Abstract
Non-alcoholic fatty liver disease (NAFLD), encompassing fatty liver and its progression into nonalcoholic steatohepatitis (NASH), fibrosis, cirrhosis, and hepatocellular carcinoma (HCC), is one of the rapidly rising health concerns worldwide. SIRT6 is an essential nuclear sirtuin that regulates numerous pathological processes including insulin resistance and inflammation, and recently it has been implicated in the amelioration of NAFLD progression. SIRT6 overexpression protects from formation of fibrotic lesions. However, the underlying molecular mechanisms are not fully delineated. Moreover, new allelic variants of SIRT6 (N308K/A313S) were recently associated with the longevity in Ashkenazi Jews by improving genome maintenance and DNA repair, suppressing transposons and killing cancer cells. Whether these new SIRT6 variants play different or enhanced roles in liver diseases is currently unknown. In this study, we aimed to clarify how these new centenarian-associated SIRT6 genetic variants affect liver metabolism and associated diseases. We present evidence that overexpression of centenarian-associated SIRT6 variants dramatically altered the metabolomic and secretomic profiles of unchallenged immortalized human hepatocytes (IHH). Most amino acids were increased in the SIRT6 N308K/A313S overexpressing IHH when compared to IHH transfected with the SIRT6 wild-type sequence. Several unsaturated fatty acids and glycerophospholipids were increased, and ceramide tended to be decreased upon SIRT6 N308K/A313S overexpression. Furthermore, we found that overexpression of SIRT6 N308K/A313S in a 3D hepatic spheroid model formed by the co-culture of human immortalized hepatocytes (IHH) and hepatic stellate cells (LX2) inhibited collagen deposition and fibrotic gene expression in absence of metabolic or dietary challenges. Hence, our findings suggest that novel longevity associated SIRT6 N308K/A313S variants could favor the prevention of NASH by altering hepatocyte proteome and lipidome.

Genflow Biosciences PLC (LSE:GENF), Europe's first listed longevity company, said it made significant progress during a 'busy' first quarter of 2023.
In the update, it also said it was in a secure financial position with enough cash and grant funding to last until March 2025.
Investors were also told it continued to make headway in two groundbreaking longevity programs: Non-alcoholic steatohepatitis (NASH) and a form of premature ageing called Werner Syndrome.
Genflow said it had reached several milestones, including the completion of a detailed application dossier for the chemistry, manufacturing, and controls (CMC) of its NASH treatment.
It will present the dossier to the Belgian regulatory authorities in early June, paving the way for accelerated clinical trials as there is currently no known medical treatment for NASH.
In collaboration with Dr Manlio Vinciguerra at the University of Liverpool, the drug developer said it has achieved a deeper understanding of the biochemical changes in NASH treatment using its centenarian SIRT6.
The research findings have been published in a peer-reviewed journal, and Genflow is exploring additional intellectual property (IP) opportunities.
It has also initiated in-vivo evaluations of its centenarian SIRT6 gene therapy in four different NASH mice models in partnership with the University of Liverpool, the University of Rochester, and the clinical research group, Physiogenex.
The studies, which have analysed over 700 mice, aim to understand the efficacy and safety of the drug candidate in animal models with NASH. The results will be crucial for seeking authorisation for clinical trials in humans.
Additionally, the company has conducted targeted biodistribution studies of its SIRT6-AAVs with partners IVEX in Estonia and Articles in Belgium. The data will form a significant part of its presentation to the regulatory authorities.
After rigorous in-house studies, Genflow has achieved consistent and satisfactory drug candidate delivery to targeted human cells, marking a crucial first step in determining the most effective dosage for cSIRT6 gene therapy in human trials.
Genflow's partnership with Exogenus Therapeutics in Portugal has led to a promising opportunity for a new patent application related to the encapsulation of AAVs into exosomes. If successful, the patent-protected delivery method could have significant positive implications for gene therapy.
Furthermore, the company has expanded its intellectual property portfolio through a provisional patent application focusing on editing its SIRT6 gene, which plays a role in longevity and age-related diseases.

I didn’t know that you were also working on gene therapies for SIRT6 activation. Do you have any preliminary results or insights?
Like I said, this is ongoing, so I don’t have a lot of results, but we’re exploring this using different models and targeting different organs. Hopefully, we’ll have the results soon, but just based on genetic models of SIRT6 overexpression, we expect to see a rejuvenating effect, because if we simply overexpress SIRT6, mice live longer.
It’s not an easily translatable system, but if we can deliver SIRT6 and even express it transiently, we hope to see improvement. I can share one type of results, which is in cell culture, not in animals. When we overexpress SIRT6 briefly in cultured human cells, we see epigenetic age going down.

Fresh from this latest success, the supermajor has now started drilling a fifth exploration probe, targeting a major prospect some distance north of its quartet of discoveries in Block 2913A.

