Why would Sareum divulge sensitive info such as this?
2 things here are what is the definition of high dosages? Secondly are the therapeutic doses anywhere near the high dosages to be administered in trials.
At 30 times treatment dose in preclinical no indication of any adverse effects or toxicity were observed.

Earlier material released by Sareum put dosing levels at a meagre 12.5 mg whole body to 10 mg per kilo up to a maximum of 1000 mg total body.

Impressive when compared to Baricitinib a JAk1/JAK2 inhibitor at a treatment dose of 4mg per day and then due to resultant side effects and black box warnings have the dose reduced to 2mg per day. This is whole body dose.

Similar profile to Bari maybe, but that is where the similarity ends. By the very nature of toxicity alone we are streets ahead.

Many pharmas have turned blue today Avacta, Physiomics, Novacyt to take 3. My feeling is that traders have moved to oil and commodities. Brent crude has dropped from 130 dollars a barrel to around 98 dollars. We are not covid 19 reliant, at the same time have proven great results in investigations.
I would hazard a guess that traders will gradually inch back here.
On good toxicity results along with the indications that we intend treating at least a 30% rise, which of course is way below where we should be. My opinion old money 3.8 to 4.2p 190 to 210 new.
We have funding for stage 1 trials in 1801. Should be no problem along with satisfactory tox results for planned stage 1 human trial.

Look at around 60 day period for granting of stage 1 trial which should be a formality. Around June/ july announcement of commencement of stage 1 trials as long as we have completion of satisfactory amount of capsule with high purity formulation contained within.

Each of these stages reach a milestone which should then reflect in an increase in market cap.
What we do not know is if and how much and at what stage Sareum intend to raise further funds if at all. Infuriates most of us here i think. I would certainly not rule out a partnership, but we must steer clear of a Sierra oncology type license, whereby a company we deal with cannot guarantee 100% financial commitment due to other pipeline drugs taking priority.

We are one quarter down since consolidation and now to balance we need go up one third.

At consolidation we were at an approx 50% ie one half down since Peel .unt came aboard. We now would need a further 100% rise to restore market cap to these levels.

Is it achievable?
Yes but can take time and a few blips.

Can we go further?
Yes. looking at circa 400 to 500 million dollars towards end of stage 1. 12 to 15p old money 600 to 750p per share.

Based the 400 million up front on Nivolumab which i believe from memory was licensed at an under priced 400 million circa 2018 2019. SRA737 should complement well.

Ok on its own in some indications, much more effective when used in combo but as yet not fully realised. 737, gemcitabine with an immuno checkpoint inhibitor ( there are several around) i believe will be outstanding. Sierra know what they have. It may prove more commercially viable than Momo they said. They know what they have.

A waiting game.

SDC1801. Toxicology results. They will kill 2 birds i feel with one stone, as independent ie non biased assessment results will form basis of CTA.
Bare in mind and we already have people in place to organise trials.

We dont know long term, but just maybe at some stage used in combo with SDC1802. In reality not sure if that would happen so just little more than an idea. As a guess in pancreatic which has a very poor prognosis at present.

Getting it 100% correct is paramount. In saying all that I reckon that we will receive half a dozen lines. Most info on report will be classified to some extent as need keep info from competitors.

ndr50. As a mono therapy SRA737 was not overly effective in high grade serious ovarian cancer. it did perform very well in colorectal and anal cancers.
Originally the first dosed person in the USA would carry a milestone payment of circa 7 million of which we would been eligible for a 27.5% payment. People have been dosed with SRA 737 outside of the USA. This figure was reduced to 2 million dollars and hence we would be eligible for an equivalent 27.5% circa 500k sterling. Sierra looking at further options to fund 737. They had cut investments to 737 for their Momo.
As it stands now, milestone payments to Sareum at 27.5% are around 70 million pounds of a an approx 300 million dollars total milestone payments through to commercialisation.

