RE: New patent application28 Aug 2025 12:15
'Scaffold proteins' patents have been filed by Avacta since 2017.
This is the World patent from the patent family: https://patents.google.com/patent/WO2019008335A1/en
"Summary of the Invention
"The inventors studied the Stefin A scaffold protein with aims of improving its properties such as its thermal stability. Building on their insights into the protein structure and function, they designed a range of changes and substitutions to improve those properties. This led to various surprising results including unpredicted anomalies where for example it might have been expected for the protein to behave in a certain manner, but for which the objective findings were at odds with predictions (for example the Q.42D/E anomaly discussed in detail below). The result of this substantial research and intellectual effort is a comprehensive teaching of how to alter the thermal stability of the Stefin A protein and how those changes in thermal stability deliver benefits to its use as a scaffold protein.
"Thus the invention relates to a polypeptide, such as an Affimer polypeptide, comprising an amino acid sequence having at least 80% identity to amino acid residues 1 to 11, 13 to 15, 17 to19,21to25,27 to28,35 to37,39,41,43 to44,46to47,49 to50,52 to53,55 to58,63 to 64, 66, 68 to 82, 84 to 85, and 87 to 98 of SEQ ID NO: 1;"

This suggests that the invention is relevant to the application of Affimers, e.g. for AffyXell, Gen 3 PDCs, other Tx applications, and diagnostics.