For decades, oil and gas exploration in South Africa had yielded what our report describes as “scattered and sporadic success.”
But all of that changed in 2019 when Total (now TotalEnergies) announced its Brulpadda discovery, followed by the Luiperd discovery one year later. Brulpadda Field is estimated to hold at least 275 million barrels of oil equivalent (MMboe), and Luiperd’s estimated reserves are approximately 340 MMboe, with 70% gas each in both discoveries.
These numbers are huge and among the largest made in our continent in their respective years.
“Brulpadda accounted for 10% of the overall discovered volumes in Africa in 2019, and Luiperd accounted for a whopping 80% of the overall volumes discovered in 2020,” our report notes.
When the Brulpadda and Luiperd fields come online, their cumulative average output is estimated to be around 35,000 barrels per day of liquids and about 100,000 barrels of oil equivalent per day (boepd) of natural gas.

A marginal field ( in shallow water ) discovered in the 1980's sounds just like Tower's prospect, and with Perenco now producing 3,800 barrels a day, Njom-3 must be on their radar. Perenco used to hold the licence but relinquished a few years ago because of their work load elsewhere in the Rio del Rey basin, which allowed Tower to obtain the licence and with Perenco now fully committed to the basin following the new 20 year contract, I can't think of any reason why Perenco wouldn't be interested. Perenco have their own jack up drilling rigs in the basin as well as the rest of the infrastructure to allow oil and gas production a rapid route to market...confirmation news of Tower's 12 month license extension could be the catalyst. It would now make perfect sense for Perenco to be turning their attention to Tower's NJOM-3 as part of their ambitious program in developing and seizing new opportunities in the Rio del Rey oil producing Basin. Gla :-)

The marginal field, discovered by a previous operator in the 1980s, is in 30 m to 50 m water depth.
Following an eight-month program—which according to the company involved innovations in terms of well architecture, pipelay and the type of platforms installed—production is currently above 3,800 bbl/d.
Earlier this month, Perenco signed a new 20-year production sharing contract with Société Nationale des Hydrocarbures for continued development of the Rio del Rey Basin, also offshore Cameroon.

Offshore drilling rig owner Northern Ocean said Wednesday that its Deepsea Mira semi-submersible drilling rig had kicked off its drilling contract with TotalEnergies in Namibia, where the French oil major last year made a giant oil discovery with its Venus-1x well.
Earlier this week, TotalEnergies was named the upstream industry's most admired explorer and has received the Discovery of the Year award in Wood Mackenzie's annual Exploration Survey. TotalEnergies also received the Discovery of the Year award for Venus offshore Namibia, in partnership with QatarEnergy, Impact Oil & Gas, and NAMCOR.
Maggy Shino, Petroleum Commissioner at Namibia’s Ministry of Mines and Energy, said this week that, in Namibia, “we have an ongoing drilling campaign with three rigs currently busy drilling appraisal and exploration wells. We are expecting two more wells to be drilled before the end of 2023 in the deep waters.”
According to African Energy Chamber, Shino said that Namibia country is seeing a rise in seismic surveys, and by the end of the year, the government is planning to announcement a series of drilling projects that will take place during 2024.

Repsol will plug and abandon the Welwitschia-1A well offshore Namibia, Tower Resources, a partner in the well, has said.

The Rowan Renaissance drillship started drilling operations on PEL0010 on 23 April 2014 and the Welwitschia-1A well spud on 1 May 2014. Repsol Exploration (Namibia) (Pty) Limited (“Repsol”), the Operator of Namibia PEL0010 (Repsol 44% working interest, Tower 30% working interest), has confirmed that the firm commitment well reached total depth of 2,454 metres TVDRT (Total Vertical Depth Below Rotary Table). Logging evaluations indicate that the Palaeocene, Maastrichtian and upper Campanian section reservoirs were less well-developed than prognosed and no hydrocarbons were encountered.
Drilling has been behind schedule initially owing to late rig-delivery and operational issues during drilling and logging, including the onset of winter weather conditions. Current expectations from the Operator are that costs will now be around 10% in excess of the $91 million gross firm well budget.
The estimated cost of continuing the current well to test the deeper targets, including the Albian, now appears to be as much as a further $40 million gross.
“Despite our interest in the deeper targets, with their large potential resources, this raises the question whether it may be better to wait for the full analysis of the current well before deciding whether and where to drill a second well and test the deeper targets with the benefit of that further information. As a result, the Partners have agreed not to drill further at this time and to evaluate the information and its implications for the Block. The well is being plugged and abandoned,” Tower said in a statement.

Tower's sp shot up from 3p to 15p following the "Glowing in the Dark" seismic update before they drilled Welwitchia-1 with their new JV partner Repsol, who made a complete dogs dinner of the drill, with the wellhead sinking into the sea bed, whereby they had to re-spud the well in a different position, costs began to escalate out of control and Repsol pulled the plug and never drilled to the source rock depths, the same carbonates and geology where Shell and TotalEnergies are having world class discoveries in the Orange Basin. The Aim market is very much different to the heady days back then and the market wants solid material news, although the Namibian Technical UPdate can only increase interest from any potential JV partner and no doubt along with the market is awaiting solid seismic analysis....in the meantime the greenlight from SA government for TotalEnergies World class gas discoveries which straddles Tower's South African offshore acerage will be very well received along with any funding news for Njom-3 in Cameroon, news is imminent on both imho...Gl :-)
