There's been a raft of commercial deals over the last year, many worth $100m's , involving early stage bio's focussed, on STAT3 or NRF2 and also SHP2 , all of which SFX-01 appears to have a potent therapeutic effect......yet Evgen's market cap is still only £15m with a phase 2/3 ARDS/ Covid patient trial underway, and awaiting data for a phase2b breast cancer trial to commence, where phase 2 data was outstanding and life changing for some patients, as well as disclosure on a collaborator on some of the best Solid Tumour preclinical data ever seen for a hard to treat cancer , as well as the rest of the promising pipeline focussed on multiple disease targets, not to mention the licensing deal with Jim Mellon worth $10.5m and royalties thereafter......sooner or later a Major will show their hand, or positive trial data and progress will propel Evgen's sp to fair market value and beyond...Gla ;-)

Scopus BioPharma Stock Price Surged Over 300% In Public Debut

The stock price of Scopus BioPharma (NASDAQ: SCPS) increased from $8.17 at 11 AM ET on December 16 to $33.96 on December 17 at 4 PM ET. Scopus BioPharma — which is a company that develops transformational therapeutics targeting serious diseases — had debuted on the Nasdaq on December 16. The company’s lead program is a targeted immuno-oncology gene therapy for the treatment of multiple cancers.

Back in June, Scopus BioPharma said it is licensing novel immuno-oncology gene therapy drug from the City of Hope. The City of Hope is a world-renowned independent research and treatment center for cancer, diabetes, and other life-threatening diseases near Los Angeles, California.

And the licensed gene therapy drug CpG-STAT3siRNA (or the STAT3 inhibitor) is a highly-selective and targeted therapy that silences the activity of the STAT3 gene by way of RNA interference while stimulating the TLR9 receptors to activate the body’s immune defense to recognize and kill cancer cells.

In pre-clinical testing, the STAT3 inhibitor successfully reduced growth and metastasis of various pre-clinical tumor models, including melanoma, colon and bladder cancers, as well as leukemia and lymphoma. A first-in-human Phase 1 clinical trial in B-cell lymphoma patients was scheduled to commence in H2 2020 at City of Hope.

Reductive stress in neuroblastoma cells aggregates protein and impairs neurogenesis

In the current study, the researchers used sulforaphane to establish RS in proliferating and differentiating Neuro 2a neuroblastoma cells grown in culture. Sulforaphane activates Nrf2/ARE signaling, leading to antioxidant augmentation. Specifically, they found that sulforaphane-mediated Nrf2 activation diminished reactive oxygen species in a dose-dependent manner leading to RS. The resulting RS abrogated oxidant signaling and impaired endoplasmic reticulum function, which promoted protein aggregation and proteotoxicity, and impaired neurogenesis. This included elevated Tau and a-synuclein and their co-localization with other protein aggregates in the cells.

Namakkal-Soorappan says they were also surprised to see that acute RS impaired neurogenesis, as measured by reduced neurite outgrowth and length, and that maintaining the cells in sustained RS conditions for five consecutive generations dramatically reduced differentiation and prevented the formation of axons.

This impairment of neurogenesis occurs through activation of the pathogenic GSK3ß/Tau cascade to promote phosphorylation of Tau and create proteotoxicity.

Intriguingly, there have been reports of increased levels of enzymes that can promote RS, both in the brains of Alzheimer’s patients and in the post-mortem brains of Alzheimer’s and Parkinson’s patients. Also, attempts to promote neurogenesis in neurodegenerative diseases using small molecule antioxidants have had poor outcomes.

Sorry SuperS, that's wishful thinking and hoping /expecting Piers to comment in such a way....if SFX-01 does indeed prove to be effective, otherwise we'd be sailing past £1 plus and more if had interviewed someone with a 90 year old Granny, who had made a miraculously recovery from Covid-19 on SFX-01...so much so that she was going to be running the London Marathon. ;-)))

News on the Solid Tumour collaboration could also be close, along with an update on progress towards the breast cancer trial and the rest of the pipeline......the pick up in buying volume, especially the 3 x 300k buys one after the other bodes very well imho...................Every chance of a commercial JV in the pipeline and every chance of a sudden and significant re-rate to reflect fair value on Evgen's undoubted transformational potential.
