Following a preliminary examination of a small group of clinically approved drugs, experimental novel compounds, and nutritional supplements, SFN was identified as a promising candidate to target the host cellular response, given that it is orally bioavailable, commercially available, and has limited side-effects 8,21.

We observed that SFN has dual antiviral and anti-inflammatory properties against coronaviruses. We determined that SFN has potent antiviral activity against HCoV-OC43 and multiple strains of SARS-CoV-2, with limited toxicity in cell culture.

The similar results observed between the coronaviruses evaluated suggest that SFN could have broad activity against coronaviruses, a feature that may prove invaluable as new strains of pathogenic coronaviruses enter the human population.

Moreover, synergistic antiviral activity was observed in vitro between SFN and remdesivir against both types of coronaviruses tested; comparable synergism in vivo would be advantageous in clinical scenarios where remdesivir is currently being used.

And finally, we need to continue pushing on treatments. In April this year, we belatedly launched our antiviral taskforce. We need to build on the success of finding Dexamethasone, to develop and stockpile a basket of other treatments to further decouple catching Covid from death (full disclosure, I have an investment in some treatment innovators). In particular, we should focus on broad-spectrum antivirals as these are likely to work on future variants.

The new Secretary of State needs to drive the completion of our vaccination programme and push hard to get greater take-up. But on its own that is unlikely to be sufficient. We need to design a much faster version of the next vaccination programme and supplement it with smart social distancing, testing and treatments. By doing that we can safely return to close to normal but, more importantly, be ready for what the virus next throws at us. And our experience to date shows that the virus is unlikely to allow us to win without some surprises. Sajid Javid needs to be ready.

All and sundry usually know when a placing is coming, very few will know If bad news or good news is coming from Prof Chalmers efficacy assessment, and the trading volume over the last few weeks doesn't suggests anyone knows imho, otherwise we would have seen lots of big sell trades and much higher volumes I reckon, more like the weak and worried getting twitchy at the time it's taking and derisking and selling up, which in turn encourages others to do the same...!!!??? KEAP1, a cysteine-based sensor and a drug target for the prevention and treatment of chronic disease

Abstract

Redox imbalance and persistent inflammation are the underlying causes of most chronic diseases. Mammalian cells have evolved elaborate mechanisms for restoring redox homeostasis and resolving acute inflammatory responses. One prominent mechanism is that of inducing the expression of antioxidant, anti-inflammatory and other cytoprotective proteins, while also suppressing the production of pro-inflammatory mediators, through the activation of transcription factor nuclear factor-erythroid 2 p45-related factor 2 (NRF2). At homeostatic conditions, NRF2 is a short-lived protein, which avidly binds to Kelch-like ECH-associated protein 1 (KEAP1). KEAP1 functions as (i) a substrate adaptor for a Cullin 3 (CUL3)-based E3 ubiquitin ligase that targets NRF2 for ubiquitination and proteasomal degradation, and (ii) a cysteine-based sensor for a myriad of physiological and pharmacological NRF2 activators. Here, we review the intricate molecular mechanisms by which KEAP1 senses electrophiles and oxidants. Chemical modification of specific cysteine sensors of KEAP1 results in loss of NRF2-repressor function and alterations in the expression of NRF2-target genes that encode large networks of diverse proteins, which collectively restore redox balance and resolve inflammation, thus ensuring a comprehensive cytoprotection. We focus on the cyclic cyanoenones, the most potent NRF2 activators, some of which are currently in clinical trials for various pathologies characterized by redox imbalance and inflammation.

Precision cancer and immune disorders focused Vividion Therapeutics has filed for a $100M IPO on the Nasdaq under the tickeR "VVID"

The per share price range and other financial details have yet to be disclosed.

J.P. Morgan, BofA Securities, and Jefferies are acting as joint book-runners.

Yesterday, Vividion received a milestone payment from Roche (OTCQX:RHHBY) for a collaboration on an oncology target. Last year, the two companies inked a deal that included a $135 upfront payment to Vividion for discovery and development of programs addressing a range of oncology and immunology targets.

Vividion's lead programs focus on transcription factor NRF2 for the potential treatment of NRF2 mutant cancers, as well as inflammatory diseases, such as irritable bowel disease and chronic kidney disease.

Perfectly poised.....and so fingers crossed for some stellar news....Gla ;-)

Yep, it would be nice to think that Prof Chalmers was allowing his researchers, Phd students and friends and families to fill their boots because of encoraging data, although personally i would want to see a significant increase in volume that might suggest that good news is leaking, to be certain....Watch this Space...all bodes well. Great post Compoundinterest, validation of SFX-01's efficacy for Covid/Ards will be a complete game changer and will propel Evgen towards global investor spotlight and focus and very likely add significant value and interest to the rest of the pipeline, whereby an early lucrative licensing deal or two could materialise. Beijing Jacobio Pharma (HK: 1167) has dosed the first two patients in a Phase I/IIa trial of its SHP2 inhibitor in patients with solid tumor cancers. One year ago, Jacobio out-licensed global rights for two small molecule SHP2 inhibitors to AbbVie (ABBV). AbbVie made a $45 million upfront payment to Jacobio, and the start of the trial triggers a $20 million milestone payment from AbbVie. The SHP2 inhibitor, JAB-3312, will be administered in combination with PD-1 antibody pembrolizumab and MEK inhibitor binimetinib.

Potential Transformational news expected anytime within the next 7 trading days, and I reckon Huw and co along with Prof Chalmers will be keeping a tight lid on results until RNS, but hopefully not on positive efficacy data, given how significant that would be....Gla holders...a rare opportunity for substantial imminent upside on good news...Excitement building!!!:-)

Evgen Pharma plc (AIM: EVG), a clinical stage drug development company, announces that its Annual Report and Accounts for the year ended 31 March 2021, the Notice of Annual General Meeting ("AGM") and a Form of Proxy will be posted to shareholders later today and will be available to download from the Company website shortly.



The Company will hold its AGM on Tuesday, 13 July 2021 at 2pm at its offices in Alderley Park.



Since laws and the UK Government's guidance regarding the COVID-19 pandemic current on Tuesday 13 July 2021 continue to include the enforcement of social distancing and restrictions on indoor meetings, shareholders will not be permitted to attend the AGM and a closed meeting will be held. Accordingly, the business at the AGM will be curtailed to the formal business section only, with no wider presentations on business performance or question and answer session and any relevant shareholder updates will be made via RNS announcements.



Similarly, proxies other than the Chairman will not be able to attend the Meeting in person or cast the shareholder's vote.
