Efficacy - Dox as a drug has proven efficacy, but it’s inefficient and constrained by the amount the body can handle, as it’s not specific.

It’s the efficacy of AVA6000 we’re interested in, not Dox. You are confusing the two.

AVA can be a roaring success at P1a but have limited success all the way up to P3. Having biopsies that proven tumour efficacy of AVA’s delivery of free dox is what we need for deals and takeovers.

Howzaty - yes we do know how dox impacts the tumour, but DLT’s restrict its impact so we need to know how this prodrug improves efficacy. That’s the point of that discussion.

The poster is saying quite a bit more than that. We have an extended half-life which implies its stable in plasma. Note, longer isn’t always better here.

We’ve got clearance data and we know free dox (the toxic part) is very low in context. Therefore, the part that can damage the body is negligible.

The cytotoxic discussion is heavily implied by the poster. What’s not confirmed is how much dox is accumulating in the tumour. This is why the biopsy data is so important.

CO - Normal tissues have low and generally undetectable levels of FAP expression. However, FAP is overexpressed in many tumor tissues.

Have another look at the AVA poster and you’ll see that free dox is around 0.018% of the cleaved plasma concentration compared to AVA. And rats have a high background FAP level (AS in video).

We will see free dox in plasma, therefore exposed to healthy tissue. It’s inevitable. But view the levels in context of the original hand grenade that has gone off in the tumour.
