regards to 737.

An excerpt from below article dated in a journal 19th July 2024.

I have not seen this detailed information before.

'At the time of study initiation, SRA737 was the only orally bioavailable CHK1-specific inhibitor in development. Prexasertib, a CHK1/2 inhibitor, is administered via I.V. and has high hematologic toxicities, which together limit its clinical utility as a monotherapy and in combination with novel agents. Other CHK1is, such as MK-8776 and AZD7762, were no longer in clinical development due to PK limitations and concerns of toxicities. Ultimately SRA737 was selected for these preclinical experiments as it represented the current best candidate for future clinical development as a monotherapy and in combination with other therapies.'

Note that acrivon has a fast track designation in areas of high unmet need, despite, intravenous means of administration and high hematologic toxicities here!

I would hazard a guess here that Sierra Onology had indeed carried out considerable preclinical work in combination therapy.

Efficacy pretty much guaranteed by dual inhibition, should out perform Deucravacitinib with no problem.

Ideally we need crack Systemic Lupus Erythematosis.

Psoratic Arthritis also looks promising and again here early inhibitors approved for use in this indication have adverse effects and severe adverse effects and even major adverse cardiac

events.

We will be pursuing this in toxicology trial and having very little Jak2 should not pose any problems at all.

Jak 2 content should prove to be of slight benefit with Tyk2 Jak2 and Jak1 Jak2 Heterodimerisation. That my understanding..

The objective is a PASI score of over 75% over 12 weeks.

Long term toxicology ie safety been mentioned here before.

Interested parties been mentioned before.

There is no concrete factual news or else an RNS would be needed.

No insider information released and we have attracted a plethora of investment.

What wording has not been used points very heavily developments going on in the background.

Subject to the longer term toxicology studies that will give a good indication as to the safety profile will correlate to the value of SDC1801.

Many Indications available and a pharma will want on as many as possible.

Rapidly accelerate development into phase 2 whereby a pharma can cherry pick the Indications.

We are not at the end of phase2 just the beginning.

Hence l would suggest modest up front payment 50 to 100 million dollars and considerable milestone payments on phase 2 success and respective indication payments plus a further milestone at end of phase3.

Total circa 1 to 1.5 billion.

With greater efficacy than Deucravacitinib and similar safety profile maybe considerably more.

Don't forget the patent protection and the added benefit of capsule formulation!

Sareum still have not defined as to who requires these further studies.

It is true to say after being carried out this data,will be required to be included in Phase 2 application.

When do Sareum intend to start these investigations?

Strange that money is raised after Sareum have planned for these toxicology studies to go ahead.

So it must have been known 2 to 3 months ago.

In this time we have not continued into 1b but chosen to progress a path to phase 2.

On success it makes us an in phase 2 clinical trial asset. You can then go into any indication you deem fit.

May well prove a tad quicker going the phase2 trial route. Phase 1b over a few weeks does not contribute a great deal to long term toxicology data.
