I agree. In fact I agree with most of what you say. But efficacy hasn’t been proven. That is the only thing I’m saying. Not that it won’t be proven. We have to wait for end of P2

It isn’t black and white. I’m not saying it doesn’t work. The most likely thing is that the results aren’t as predictable as had been expected and there is a lot more to be done on tuning the dosing regime.

That is good but what some are refusing to recognise is that there are still high risks:
1. Does it work well enough across a large number of patients
2. Will it generate enough profit to support development of the ensuing platform and pay us a decent dividend
3. Will BP buy it and any profits be watered down supporting the rest of their product range.
To me it’s still not worth losing my shirt over.

I’ve never argued that it hasn’t been released to the TME or that patients have died. What I am considering is the reasons why we might be where we are and one of those could be slower than expected cleaving in patients with certain characteristics
How does it compare to 1540mg/m2 ?

If MTD in mice was 2mg/kg then what are the equivalent doses in humans in C7?
Jeez this is painful. It doesn’t say how much other than therapeutic levels whatever that means. Presumable any amount no matter how small is therapeutic because we know the effect that dox has on tumours.
It’s all down to Al’s propensity for ambiguous phrases. And up to now it isn’t the most positive interpretation that has been how things have turned out.

But none of these give us any clue as to dox in the tumour and yes I know it was a safety trial which is what I’m trying to impress on the rampers.
Anyway hopefully with rich data reveal we will (soon) find out.

Ok I’ve rewatched the video. Nothing there precludes my concerns about excretion versus cleaving. It is great that safety profile allows more frequent dosing and better still that it keeps a higher concentration of AVA6000 in the blood for longer. Anyway we will see what is actually happening when arm 2 finishes whenever that is.
And my point about how the trial had to be extended being a surprise because the “results were far better than expected” still stands The rat trial results certainly predicted those results. And I can’t remember Avacta saying “better than expected “ I think they just said “unexpected “. But I am more than happy if someone finds that quote.
Finally I am invested and want it to work but I do take offence to the constant black and white “it works”.

AVA6000 safety is proven. Efficacy is not. It isn’t enough for it not to kill people. I think that there is a possibility that excretion rate exceeds cleaving rate for some patient characteristics hence 2 week dosing trial and stony silence on C7. IMO

Why wouldn’t we believe the timelines? Some posters keep telling us that because of the changed design of the trails, we are ahead of the target to get onto the market.
It is fantastic that SP is back up to 54p though I wonder how high it needs to get before profit taking steps in though I would only expect that to cause a small dip before carrying on up to find a stable value.

I don’t think that anyone on here is questioning the marvellous results (similar to rat trials) that Avacta have announced for 4 out of 40 patients. Neither the safety profile achieved. But 4 is not statistically significant.
Regarding AACR prepare to be disappointed. All the stuff you mentioned (which we desperately need to know) has not been promised by Avacta.
So stop ramping and wait
My point is that they would have to have a presence anyway but I’m not saying that there won’t be an excellent presentation. Just not very likely.
