Rather than calling an EGM, surely a face to face with one or two level-headed shareholders could answer questions that don't warrant an RNS. A brief rundown of a meeting could then be posted here - clarity without need for threats of an EGM or revealing any sensitive info.

Hi PCS/Aber et al - is it feasible then for one or two of our main investors to get in touch with the BoD after the convention and demand a face to face meeting with Tim or Stephen to clarify what's going on re roles & responsibilities etc. etc.?

Hi Superritch - not sure why you consider SAR a write-off. They announced the extended tox trial has been initiated (for those getting their knickers in a twist over the wording, initiated means to have begun or set something in motion, such as a process or action), and if all goes well 1801 will be P2-ready with an acceptable or maybe even an excellent safety profile before the end of the year.

I understand all the frustrations re communications etc. but we know where we stand - tox study on-going, control over 737, 2 new CNS candidates, good old 1802 still(!!) in translational studies and JR writing up a paper on the Oz trial. I feel selling up now would be like going round to a new girlfriend's place and she invites you to stay the night but you explain you've got a new flavour of crisps to try back at home so, y'know, maybe some other time. Each to their own.

Just been looking at the Bio Convention website - 19608 registrants and 61508 meetings arranged. Do people seriously think Tim'll just be standing in a corner wearing an 'Ask me about Chk1' t-shirt? Some may think I'm some sort of SAR apologist, I'm not, I'm just defending my investment.

Melanoma has a rising incidence worldwide. Current treatments are effective, although the development of resistance is common. A novel anti-cancer treatment using checkpoint kinase 1 inhibitor (CHK1i), SRA737, in combination with low-dose hydroxyurea (LDHU), has been demonstrated to effectively kill tumour cells and promote an anti-tumour immune response through the treatment-induced release of pro-inflammatory chemokines and cytokines. These chemokines/cytokines modify the tumour microenvironment from an immunosuppressive to an inflamed state to recruit anti-tumour immune cells.

Results: We demonstrate that SRA737 + LDHU upregulates pro-inflammatory chemokines in human melanoma cells in response to SRA737 + LDHU through the ATM-NF-κB signalling pathway. The increased chemokine expression corresponded to the increase in secretion of pro-inflammatory chemokines from tumour cells following SRA737 + LDHU treatment. However, inhibiting NF-κB and ATM did not affect SRA737 + LDHU-induced cell killing. Increased expression of non-NF-κB target genes with SRA737 + LDHU suggests that other transcriptional pathways are also activated and may contribute to the increasing cytokine/chemokine gene expression in response to treatment.

Conclusions: SRA737 + LDHU upregulates pro-inflammatory chemokine expression through an ATM-NF-κB-dependent mechanism.

Matty - It'd have to be a helluva backhander. I think SP has 400k shares plus whatever options he may have. If the sp was still at £4 then that's an easy £1.6m+ so how much would he have to take from a company to top that?

Hi Meg - I think us punters are in a really difficult position re Parker simply because we'll never know the truth about how hard or not he's working for us. For all we know he's trying to engineer a massive takeover or he's taking the mickey - both could be covered by a 'need to know/business sensitive' umbrella.

Since we appear to be funded until early next year, if we don't make any significant progress once the tox report is in then it could be time for a shareholder revolt but we should be careful what we wish for.

Evening PCS - I'm as frustrated as everyone else seems to be with the lack of sp-boosting news etc. but at least 1801's extended tox trial is underway.

I presume Tim is off to Boston instead of SP in order to answer questions on the science side. It wouldn't look good if SP answered most questions with, 'umm, I have to get back to you on that...' I think at this stage of the game us punters are not the target audience as we're already bought in hence the lack of PR being aimed our way. SP should however be firing off e-mails with data excerpts to every contact in his little black book but I don't expect to be informed of that as it must surely come under the heading of 'business as usual' - in much the same way that GSK or Pfizer don't hold a press conference every time they hold a teambuilding event. If a company has any interest in what we do, they'll go direct to the BoD rather than look on Twitter/X for info.

That said, if anyone connected to the BoD reads this(!), they might suggest either adding some sort of progress page to the website for updates that don't warrant an RNS or maybe get a bit more active on Twitter/X for the benefit of punters e.g.

15/5/25 first species in extended tox study now dosed with 1801, vital signs to be monitored for X days

20/5/25 patent application for xyz submitted to Spanish authorities for validation

23/5/25 increased dosage administered etc etc.

Yes, this would require some additional cost in maintaining the website but it might placate those looking for news when an RNS might not be wholly justified.

If Tim emerges from the event with his face painted like a tiger and carrying a balloon animal then we'll know for sure it's a lifestyle company, otherwise the event might just be a backup plan in case our 'advisor' loses interest (for whatever reason) in the extended tox study. IF this interested party is our preferred candidate for a partnership/sale and they're waiting to confirm 1801 really has a clean tox profile before making an offer, the BoD may still have to be doing the rounds as a way of keeping some options open.

At the start of the year the BoD said we're funded for approx 18mths however that was before April's announcement that we now have a couple of CNS candidates - depending on what development the BoD want to carry out on those, we may see a shorter cash runway so Tim might be in the US to try and sell off 737 once and for all ahead of greater cash needs should they be required before completion of the tox study.

In patent news, although the EU patent below was granted in 2023, I see it has been published in the last few months in Poland, Portugal & Spain. When a patent is granted through the European Patent Office, it isn't automatically valid across all European countries. Instead, it creates a bundle of potential national rights that need to be activated through a process called "validation". Each validated patent becomes independent after grant, allowing flexibility in maintaining rights in different countries - it's not redundant filing, but rather a necessary step to activate and maintain enforceable patent rights in specific European countries.

Evening JB - so is it also standard practice for a company to announce who is carrying out a tox study for them? And when you say, "...they could just google it like I did", are you telling me you know which lab is involved? (My guess is you don't, unless you snuck in to SAR HQ via an air vent, shot a tranquilizer dart into the security guard, stole a laptop and cracked the password (biscuits37).)

Do you really think it a good idea to announce which lab is conducting animal testing? Wouldn't it be marvelous if the lab was then attacked by activists and our trial got derailed?

A Chinese group published a paper today (link below) on "Overcoming temozolomide resistance in glioma: recent advances and mechanistic insights" in which 737 gets a fleeting but crucial mention, "For example, TMZ treatment activates CHK1, promoting structural changes in lnc01956. These changes allow lnc01956 to move from the nucleus to the cytoplasm, where it binds MGMT and induces chemoresistance. Blocking CHK1 with the inhibitor SRA737 reversed this effect, restoring TMZ sensitivity."

Hi NavM - it's only a news vacuum if we get nothing on the publishing of the 1801 paper, no plan of action for 737, no end to the 1802 translational studies and no decision on what to do with the new CNS candidates. But other than that, yeah, a total vacuum (rolls eyes).

I hope we've been steered down this path by a strong candidate for partnering/licensing rather than through a casual conversation with a CRO.

As for being first in class v's best in class - clearly there are advantages/disadvantages to both. Being first pays well but only whilst there's no competition plus you run the risk of falling into unforeseen complications.