From memory a 2nd milestone payment would be payable on stage 2 clinical studies in human trials. on reaching this stage the 1st milestone payment would become payable. Not sure if this stage 2 trial would have to be administered in the US. Nothing I have looked at confirms if this is the case or not.

All in all very promising results regarding 737 as a therapy although significant delays have been created by the lack of investment by Sierra into the further development of 737 both in mono and combination therapies. They have carried out some great work give them their due.

737 has indicated so far that it should yield extremely encouraging results when used with Wee1, Gemcitibine, and especially Gemcitibine with an immuno checkpoint inhibitor in later stage human trials.

Pleased that 89 of 107 patients have survived the 3 years. Would be interesting to know how long after the trials ended that those that benefitted from 737 continued further treatment and how their conditions are today.

Basically 737 causes cell cycle arrest through P53 checkpoint and prevent cancer reaching metastase due to the DNA damage repair pathway.

Very positive results. Advanced cancer. Continuation of treatment after trial which many received as they benefitted from. This was a mono therapy from memory. It was also used in combo with Gemcitibine. There were a teeny bit on Sierra website a short while ago. SRA 737 plus Gemcitibine and immunotherapy checkpoint inhibitor. As near as could make from it the mutts nuts. Not available on their slide any more.

Lethally irradiated mice with 20% survival after 30 days. IL-6 and Il-3 90% survival rate after 30 days.
SDC 1802 in combo with other therapies or even in combo with 737. Not saying it will happen as not know if compatible.
SRA 737 well worth keeping an eye on especially in combo gemcitibine and immuno checkpoint inhibitors. I did read on Sierra website a few weeks ago but no longer available.

SRA 737 and DNA damage repair mechanism that profusely replicates cell production in an effort to repair a tumour. Like most injuries the body makes many cells to repair damage. Unfortunately our bodies cant recognise beyond a certain stage ( ie single cells) that the cancer is not what our body wants.
As you already know cancer cells do not suffer from apoptosis, which is a programmed cell death. Our cells die and are replaced all the time. It is a vital part of ageing. Chemo and radiotherapy can destroy these non apoptosis cells. The downside has been DDR resulting in metastase cancer which 737, may on its own and certainly when used in combo can conquer. Not all but a minimum of 50% of tumours are reliant on P53 checkpoint.

Looks like a world beater. one of 3haha.

Forgot to add Citizen 79, great posts from you. I do confess I have difficulty in understanding some of the nomenclature in some reports. I have gone as far as I can to envisage toxicity problems with our compounds. I went down the IL-15 route as inhibiting this can result in severe infection. Some sub units are shared with IL-2. We do not inhibit Il-15 completely.

It is also the ratio and degree of inhibition of certain interleukins that look to be of importance. Sareum have clearly learned of the pitfalls and lack of selectivity with early stage kinas inhibitors. In depth results I feel to be kept a closely guarded secret.

Both Tyk2 and Jak2 signal granulocyte colony stimulating factor ( effectively the white cells that are produced in bone marrow)
However, it is generally accepted now that TYk2 is far more selective than Jak2. We are a later stage generation kinase inhibitor. This may have been the reason as to why Tim did not go down the JAK2 route much preferring the better safety profile associated with Tyk2.

I dare say the Sareum kinase inhibitor library paid a very big part in this.

To ensure top notch, capsule as the form of oral route, protection by patented 17 year crystallisation of compound formulations which can achieve 100% purity. This should give us the edge with regards LADMET and provide value with this considerable patent protection level.
Il-6 is topic worthy of discussion on its own.

Correct me if I am wrong citizen but Baricitinib is a Jak1 Jak2 inhibitor and thus not as selective as Tyk2.
Use of steroids maybe more effective than the use of baricitinib on its own. The world will still require an effective treatment I feel in the advent of any similar virus causing acute respiratory distress syndrome outbreak, irrespective of Covid 19.
