The answer is very simple. FAP cleaves the peptidic (-CO-NH-) bond between the D-Ala-L-Pro (which is the core of the invention) and the drug. That peptidic bond is created by linking the acid end (-CO2H) of proline to an amino (H2NH-) group on the drug. If you look at any of the diagrams of AVA6000 being split, you'll see what this looks like.

So, in short, any drug with an available amino group can be pre|CISIONed. Bortezomib doesn't, so a similar structure retaining that part of bortezomib that is responsible for its pharmacological activity but with a free amino group was required.

Just a reminder that, as per the patents that are the IP for pre|CISION, this platform can be used to target drugs to ANY tissue in which FAP has been upregulated, not just epithelial cell cancer tumours. Opportunities for prodrugs targetting inflammation, fibrosis and wound healing have yet to be exploited.
