Hi Celtic. Ultimately, every expert in the field will have a slightly different opinion on this but he is backing their tyk2 as it's safer than having Jak1 and made incredibly well. He does concede though that having jak 1 is more effective. So if we can nail the safety aspects, IE what's going on right now, then it bodes very well indeed.

When push comes to shove, the squeaky chair, the awful PR, lack of news are really immaterial. If John and Tim have nailed the science, we're onto a winner. Need to pass the safety first though.

Thanks for all the replies. I'd be very wary of these buyouts with a sum up front and then staggered milestones. We all know from SO that strategies can change and milestones are no guarantee of payment.

Can I ask a question about how these sorts of deals work in practice?

Say we are bought for £500m upfront and up to £1bn in milestones. How does that work? How would they pay the subsequent money for milestones if the company doesn't exist more? Would our shares still technically be active? So it would officially be a buyout as SAR would have 0 control over operations but the shares still bought and sold? I can't quite understand how we are bought out but as shareholders be owed money potentially 2 years down the line...

I think this a great idea and although I'm sure HBD will be able to utilise his clinical trial hunting skills, if Sierra changed the name to SRA737, I wouldn't be surprised if the compound is given a new name.

Therefore it may be better to search for trials with gemcitabine etc. i.e. the other compounds..

Or more simply

So this is the timeline for this section:

Day 1 (pre-dose, 8 hours post-dose)

Day 2 (24 hours post-dose)

Day 4 (48 hours post-dose)

Day 6 (120 hours post-dose)

Day 14 (on presentation to the clinic)

Day 15 (pre second-dose, 8 hours post second-dose)

Day 16 (24 hours post-second dose)

Day 17 (48 hours post-second dose)

Day 20 (120 hours post-second dose)

Day 29 (final follow-up visit)

But then it also says:

Each participant will be in the study for approximately 9 weeks (Screening Visit to Follow-up Visit) and will receive 2 doses of SDC-1801.

So maybe 3-4 more weeks until completion, bearing in mind potential disruption over the festive period. I would like to see completion RNS by the end of January at the latest. Hopefully we will have 737 news before then. Could be an exciting Q1 for SAR...

The food dosing part of the trial started just before 9th November, according to the RNS on that day.

Does anyone have any idea how long that would part of the trial was supposed to have taken?

Also, is this the last part of the trial or is there one more?

I would like to think the BOD, after the flak they received at the AGM, now recognise the important of timely reporting of milestones, if only to ally fears from us shareholders and boost confidence all is still going well.

(I know there is a page that has been posted which outlines the full structure of the trial, but for the life of me I can't find it anywhere.)

3 years ago there was a strategy. April 2020, we were presented with a lovely PowerPoint that outlined the buy back option. The strategy was to define a resource and sell it to AA within 2-3 years.

We were sold the dream. RNSs every 3 days telling us we'd hit the mother load and everything was hunk dory.

3 years down the line it's evident that not only Colin missold us the dream, but also he has no strategy for ANY shareholder value. Quite frankly (to use his favourite phrase), there is no obvious plan for any short to medium return of shareholder value. I do hope conversations are happening behind the scenes for a buyout by XYZ for 50million because I would love my money back.

It wasn't long ago I considered Kwadoku completely mental. Turns out he's not far off - from the evidence we have before us (down 80% and ZERO news), Colin doesn't appear to care about the shareholders.

I look forward to being proven wrong. As I've said before, he needs to step up or he will be canned at the AGM 2024.

Fortunately (for us) Bezant and Xtract are poles apart. Their current market cap of 1.7m, is almost a tenth of ours and they have taken a placing for around half their MC. It really is dire over there. Their fate rests on the commission of one project in the Phillipines. It's a one basket company.

However, nothing explains Xtract's marketing being significantly less than Empress's, despite them having a fraction of the economic interest in Manica and having multiple other share price drivers (IE. Other distractions and things to market).

God knows how much they'd be shouting from the rooftops if this was their only project.

The higher it goes, the higher the profit margin. The higher the profit, the more income to Xtract. Currently, Xtract flow of income is inversely proportional to our CEO's interest in marketing us. Would be nice if he started working for his money, IE. 125k for a part time salary. Else he'll get the boot come AGM, summer 2024, should the current sp malaise continue or his disinterest/ lack of effort showcasing us to the wider market not pick up.

I note from the photo in the tweet that TLL-018 in phase 1 was only tested up to 120mg in healthy volunteers.

Then, in the phase 2 trial, it was only tested up to 30mg however "TLL-018 20 and 30mg demonstrated superior efficacy over tofacitinib in RA patients, suggesting that inhibition of TYK2, in addition to JAK1, enhances efficacy."

It certainly bodes very well that we are testing up to 300mg while only 30mg of another TYK2/JAK1 seemed to have a positive impact.

What do you mean by: "report on each cohort"? How much information will we receive? I would argue it would be absolutely minimal information especially in terms of efficacy. We all know how conservative the BOD are both in terms of news output and in terms of information given DURING the trial. We are literally experiencing it right now. The share price isn't going anywhere and over the past 4 months in trial we have been getting the feeblest levels of information apart from tidbits stating next stage has commenced. This will only confirm safety requirements are being adhered to, and NOT efficacy and key objectives achieved.

I am as confident as the next person on here, but evidence from the current trial AND tempest therapeutics would suggest a) Share price doesn't move upwards whilst the trial is going on and b) We will not receive indication of success in efficacy from THIS BOD DURING the trial.

This suggests two things. It will probably take a little longer than expected. But when it does, it will go BOOM. (a la Tempest - 4000% in one day) I.e. Well worth waiting for. Unless we are bought out before that because big pharma would like to remove the 4000% increase bit...
